    Increase in environmental temperature affects exploratory behaviour, anxiety and social preference in Danio rerio

    The aim of this work is to investigate the effect of a temperature increase on the behaviour of adult zebrafish (Danio rerio) maintained for 21 days at 34 °C (treatment) and 26 °C (control). The temperatures chosen are within the vital range of zebrafish and correspond to temperatures that this species encounters in the natural environment. Previous results showed that the same treatment affects the brain proteome and the behaviour of adult zebrafish by producing alterations in the proteins involved in neurotransmitter release and synaptic function and impairing fish exploratory behaviour. In this study, we have investigated the performance of treated and control zebrafish during environmental exploration by using four behavioural tests (novel tank diving, light and dark preference, social preference and mirror biting) that are paradigms for assessing the state of anxiety, boldness, social preference and aggressive behaviour, respectively. The results showed that heat treatment reduces anxiety and increases the boldness of zebrafish, which spent more time in potentially dangerous areas of the tank such as the top and the uncovered bright area and at a distance from the social group, thus decreasing protection for the zebrafish. These data suggest that the increase in ambient temperature may compromise zebrafish survival rate in the natural environment

    Emerging Neuroblastoma 3D In Vitro Models for Pre-Clinical Assessments

    The potential of tumor three-dimensional (3D) in vitro models for the validation of existing or novel anti-cancer therapies has been largely recognized. During the last decade, diverse in vitro 3D cell systems have been proposed as a bridging link between two-dimensional (2D) cell cultures and in vivo animal models, both considered gold standards in pre-clinical settings. The latest awareness about the power of tailored therapies and cell-based therapies in eradicating tumor cells raises the need for versatile 3D cell culture systems through which we might rapidly understand the specificity of promising anti-cancer approaches. Yet, a faithful reproduction of the complex tumor microenvironment is demanding as it implies a suitable organization of several cell types and extracellular matrix components. The proposed 3D tumor models discussed here are expected to offer the required structural complexity while also assuring cost-effectiveness during pre-selection of the most promising therapies. As neuroblastoma is an extremely heterogenous extracranial solid tumor, translation from 2D cultures into innovative 3D in vitro systems is particularly challenging. In recent years, the number of 3D in vitro models mimicking native neuroblastoma tumors has been rapidly increasing. However, in vitro platforms that efficiently sustain patient-derived tumor cell growth, thus allowing comprehensive drug discovery studies on tailored therapies, are still lacking. In this review, the latest neuroblastoma 3D in vitro models are presented and their applicability for a more accurate prediction of therapy outcomes is discussed

    Measurement of the Omega_c Lifetime

    We present the measurement of the lifetime of the Omega_c we have performed using three independent data samples from two different decay modes. Using a Sigma- beam of 340 GeV/c we have obtained clean signals for the Omega_c decaying into Xi- K- pi+ pi+ and Omega- pi+ pi- pi+, avoiding topological cuts normally used in charm analysis. The short but measurable lifetime of the Omega_c is demonstrated by a clear enhancement of the signals at short but finite decay lengths. Using a continuous maximum likelihood method we determined the lifetime to be tau(Omega_c) = 55 +13-11(stat) +18-23(syst) fs. This makes the Omega_c the shortest living weakly decaying particle observed so far. The short value of the lifetime confirms the predicted pattern of the charmed baryon lifetimes and demonstrates that the strong interaction plays a vital role in the lifetimes of charmed hadrons.Comment: 15 pages, including 7 figures; gzipped, uuencoded postscrip

    Acute environmental temperature variation affects brain protein expression, anxiety and explorative behaviour in adult zebrafish

    This study investigated the effect of 4-d acute thermal treatments at 18 \ub0C, 26 \ub0C (control) and 34 \ub0C on the nervous system of adult zebrafish (Danio rerio) using a multidisciplinary approach based on behavioural tests and brain proteomic analysis. The behavioural variations induced by thermal treatment were investigated using five different tests, the novel tank diving, light and dark preference, social preference, mirror biting, and Y-Maze tests, which are standard paradigms specifically tailored for zebrafish to assess their anxiety-like behaviour, boldness, social preference, aggressiveness, and explorative behaviour, respectively. Proteomic data revealed that several proteins involved in energy metabolism, messenger RNA translation, protein synthesis, folding and degradation, cytoskeleton organisation and synaptic vesiculation are regulated differently at extreme temperatures. The results showed that anxiety-like behaviours increase in zebrafish at 18 \ub0C compared to those at 26 \ub0C or 34 \ub0C, whereas anxiety-related protein signalling pathways are downregulated. Moreover, treatments at both 18 \ub0C and 34 \ub0C affect the exploratory behaviour that appears not to be modulated by past experiences, suggesting the impairment of fish cognitive abilities. This study is the continuation of our previous work on the effect of 21-d chronic treatment at the same constant temperature level and will enable the comparison of acute and chronic treatment effects on the nervous system function in adult zebrafish

    Enhanced CP Violation with BKD0(D0)B\to K D^0 (\overline D^0) Modes and Extraction of the CKM Angle gamma

    The Gronau-London-Wyler (GLW) method extracts the CKM angle γ\gamma by measuring B±B^\pm decay rates involving D0/D0D^0/\overline D^0 mesons. Since that method necessitates the interference between two amplitudes that are significantly different in magnitude, the resulting asymmetries tend to be small. CP violation can be greatly enhanced for decays to final states that are common to both D^0 and D0\overline D^0 and that are not CP eigenstates. In particular, large asymmetries are possible for final states f such that D0fD^0\to f is doubly Cabibbo suppressed while D0f\overline D^0\to f is Cabibbo allowed. The measurement of interference effects in two such modes allows the extraction of γ\gamma without prior knowledge of Br(BKD0)Br(B^-\to K^- \overline D^0), which may be difficult to determine due to backgrounds.Comment: 12 pages, LaTeX, no figure

    Measurements of Lifetimes and a Limit on the Lifetime Difference in the Neutral D-Meson System

    Using the large hadroproduced charm sample collected in experiment E791 at Fermilab, we report the first directly measured constraint on the decay-width difference Delta Gamma for the mass eigenstates of the D0-D0bar system. We obtain our result from lifetime measurements of the decays D0 --> K-pi+ and D0 --> K-K+, under the assumption of CP invariance, which implies that the CP eigenstates and the mass eigenstates are the same. The lifetime of D0 --> K-K+ (the CP-even final state is \tau_KK = 0.410 +/- 0.011 +/- 0.006 ps, and the lifetime of D0 --> K-pi+ (an equal mixture of CP-odd and CP-even final states is tau_Kpi = 0.413 +/- 0.003 +/- 0.004 ps. The decay-width difference is Delta Gamma = 2(Gamma_KK - Gamma_Kpi) = 0.04 +/- 0.14 +/- 0.05 ps^-1.     Near Elimination of Ventricular Pacing in SafeR Mode Compared to DDD Modes: A Randomized Study of 422 Patients

    Aims:SafeR performance versus DDD/automatic mode conversion (DDD/AMC) and DDD with a 250-ms atrioventricular (AV) delay (DDD/LD) modes was assessed toward ventricular pacing (Vp) reduction. Methods:After a 1-month run-in phase, recipients of dual-chamber pacemakers without persistent AV block and persistent atrial fibrillation (AF) were randomly assigned to SafeR, DDD/AMC, or DDD/LD in a 1:1:1 design. The main endpoint was the percentage of Vp (%Vp) at 2 months and 1 year after randomization, ascertained from device memories. Secondary endpoints include %Vp at 1 year according to pacing indication and 1-year AF incidence based on automatic mode switch device stored episodes. Results:Among 422 randomized patients (73.2 ± 10.6 years, 50% men, sinus node dysfunction 47.4%, paroxysmal AV block 30.3%, bradycardia-tachycardia syndrome 21.8%), 141 were assigned to SafeR versus 146 to DDD/AMC and 135 to DDD/LD modes. Mean %Vp at 2 months was 3.4 ± 12.6% in SafeR versus 33.6 ± 34.7% and 14.0 ± 26.0% in DDD/AMC and DDD/LD modes, respectively (P < 0.0001 for both). At 1 year, mean %Vp in SafeR was 4.5 ± 15.3% versus 37.9 ± 34.4% and 16.7 ± 28.0% in DDD/AMC and DDD/LD modes, respectively (P < 0.0001 for both). The proportion of patients in whom Vp was completely eliminated was significantly higher in SafeR (69%) versus DDD/AMC (15%) and DDD/LD (45%) modes (P < 0.0001 for both), regardless of pacing indication. The absolute risk of developing permanent AF or of remaining in AF for >30% of the time was 5.4% lower in SafeR than in the DDD pacing group (ns). Conclusions:In this selected patient population, SafeR markedly suppressed unnecessary Vp compared with DDD modes. PACE 2012; 35:392–402

    The Leading Particle Effect from Heavy-Quark Recombination

    The leading particle effect in charm hadroproduction is an enhancement of the cross section for a charmed hadron D in the forward direction of the beam when the beam hadron has a valence parton in common with the D. The large D+/D- asymmetry observed by the E791 experiment is an example of this phenomenon. We show that the heavy-quark recombination mechanism provides an economical explanation for this effect. In particular, the D+/D- asymmetry can be fit reasonably well using a single parameter whose value is consistent with a recent determination from charm photoproduction.Comment: Revtex file, 4 pages, 3 figure
