Early Interferon-γ Production in Human Lymphocyte Subsets in Response to Nontyphoidal Salmonella Demonstrates Inherent Capacity in Innate Cells
Background
Nontyphoidal Salmonellae frequently cause life-threatening bacteremia in sub-Saharan Africa. Young children and HIV-infected adults are particularly susceptible. High case-fatality rates and increasing antibiotic resistance require new approaches to the management of this disease. Impaired cellular immunity caused by defects in the T helper 1 pathway lead to intracellular disease with Salmonella that can be countered by IFNγ administration. This report identifies the lymphocyte subsets that produce IFNγ early in Salmonella infection.
Methodology
Intracellular cytokine staining was used to identify IFNγ production in blood lymphocyte subsets of ten healthy adults with antibodies to Salmonella (as evidence of immunity to Salmonella), in response to stimulation with live and heat-killed preparations of the D23580 invasive African isolate of Salmonella Typhimurium. The absolute number of IFNγ-producing cells in innate, innate-like and adaptive lymphocyte subpopulations was determined.
Principal Findings
Early IFNγ production was found in the innate/innate-like lymphocyte subsets: γδ-T cells, NK cells and NK-like T cells. Significantly higher percentages of such cells produced IFNγ compared to adaptive αβ-T cells (Student's t test, P<0.001 and ≤0.02 for each innate subset compared, respectively, with CD4+- and CD8+-T cells). The absolute numbers of IFNγ-producing cells showed similar differences. The proportion of IFNγ-producing γδ-T cells, but not other lymphocytes, was significantly higher when stimulated with live compared with heat-killed bacteria (P<0.0001).
Conclusion/Significance
Our findings indicate an inherent capacity of innate/innate-like lymphocyte subsets to produce IFNγ early in the response to Salmonella infection. This may serve to control intracellular infection and reduce the threat of extracellular spread of disease with bacteremia which becomes life-threatening in the absence of protective antibody. These innate cells may also help mitigate against the effect on IFNγ production of depletion of Salmonella-specific CD4+-T lymphocytes in HIV infection
Individual behavioral rules sustain the cell allocation pattern in the combs of honey bee colonies (Apis mellifera)
In the beeswax combs of honey bees, the cells of brood, pollen, and honey
have a consistent spatial pattern that is sustained throughout the life of a
colony. This spatial pattern is believed to emerge from simple behavioral rules
that specify how the queen moves, where foragers deposit honey/pollen and how
honey/pollen is consumed from cells. Prior work has shown that a set of such
rules can explain the formation of the allocation pattern starting from an
empty comb. We show that these rules cannot maintain the pattern once the brood
start to vacate their cells, and we propose new, biologically realistic rules
that better sustain the observed allocation pattern. We analyze the three
resulting models by performing hundreds of simulation runs over many
gestational periods and a wide range of parameter values. We develop new
metrics for pattern assessment and employ them in analyzing pattern retention
over each simulation run. Applied to our simulation results, these metrics show
alteration of an accepted model for honey/pollen consumption based on local
information can stabilize the cell allocation pattern over time. We also show
that adding global information, by biasing the queen's movements towards the
center of the comb, expands the parameter regime over which pattern retention

Early affective changes and increased connectivity in preclinical Alzheimer's disease.
IntroductionAffective changes precede cognitive decline in mild Alzheimer's disease and may relate to increased connectivity in a "salience network" attuned to emotionally significant stimuli. The trajectory of affective changes in preclinical Alzheimer's disease, and its relationship to this network, is unknown.MethodsOne hundred one cognitively normal older adults received longitudinal assessments of affective symptoms, then amyloid-PET. We hypothesized amyloid-positive individuals would show enhanced emotional reactivity associated with salience network connectivity. We tested whether increased global connectivity in key regions significantly related to affective changes.ResultsIn participants later found to be amyloid positive, emotional reactivity increased with age, and interpersonal warmth declined in women. These individuals showed higher global connectivity within the right insula and superior temporal sulcus; higher superior temporal sulcus connectivity predicted increasing emotional reactivity and decreasing interpersonal warmth.ConclusionsAffective changes should be considered an early preclinical feature of Alzheimer's disease. These changes may relate to higher functional connectivity in regions critical for social-emotional processing
Risk of major depressive disorder among older persons living in HIV-endemic central and southwestern Uganda.
: Major depressive disorder (MDD) is projected to become the second most common cause of disability by 2020 calling for a better understanding its antecedents across the lifespan and in diverse socio-cultural settings. In this paper we describe the risk factors of MDD among older people (50 years +) living in HIV-endemic central and southwestern Uganda. A cross-sectional study was undertaken among 471 respondents (50 years +) participating in the Wellbeing of Older People's Study cohort of the MRC/UVRI Uganda research Unit on AIDS in Uganda. Participants were from five strata: HIV negative, HIV positive on ART, HIV positive not on ART, having an adult child on ART, and having an adult child who died of HIV. Overall MDD prevalence was 9.2% (95% CI 6.7-12.2%) with a prevalence among males of 7.4% (95% CI 4.0-12.3%) and females of 10.3% (95% CI 7.0-14.3%). Factors significantly associated with MDD included: declining socio-economic status, increasing disability scores, decreasing mean grip strength, reported back pain, and not having hypertension. Marginally associated with MDD was being HIV infected and not on ART.<br/

Vanishing Viscosity Limits and Boundary Layers for Circularly Symmetric 2D Flows
We continue the work of Lopes Filho, Mazzucato and Nussenzveig Lopes [LMN],
on the vanishing viscosity limit of circularly symmetric viscous flow in a disk
with rotating boundary, shown there to converge to the inviscid limit in
-norm as long as the prescribed angular velocity of the
boundary has bounded total variation. Here we establish convergence in stronger
and -Sobolev spaces, allow for more singular angular velocities
, and address the issue of analyzing the behavior of the boundary
layer. This includes an analysis of concentration of vorticity in the vanishing
viscosity limit. We also consider such flows on an annulus, whose two boundary
components rotate independently.
