    Long Duration Exposure Facility: A general overview

    The Long Duration Exposure Facility (LDEF) is a large, low-cost, reusable, unmanned, free-flying spacecraft which accommodates technology, science, and applications experiments for long-term exposure to the space environment. The LDEF was designed and built by the NASA Langley Research Center (LaRC) for NASA's Office of Aeronautics and Space Technology. Specifically, the LDEF was designed to transport experiments into space via the Space Shuttle, to free fly in Earth orbit for an extended period, and be retrieved on a later Space Shuttle flight allowing experiments to be returned to Earth for postflight analysis in the laboratory. The LDEF with a full complement of experiments was placed in Earth orbit in April 1984 by Challenger and retrieved from orbit in January 1990 by Columbia.     I am currently working as an assistant in developing a reading program for Charles E. Merrill Publishing Company. As might be expected, I am encountering some difficulty in getting the publisher to represent girls and women in the materials to be selected for the program. I hope you can give me some suggestions, support, pressure—whatever. Briefly the situation is this. The publisher is responsible for choosing selections to go into 4th-6th grade reader anthologies. Their art department has minority quotas (20 percent black, 10 percent Spanish-descended, etc.) but none for girls and women. Although the project editor seems to be aware of the need for representation in this area, there are other considerations that allow her to put off or minimize finding selections that are intelligently representative (i.e., non-sexist). Since, further, the two men I am working with here (two professors hired by Merrill to develop the program) are more-or-less unsympathetic to the problems of sexism in reading programs, I can exert no pressure through them. One of the professors, in fact, is pushing the inclusion of boy-relevant materials in order to get boy performance up to girl performance at the elementary level. There is little care to consider the long-run socialization process by which boys come to outperform girls

    Opportunities to improve health, production efficiency and sustainability through applied gene editing

    oai:ojs.pkp.sfu.ca:article/1741Recent advances in gene editing technologies and in the application of these technologies to livestock animals have created a wealth of opportunity for improving animal health and well being and thereby the production and sustainability of animal protein productions.  I review two technology examples in porcine and bovine systems that Genus is engaged in advancing through development.  In porcine, recent published work has demonstrated that a simple edit producing a loss of function variant for the gene product CD163 can produce full resistance to the devastating pig disease porcine reproductive and respiratory syndrome virus (PRRSv) .  In cattle, a more subtle edit, involving an edit of the -5 amino acid before the signal cleavage site of the CD 18 gene product from glutamine to glycine has been shown in cell model systems to confer resistance to the Mannheimia haemolytica leukotoxin , hypothesized to improve resilience to bovine respiratory disease (BRD).  Among other challenges, the development and successful commercialization of these types of gene editing technologies will require the creation of multiple, consistent, reproducible edits in commercial founder lines of elite genetics.  The practical challenges of deploying these technologies in beef, dairy and pork production systems are considered

    Localization of tenascin in human skin wounds

    A total of 56 surgically treated human skin wounds with a wound age between 8h and 7 months were investigated. Tenascin was visualized by immunohistochemistry and appeared first in the wound area pericellularly around fibroblastic cells approximately 2 days after wounding. A network-like interstitial positive staining pattern was first detectable in 3-day-old skin wounds. In all wounds with an age of 5 days or more, intensive reactivity for tenascin could be observed in the lesional area (dermal-epidermal junction, wound edge, areas of bleeding). In wounds with an age of more than approximately 1.5 months no positive staining occurred in the scar tissue. In conclusion, for forensic purposes, positive staining for tenascin restricted to the pericellular area of fibroblastic cells indicates a wound age of at least 2 days. Network-like structures appear after approximately 3 days or more. Since tenascin seems to be regularly detectable in skin wounds older than 5 days, the lack of a positive reaction in a sufficient number of specimens indicates a wound age of less than 5 days. The lack of a positive reaction in the granulation tissue of wounds with advanced wound age indicates a survival time of more than about 1.5 months, but a positive staining in older wounds cannot be excluded

    Vibrational Circular Dichroism Detects Symmetry Breaking due to Conformational Mobility in C2-Symmetry Chiral Molecules and Provides Further Insight into Inter-Chromophoric Interactions

    Bicyclo[3.3.1]nonane-2,6-dione (1) and bicyclo[3.3.1]nona-3,7-diene-2,6-dione (2) have been examined by vibrational circular dichroism (VCD), which, as for most C2-symmetric systems, exhibits strong VCD signals. In the case of 2, VCD signals are stronger and sharper with several bisignate doublets; for 1, signals are less intense and broader. The VCD and IR spectra are excellently predicted by DFT calculations: only one conformer is present for 2, while for 1, three main conformers, related through concerted skeleton torsional motions are present (two of them being interchanged by C2-rotation). The VCD spectrum shows specific features for the different conformers, such that correct population factors are crucial for reproducing experimental data. Also, the TD-DFT prediction of ECD (electronic circular dichroism) spectra is good. By comparing the spectroscopic signature of the two molecules (both VCD and ECD) and by careful analysis of the theoretical results, the role of the C=C double bond in compound (2) is evidenced. The double bond contributes toward enhancing the CD response both electronically and vibrationally

    Upregulation of pERK and c-JUN by γ-Tocotrienol and Not α-Tocopherol Are Essential to the Differential Effect on Apoptosis in Prostate Cancer Cells

    BACKGROUND: α-tocopherol (AT) and γ-tocotrienol (GT3) are vitamin E isoforms considered to have potential chemopreventive properties. AT has been widely studied in vitro and in clinical trials with mixed results. The latest clinical study (SELECT trial) tested AT in prostate cancer patients, determined that AT provided no benefit, and could promote cancer. Conversely, GT3 has shown antineoplastic properties in several in vitro studies, with no clinical studies published to date. GT3 causes apoptosis via upregulation of the JNK pathway; however, inhibition results in a partial block of cell death. We compared side by side the mechanistic differences in these cells in response to AT and GT3. METHODS: The effects of GT3 and AT were studied on androgen sensitive LNCaP and androgen independent PC-3 prostate cancer cells. Their cytotoxic effects were analyzed via MTT and confirmed by metabolic assays measuring ATP. Cellular pathways were studied by immunoblot. Quantitative analysis and the determination of relationships between cell signaling events were analyzed for both agents tested. Non-cancerous prostate RWPE-1 cells were also included as a control. RESULTS: The RAF/RAS/ERK pathway was significantly activated by GT3 in LNCaP and PC-3 cells but not by AT. This activation is essential for the apoptotic affect by GT3 as demonstrated the complete inhibition of apoptosis by MEK1 inhibitor U0126. Phospho-c-JUN was upregulated by GT3 but not AT. No changes were observed on AKT for either agent, and no release of cytochrome c into the cytoplasm was detected. Caspases 9 and 3 were efficiently activated by GT3 on both cell lines irrespective of androgen sensitivity, but not in cells dosed with AT. Cell viability of non-cancerous RWPE-1 cells was affected neither by GT3 nor AT. CONCLUSIONS: c-JUN is a recognized master regulator of apoptosis as shown previously in prostate cancer. However, the mechanism of action of GT3 in these cells also include a significant activation of ERK which is essential for the apoptotic effect of GT3. The activation of both, ERK and c-JUN, is required for apoptosis and may suggest a relevant step in ensuring circumvention of mechanisms of resistance related to the constitutive activation of MEK1

    A comparative study of overtone CH- stretching vibrational circular dichroism spectra of fenchone and camphor

    Near-infrared vibrational circular dichroism (VCD) spectra in the region 1500-850 nm are recorded for both enantiomers of camphor and fenchone using a home-built dispersive instrument. Two CH-stretching overtone regions and two CH-stretching/HCH-bending combination regions are investigated. The near-infrared VCD data are examined and compared with the corresponding previously published data of (3R)-(+)-methylcyclopentanone and (3R)-(+)-methylcyclohexanone. The absorption spectra of both the fundamental and overtone regions are also considered, and the fundamental CH-stretching VCD data are taken into account, where possible. An empirical assignment of the spectra is first considered, with special regard to the second CHstretching overtone region. This region is then investigated by a heuristic approach based on DFT calculations, using the hybrid functional B3LYP at the 6-31G** basis set level. Subsequently rotational strengths have been evaluated by use of a classical model based on the contribution of polarizable groups to the optical activity. The same models are also used to interpret the VCD spectra between 2000 and 3000 wavenumbers for (3R)-(+)-methylcyclopentanone-d4

    Insights into the Structures of Bilirubin and Biliverdin from Vibrational and Electronic Circular Dichroism: History and Perspectives

    : In this work we review research activities on a few of the most relevant structural aspects of bilirubin (BR) and biliverdin (BV). Special attention is paid to the exocyclic C=C bonds being in mostly Z rather than E configurations, and to the overall conformation being essentially different for BR and BV due to the presence or absence of the double C=C bond at C-10. In both cases, racemic mixtures of each compound of either M or P configuration are present in achiral solutions; however, imbalance between the two configurations may be easily achieved. In particular, results based on chiroptical spectroscopies, both electronic and vibrational circular dichroism (ECD and VCD) methods, are presented for chirally derivatized BR and BV molecules. Finally, we review deracemization experiments monitored with ECD data from our lab for BR in the presence of serum albumin and anesthetic compounds
