TAG1 regulates the endocytic trafficking and signaling of the semaphorin3A receptor complex.
Endocytic trafficking of membrane proteins is essential for neuronal structure and function. We show that Transient Axonal Glycoprotein 1 (TAG1 or CNTN2), a contactin-related adhesion molecule, plays a central role in the differential trafficking of components of the semaphorin3A (Sema3A) receptor complex into distinct endosomal compartments in murine spinal sensory neuron growth cones. The semaphorin3A receptor is composed of Neuropilin1 (NRP1), PlexinA4, and L1, with NRP1 being the ligand-binding component. TAG1 interacts with NRP1, causing a change in its association with L1 in the Sema3A response such that L1 is lost from the complex following Sema3A binding. Initially, however, L1 and NRP1 endocytose together and only become separated intracellularly, with NRP1 becoming associated with endosomes enriched in lipid rafts and colocalizing with TAG1 and PlexinA4. When TAG1 is missing, NRP1 and L1 fail to separate and NRP1 does not become raft associated; colocalization with PlexinA4 is reduced and Plexin signaling is not initiated. These observations identify a novel role for TAG1 in modulating the intracellular sorting of signaling receptor complexes
Formation of coated vesicles from coated pits in broken A431 cells
Biochemical and morphological techniques were used to demonstrate the early steps in the endocytosis of transferrin in broken A431 cells. After binding 125I-transferrin, the cells were broken by scraping and then warmed. 125I-transferrin became inaccessible to exogenous anti-transferrin antibody providing a measure of the internalization process. Parallel morphological experiments using transferrin coupled to horseradish peroxidase confirmed internalization in broken cells. The process was characterized and compared with endocytosis in intact cells and showed many similar features. The system was used to show that both the appearance of new coated pits and the scission of coated pits to form coated vesicles were dependent on the addition of cytosol and ATP whereas invagination of pits was dependent on neither
Fiber-diffraction Interferometer using Coherent Fiber Optic Taper
We present a fiber-diffraction interferometer using a coherent fiber optic
taper for optical testing in an uncontrolled environment. We use a coherent
fiber optic taper and a single-mode fiber having thermally-expanded core. Part
of the measurement wave coming from a test target is condensed through a fiber
optic taper and spatially filtered from a single-mode fiber to be reference
wave. Vibration of the cavity between the target and the interferometer probe
is common to both reference and measurement waves, thus the interference fringe
is stabilized in an optical way. Generation of the reference wave is stable
even with the target movement. Focus shift of the input measurement wave is
desensitized by a coherent fiber optic taper
Endocytosis Occurs Independently Of Annexin-Vi In Human A431 Cells
Annexin VI is one of a family of calcium-dependent phospholipid-binding proteins. Although the function of this protein is not known, various physiological roles have been proposed, including a role in the budding of clathrin-coated pits (Lin et al., 1992. Cell. 70:283-291.). In this study we have investigated a possible endocytotic role for annexin VI in intact cells, using the human squamous carcinoma cell line A431, and report that these cells do not express endogenous annexin VI, as judged by Western and Northern blotting and PCR/Southern blotting. To examine whether endocytosis might in some way be either facilitated or inhibited by the presence of annexin VI, a series of A431 clones were isolated in which annexin VI expression was achieved by stable transfection. These cells expressed annexin VI at similar levels to other human cell types. Using assays for endocytosis and recycling of the transferrin receptor, we report that each of these cellular processes occurs with identical kinetics in both transfected and wild-type A431 cells. In addition, purified annexin VI failed to support the scission of coated pits in permeabilized A431 cells. We conclude that annexin VI is not an essential component of the endocytic pathway, and that in A431 cells, annexin VI fails to exert any influence on internalization and recycling of the transferrin receptor
Force on a neutral atom near conducting microstructures
We derive the non-retarded energy shift of a neutral atom for two different
geometries. For an atom close to a cylindrical wire we find an integral
representation for the energy shift, give asymptotic expressions, and
interpolate numerically. For an atom close to a semi-infinite halfplane we
determine the exact Green's function of the Laplace equation and use it derive
the exact energy shift for an arbitrary position of the atom. These results can
be used to estimate the energy shift of an atom close to etched microstructures

Long range polarization attraction between two different likely charged macroions
It is known that in a water solution with multivalent counterions (Z-ions),
two likely charged macroions can attract each other due to correlations of
Z-ions adsorbed on their surfaces. This "correlation" attraction is
short-ranged and decays exponentially with increasing distance between
macroions at characteristic distance A/2\pi, where A is the average distance
between Z-ions on the surfaces of macroions. In this work, we show that an
additional long range "polarization" attraction exists when the bare surface
charge densities of the two macroions have the same sign, but are different in
absolute values. The key idea is that with adsorbed Z-ions, two insulating
macroions can be considered as conductors with fixed but different electric
potentials. Each potential is determined by the difference between the entropic
bulk chemical potential of a Z-ion and its correlation chemical potential at
the surface of the macroion determined by its bare surface charge density. When
the two macroions are close enough, they get polarized in such a way that their
adjacent spots form a charged capacitor, which leads to attraction. In a salt
free solution this polarization attractive force is long ranged: it decays as a
power of the distance between the surfaces of two macroions, d. The
polarization force decays slower than the van der Waals attraction and
therefore is much larger than it in a large range of distances. In the presence
of large amount of monovalent salt, when A/2\pi<< d<< r_s (r_s is the
Debye-H\"{u}ckel screening radius), this force is still much stronger than the

The effects of metals and inhibitors on thermal oxidative degradation reactions of unbranched perfluoroalkylethers
Thermal oxidative degradation studies were performed on unbranched perfluoroalkylethers at 288 C in oxygen. Metals and alloys studied included Ti, Al, and Ti (4 Al, 4 Mn). The mechanism of degradation was by chain scission. Ti and Al promoted less degradation than Ti (4 Al, 4 Mn). The two inhibitors investigated (a perfluorophenyl phosphine and a phosphatriazine) reduced degradation rates by several orders of magnitude. Both inhibitors were effective for the same duration (75 to 100 hours). The phosphatriazine appeared to provide more surface protection
Equation of the field lines of an axisymmetric multipole with a source surface
Optical spectropolarimeters can be used to produce maps of the surface magnetic fields of stars and hence to determine how stellar magnetic fields vary with stellar mass, rotation rate, and evolutionary stage. In particular, we now can map the surface magnetic fields of forming solar-like stars, which are still contracting under gravity and are surrounded by a disk of gas and dust. Their large scale magnetic fields are almost dipolar on some stars, and there is evidence for many higher order multipole field components on other stars. The availability of new data has renewed interest in incorporating multipolar magnetic fields into models of stellar magnetospheres. I describe the basic properties of axial multipoles of arbitrary degree ℓ and derive the equation of the field lines in spherical coordinates. The spherical magnetic field components that describe the global stellar field topology are obtained analytically assuming that currents can be neglected in the region exterior to the star, and interior to some fixed spherical equipotential surface. The field components follow from the solution of Laplace’s equation for the magnetostatic potential

"Engaging with birth stories in pregnancy: a hermeneutic phenomenological study of women's experiences across two generations"
BACKGROUND: The birth story has been widely understood as a crucial source of knowledge about childbirth. What has not been reported is the effect that birth stories may have on primigravid women's understandings of birth. Findings are presented from a qualitative study exploring how two generations of women came to understand birth in the milieu of other's stories. The prior assumption was that birth stories must surely have a positive or negative influence on listeners, steering them towards either medical or midwifery-led models of care.
METHODS: A Heideggerian hermeneutic phenomenological approach was used. Twenty UK participants were purposively selected and interviewed. Findings from the initial sample of 10 women who were pregnant in 2012 indicated that virtual media was a primary source of birth stories. This led to recruitment of a second sample of 10 women who gave birth in the 1970s-1980s, to determine whether they were more able to translate information into knowledge via stories told through personal contact and not through virtual technologies
RESULTS: Findings revealed the experience of 'being-in-the-world' of birth and of stories in that world. From a Heideggerian perspective, the birth story was constructed through 'idle talk' (the taken for granted assumptions of things, which come into being through language). Both oral stories and those told through technology were described as the 'modern birth story'. The first theme 'Stories are difficult like that', examines the birth story as problematic and considers how stories shape meaning. The second 'It's a generational thing', considers how women from two generations came to understand what their experience might be. The third 'Birth in the twilight of certainty,' examines women's experience of Being in a system of birth as constructed, portrayed and sustained in the stories being shared.
CONCLUSIONS: The women pregnant in 2012 framed their expectations in the language of choice, whilst the women who birthed in the 1970s-1980s framed their experience in the language of safety. For both, however, the world of birth was the same; saturated with, and only legitimised by the birth of a healthy baby. Rather than creating meaningful understanding, the 'idle talk' of birth made both cohorts fearful of leaving the relative comfort of the 'system', and of claiming an alternative birth
