    Chronic oligodendrocyte injury in central nervous system pathologies

    Myelin, the membrane surrounding neuronal axons, is critical for central nervous system (CNS) function. Injury to myelin-forming oligodendrocytes (OL) in chronic neurological diseases (e.g. multiple sclerosis) ranges from sublethal to lethal, leading to OL dysfunction and myelin pathology, and consequent deleterious impacts on axonal health that drive clinical impairments. This is regulated by intrinsic factors such as heterogeneity and age, and extrinsic cellular and molecular interactions. Here, we discuss the responses of OLs to injury, and perspectives for therapeutic targeting. We put forward that targeting mature OL health in neurological disease is a promising therapeutic strategy to support CNS function

    Applications and optimization of cryopreservation technologies to cellular therapeutics

    Delivery of cell therapies often requires the ability to hold products in readiness whilst logistical, regulatory and potency considerations are dealt with and recorded. This requires reversibly stopping biological time, a process which is often achieved by cryopreservation. However, cryopreservation itself poses many biological and biophysical challenges to living cells that need to be understood in order to apply the low temperature technologies to their best advantage. This review sets out the history of applied cryopreservation, our current understanding of the various processes involved in storage at cryogenic temperatures, and challenges for robust and reliable uses of cryopreservation within the cell therapy arena

    Variation of bone mineral density induced by exercise in postmenopausal women

    OBJETIVO: Evaluar la variación de la densidad mineral ósea (DMO) en columna lumbar (CL) y cuello femoral (CF).MÉTODO: 77 mujeres con osteopenia y menopausia, formaron cuatro grupos. Dos con ejercicio físico: uno tratamiento de estrógeno/Calcitonina (n=16), y otro ingesta de Calcio/Vitamina D (n=7). Dos sin ejercicio físico, uno tratamiento estrógeno/Calcitonina (n=27) y otro ingesta Calcio/Vitamina D (n=27). El ejercicio consistió en Fuerza Muscular (65% a 75% de 1RM) y multisaltos de baja a moderada intensidad. La DMO se midió mediante absorciometría. RESULTADOS: En ANOVA 2x2 se encontró homogeneidad (p<0,05) en la DMO tanto en CL y CF entre los tratamientos farmacológicos y nutricionales, heterogeneidad entre los grupos activos y sedentarios (CL p<0,05, CF p<0,01) y heterogeneidad en la interacción (p<0,01) de ambos tratamientos con el ejercicio físico. CONCLUSIONES: El ejercicio físico programado junto con los tratamientos habituales conduce a un mayor incremento de la DMO de en mujeres postmenopáusicasOBJECTIVE: Assess the variation of Bone Mineral Density (BMD) in Lumbar Spine (LS) and Femoral Neck (FN). METHOD: 77 postmenopausal women with osteopenia. Four groups, two groups with exercise: one Estrogen/Calcitonin treatment (n=16), another supplement Calcium/Vitamin D (n=7); two groups without exercise, one Estrogen/Calcitonin treatment (n=27), and another supplement Calcium/Vitamin D (n=27). The exercise program consisted of resistance training (65% to 75% 1RM) and multi-jumps of low-to-moderate intensity. The BMD was evaluated by absorptiometry. RESULTS. ANOVA 2x2, homogeneity (p<0,05) in the BMD in both LS and FN between the pharmacological and nutritional treatments, and heterogeneity between the active and sedentary groups (LS p <0,05, FN p<0,01) as also in the interaction (p<0,01) of the pharmacological/nutritional treatments with the physical exercise were found.     INTRODUCTION: In the last years, new technologies such as the brain-machine interfaces (BMI) have been incorporated in the rehabilitation process of subjects with stroke. These systems are able to detect motion intention, analyzing the cortical signals using different techniques such as the electroencephalography (EEG). This information could guide different interfaces such as robotic devices, electrical stimulation or virtual reality. CASE REPORT: A 40 years-old man with stroke with two months from the injury participated in this study. We used a BMI based on EEG. The subject's motion intention was analyzed calculating the event-related desynchronization. The upper limb motor function was evaluated with the Fugl-Meyer Assessment and the participant's satisfaction was evaluated using the QUEST 2.0. The intervention using a physical therapist as an interface was carried out without difficulty. CONCLUSIONS: The BMI systems detect cortical changes in a subacute stroke subject. These changes are coherent with the evolution observed using the Fugl-Meyer Assessment

    Effects of resistance training program on muscle mass and muscle strength and the relationship with cognition in Older Women

    The aim of this study was to study the effects of a resistance training programme on Maximal Dynamic Strength (MDS) and muscle morphology of the upper limbs (UL) and lower limbs (LL), as well as to analyse their association with cognition, in a population of older women. The study had a duration of 24 months and a total of 93 Chilean older women participated. The participants were divided into two groups: The Physical Activity Group (PAG, n = 45, age (X ± SD) 77.93 ± 3.54 years), and the Sedentary Group (SG, n = 48, age (X ± SD) 77.71 ± 3.41 years). The PAG carried out a muscle strength training routine twice per week. The following variables were evaluated: Muscle function through maximal dynamic strength (1RM), muscle morphology through arm and calf circumference (AC and CC, respectively), and cognition (Mini Mental State Examination: MMSE). The results show that the SG recorded significant decreases (percent changes; p < 0.05) in the analysed variables: MMSE (-3.5%), MDS in UL (-3.3%), MDS in LL (-4.1%), AC (-4.5%), CC (-4.1%), and BMI (-3.1%). However, the PAG improved significantly in all the analysed variables except in BMI: MMSE (3.9%), MDS in UL (3.6%), MDS in LL (3.5%), AC (1.8%), and CC (2.5%). Moreover, there was a significant association (p < 0.05) between the changes in the muscle strength variables and the changes in cognition level. Therefore, it can be concluded that a two-year muscle strength training programme (load intensity between 30-55% 1RM) in older women improves Maximal Dynamic Strength in UL and LL, as well as muscle mass in arms and calves. Furthermore, it can be asserted that the changes in muscle strength levels could predict the changes in the levels of cognition in older women. © 2021 by the authors.     Spanish study of anticoagulation in haemodialysis

    This study's objectives were to determine which anticoagulation methods are commonly used in patients who are undergoing haemodialysis (HD) in Spain, on what criteria do they depend, and the consequences arising from their use. MATERIAL AND METHOD: Ours was a cross-sectional study based on two types of surveys: a "HD Centre Survey" and a "Patient Survey". The first survey was answered by 87 adult HD units serving a total of 6093 patients, as well as 2 paediatric units. Among these units, 48.3% were part of the public health system and the remaining 51.7% units were part of the private health system. The patient survey analysed 758 patients who were chosen at random from among the aforementioned 78 HD units. RESULTs: A) HD Centre Survey: The majority of adult HD units (n=61, 70.2%) used both kinds of heparin, 19 of them (21.8%) only used LMWH and 7 of them (8%) only used UFH. The most frequently applied criteria for the use of LMWH were medical indications (83.3% of HD units) and ease of administration (29.5%). The most frequently used methods for adjusting the dosage were clotting of the circuit (88.2% of units), bleeding of the vascular access after disconnection (75.3%), and patient weight (57.6%). B) Patient Survey: The distribution of the types of heparin used was: UFH: 44.1%, LMWH: 51.5%, and dialysis without heparin in 4.4% of patients. LMWH was more frequently used in public medical centres (64.2% of patients) than in private medical centres (46.1%) (P<.001). LMWH was more frequently used in on-line haemodiafiltration (HF) than in high-flux HD (P<.001). Antiplatelet agents were given to 45.5% of patients, oral anticoagulants to 18.4% of patients, and both to 5% of patients. Additionally, 4.4% of patients had suffered bleeding complications during the previous week, and 1.9% of patients suffered thrombotic complications. Bleeding complications were more frequent in patients with oral anticoagulants (P=.001), although there was no association between the type of heparin and the occurrence of bleeding or thrombotic complications. CONCLUSIONS: We are able to conclude that there is a great amount of disparity in the criteria used for the medical prescription of anticoagulation in HD. It is advisable that each HD unit revise their own results as well as those from other centres, and possibly to create an Anticoagulation Guide in Haemodialysis

The developing fetus gives priority to brain development ("brain sparing"), but the risk exists of neurological and cognitive deficits at short or long term. On the other hand, diets rich in fat exert pernicious effects on brain function. Using a pig model of spontaneous IUGR, we have studied the effect on the adult of a long-term high-fat diet (HFD) on the neurotransmitter profile in several brain areas, and the morphology and the proteome of the hippocampus. Our hypothesis was that animals affected by IUGR (born with low birth weight) would present a different susceptibility to an HFD when they become adults, compared with normal birth-weight animals. Our results indicate that HFD affected the serotoninergic pathway, but it did not provoke relevant changes in the morphology of the hippocampus. Finally, the proteomic analysis revealed that, in some instances, NBW and LBW individuals respond to HFD in different ways. In particular, NBW animals presented changes in oxidative phosphorylation and the extracellular matrix, whereas LBW animals presented differences in RNA splicing, anterograde and retrograde transport and the mTOR pathway

    Wunderlich syndrome (spontaneous renal hematoma) as a cause of acute abdomen: a case report

    Spontaneous renal hemorrhage, also known as Wünderlich syndrome, is a rare condition that can be life-threatening and consists of the onset of sudden hemorrhage into the subcapsular and perirenal spaces. It can be lethal as it goes unnoticed and requires aggressive treatment. The entity lies mainly in neoplasms, the malignant ones being the most common. We present the case of a 63-year-old female patient with acute abdomen clinic, a diagnostic laparoscopy was performed and renal hematoma was evidenced as the cause of the symptoms. It was managed conservatively without any complications. We emphasize the importance of keeping in mind Wünderlich syndrome as a differential diagnosis to instigate early treatment for a better outcome.
