    AbstractTraceability of meat has become a key aspect of food-quality assurance and a priority for EU countries, to meet consumer demand for comprehensive and integrated food safety policies. In this context, the traceability of animals and animal products at the breed level might play a key role as it would enable the certification of regional products linked to particular breeds.     Near-infrared reflectance spectroscopy for predicting the phospholipid fraction and the total fatty acid composition of freeze-dried beef

    Research on fatty acids (FA) is important because their intake is related to human health. NIRS can be a useful tool to estimate the FA of beef but due to the high moisture and the high absorbance of water makes it difficult to calibrate the analyses. This work evaluated near-infrared reflectance spectroscopy as a tool to assess the total fatty acid composition and the phospholipid fraction of fatty acids of beef using freeze-dried meat. An average of 22 unrelated pure breed young bulls from 15 European breeds were reared on a common concentrate-based diet. A total of 332 longissimus thoracis steaks were analysed for fatty acid composition and a freeze-dried sample was subjected to near-infrared spectral analysis. 220 samples (67%) were used as a calibration set with the remaining 110 (33%) being used for validation of the models obtained. There was a large variation in the total FA concentration across the animals giving a good data set for the analysis and whilst the coefficient of variation was nearly 68% for the monounsaturated FA it was only 27% for the polyunsaturated fatty acids (PUFA). PLS method was used to develop the prediction models. The models for the phospholipid fraction had a low R2 p and high standard error, while models for neutral lipid had the best performance, in general. It was not possible to obtain a good prediction of many individual PUFA concentrations being present at low concentrations and less variable than other FA. The best models were developed for Total FA, saturated FA, 9c18:1 and 16:1 with R2 p greater than 0.76. This study indicates that NIRS is a feasible and useful tool for screening purposes and it has the potential to predict most of the FA of freeze-dried beef. © 2021 by the authors.     Can Medication Free, Treatment-Resistant, Depressed Patients Who Initially Respond to TMS Be Maintained Off Medications? A Prospective, 12-Month Multisite Randomized Pilot Study

    AbstractBackgroundRepetitive transcranial magnetic stimulation (TMS) is efficacious for acute treatment of resistant major depressive disorder (MDD), but there is little information on maintenance TMS after acute response.Objective/hypothesisThis pilot feasibility study investigated 12-month outcomes comparing two maintenance TMS approaches – a scheduled, single TMS session delivered monthly (SCH) vs. observation only (OBS).MethodsAntidepressant-free patients with unipolar, non-psychotic, treatment-resistant MDD participated in a randomized, open-label, multisite trial. Patients meeting protocol-defined criteria for improvement after six weeks of acute TMS were randomized to SCH or OBS regimens. TMS reintroduction was available for symptomatic worsening; all patients remained antidepressant-free during the trial.ResultsSixty-seven patients enrolled in the acute phase, and 49 (73%) met randomization criteria. Groups were matched, although more patients in the SCH group had failed ≥2 antidepressants (p = .035). There were no significant group differences on any outcome measure. SCH patients had nonsignificantly longer time to first TMS reintroduction, 91 ± 66 days, vs. OBS, 77 ± 52 days; OBS patients were nonsignificantly more likely to need reintroduction (odds ratio = 1.21, 95% CI .38–3.89). Reintroduction lasted 14.3 ± 17.8 days (SCH) and 16.9 ± 18.9 days (OBS); 14/18 (78%) SCH and 17/27 (63%) OBS responded to reintroduction. Sixteen patients (32.7%) completed all 53 weeks of the study.ConclusionsMaintaining treatment-resistant depressed patients off medications with periodic TMS appears feasible in some cases. There was no statistical advantage of SCH vs. OBS, although SCH was associated with a nonsignificantly longer time to relapse. Those who initially respond to TMS have a strong chance of re-responding if relapse occurs

    The Atacama Cosmology Telescope: The polarization-sensitive ACTPol instrument

    The Atacama Cosmology Telescope (ACT) is designed to make high angular resolution measurements of anisotropies in the Cosmic Microwave Background (CMB) at millimeter wavelengths. We describe ACTPol, an upgraded receiver for ACT, which uses feedhorn-coupled, polarization-sensitive detector arrays, a 3 degree field of view, 100 mK cryogenics with continuous cooling, and meta material anti-reflection coatings. ACTPol comprises three arrays with separate cryogenic optics: two arrays at a central frequency of 148 GHz and one array operating simultaneously at both 97 GHz and 148 GHz. The combined instrument sensitivity, angular resolution, and sky coverage are optimized for measuring angular power spectra, clusters via the thermal Sunyaev-Zel'dovich and kinetic Sunyaev-Zel'dovich signals, and CMB lensing due to large scale structure. The receiver was commissioned with its first 148 GHz array in 2013, observed with both 148 GHz arrays in 2014, and has recently completed its first full season of operations with the full suite of three arrays.     Differential gene expression in tilapia Oreochromis niloticus (L., 1758) under feeding stress

    Feeding stress or suboptimal feeding in fish can have a direct impact on their energy metabolism and feed use efficiency, and thus on productivity. That impact can be quantified objectively by comparing the expression of genes between fish fed ad libitum and fish fed suboptimally. Moreover, some of the genes expressed in animals fed suboptimally can also be used as candidate genes to identify feeding stress in general. To identify some of those genes, a group of 60 tilapia Oreochromis niloticus (L., 1758) was divided into two batches (fed ad libitum or suboptimally for 22 days) with two replicates. For this purpose, we carried out differential display analysis in brain tissue samples using 36 combinations of 3 polyT anchoring primers and 12 random primers. An average of 25 bands was produced per primer pair, with 31 bands expressed differentially which were excised from the gels, sequenced, and annotated with information available on public databases. Some sequences matched genes with different binding activities, regulation of transcription, and those playing a role in synapses. One of the most interesting findings was the ALOX5 gene, which plays a role in the production of lukotrienes, important mediators in inflammatory processes. The identification of genes corresponding to those bands will provide more information about mechanisms that play a role in stressful situations produced by suboptimal feeding, as well as other causes.El estrés alimentario o subalimentación de los peces afecta directamente al metabolismo energético, a la eficacia de utilización del alimento y, por tanto, a su productividad. Este efecto se puede cuantificar de forma objetiva mediante comparaciones de expresión génica con respecto a aquellos peces que han sido alimentados a saciedad. Además, algunos de los genes expresados por los peces subalimentados pueden usarse como genes candidatos para identificar estrés alimentario en general. Para identificar estos genes se dividió un grupo de 60 tilapias Oreochromis niloticus (L., 1758) en dos tratamientos (alimentado ad líbitum o con alimentación subóptima, durante 22 días) con dos réplicas. Se utilizó la técnica de differential display en muestras de cerebro con 36 combinaciones resultado de tres cebadores polyT de anclaje y 12 cebadores aleatorios. Se expresaron una media de 25 bandas por pareja de cebadores y un total de 31 bandas expresadas diferencialmente, se extrajeron, secuenciaron y anotaron con la ayuda de la información disponible en bases de datos públicas. Algunas secuencias coinciden con genes cuya función es de unión a diferentes proteínas y de regulación de la transcripción, y juegan un papel importante en la sinapsis. Es destacable el gen ALOX5, que interviene en la producción de leucotrienos, importantes mediadores de procesos inflamatorios. La identificación de los genes que corresponden a estas bandas aportará conocimiento sobre los mecanismos que intervienen en situaciones de estrés, tanto producidas por una alimentación subóptima como por otras causas.Instituto Español de Oceanografí
