    Bakalářská práce se zabývá výpočtem jízdní dynamiku vozu upraveného pro závody do vrchu. Tímto vozidlem je BMW E36 Compact s motorem M52B25 a 139kW. Je vypočtena kompletní jízdní dynamika tohoto vozidla a jsou navrženy úpravy za účelem vylepšení jízdních vlastností. Výpočty probíhaly v programu Microsoft Excel.The bachelor thesis deals with the calculation of driving dynamics of the car adapted for racing to uphill. This vehicle is the BMW E36 Compact with the engine M52B25 and 139kW. The complete driving dynamics of this vehicle are calculated and changes are proposed to improve driving performance.     Obsahem této diplomové práce je studie membránového čerpadla s lineárním pohonem. Jako lineární pohon byl použit lineární motor. Práce je podložena teoretickými výpočty čerpadla a jeho konstrukčními návrhy. Taktéž obsahuje optimalizaci sacího, resp. výtlačného, ventilu pro čerpadlo, vytvořené pomocí programu pro výpočtové modelovaní proudění. Práce je doplněna základním pohledem na různé druhy čerpadel na základě rozdělení na hydrodynamická a hydrostatická čerpadla. Následně je uveden přehled čerpadel užívaných k čerpání krve.The content of this master‘s thesis is a study of diaphragm pump with linear drive. Linear motor was used as the linear drive. Thesis is well – founded by theoretic calculation of pump and its design draft. Likewise, the thesis includes optimalization of suction, or pressure, valve for pump, created by using computer fluid dynamics software. Thesis is complemented by basic view of different types of pumps based on the classification by hydrodynamic and hydrostatic pumps. Subsequently, the preview of blood – flow pumps is presented.

    A Conditioning Sciatic Nerve Lesion Triggers a Pro-regenerative State in Primary Sensory Neurons Also of Dorsal Root Ganglia Non-associated With the Damaged Nerve

    The primary sensory neurons of dorsal root ganglia (DRG) are a very useful model to study the neuronal regenerative program that is a prerequisite for successful axon regeneration after peripheral nerve injury. Seven days after a unilateral sciatic nerve injury by compression or transection, we detected a bilateral increase in growth-associated protein-43 (GAP-43) and superior cervical ganglion-10 (SCG-10) mRNA and protein levels not only in DRG neurons of lumbar spinal cord segments (L4-L5) associated with injured nerve, but also in remote cervical segments (C6-C8). The increase in regeneration-associated proteins in the cervical DRG neurons was associated with the greater length of regenerated axons 1 day after ulnar nerve crush following prior sciatic nerve injury as compared to controls with only ulnar nerve crush. The increased axonal regeneration capacity of cervical DRG neurons after a prior conditioning sciatic nerve lesion was confirmed by neurite outgrowth assay of in vitro cultivated DRG neurons. Intrathecal injection of IL-6 or a JAK2 inhibitor (AG490) revealed a role for the IL-6 signaling pathway in activating the pro-regenerative state in remote DRG neurons. Our results suggest that the pro-regenerative state induced in the DRG neurons non-associated with the injured nerve reflects a systemic reaction of these neurons to unilateral sciatic nerve injury

    Role of TNF-alpha during central sensitization in preclinical studies

    Tumor necrosis factor-alpha (TNF-α) is a principal mediator in pro-inflammatory processes that involve necrosis, apoptosis and proliferation. Experimental and clinical evidence demonstrate that peripheral nerve injury results in activation and morphological changes of microglial cells in the spinal cord. These adjustments occur in order to initiate an inflammatory cascade in response to the damage. Between the agents involved in this reaction, TNF-α is recognized as a key player in this process as it not only modulates lesion formation, but also because it is suggested to induce nociceptive signals. Nowadays, even though the function of TNF-α in inflammation and pain production seems to be generally accepted, diverse sources of literature point to different pathways and outcomes. In this review, we systematically searched and reviewed original articles from the past 10 years on animal models of peripheral nervous injury describing TNF-α expression in neural tissue and pain behavior
