    Comparison of Parent and Child Ratings of Fruit and Vegetable Liking to Assess Parent Accuracy as Proxy Reporters

    This study examined the accuracy of parent-report child fruit/ vegetable (FV) liking. Child/parent dyads (n = 24) were recruited from six Head Start preschools in North Carolina. Liking for 10 FVs was assessed using a validated pictorial tool for children; a similar scale was used for parents. Negative relationships were observed between parent/child for one fruit (grapes) and one vegetable (broccoli). Positive relationships were observed among oranges, grapes, and overall fruit rankings. Parents tended to rank children’s liking of fruits higher than their children, while children ranked liking vegetables higher. Findings suggest parents may not be accurate respondents for preschool children’s FV liking

    Sequencing and Analysis of Lumpy Skin Disease Virus Whole Genomes Reveals a New Viral Subgroup in West and Central Africa

    Lumpy skin disease virus (LSDV) is a member of the capripoxvirus (CPPV) genus of the Poxviridae family. LSDV is a rapidly emerging, high-consequence pathogen of cattle, recently spreading from Africa and the Middle East into Europe and Asia. We have sequenced the whole genome of historical LSDV isolates from the Pirbright Institute virus archive, and field isolates from recent disease outbreaks in Sri Lanka, Mongolia, Nigeria and Ethiopia. These genome sequences were compared to published genomes and classified into different subgroups. Two subgroups contained vaccine or vaccine-like samples ("Neethling-like" clade 1.1 and "Kenya-like" subgroup, clade 1.2.2). One subgroup was associated with outbreaks of LSD in the Middle East/Europe (clade 1.2.1) and a previously unreported subgroup originated from cases of LSD in west and central Africa (clade 1.2.3). Isolates were also identified that contained a mix of genes from both wildtype and vaccine samples (vaccine-like recombinants, grouped in clade 2). Whole genome sequencing and analysis of LSDV strains isolated from different regions of Africa, Europe and Asia have provided new knowledge of the drivers of LSDV emergence, and will inform future disease control strategies.</p

    Activation of Akt by the Bacterial Inositol Phosphatase, SopB, is Wortmannin Insensitive

    Salmonella enterica uses effector proteins translocated by a Type III Secretion System to invade epithelial cells. One of the invasion-associated effectors, SopB, is an inositol phosphatase that mediates sustained activation of the pro-survival kinase Akt in infected cells. Canonical activation of Akt involves membrane translocation and phosphorylation and is dependent on phosphatidyl inositide 3 kinase (PI3K). Here we have investigated these two distinct processes in Salmonella infected HeLa cells. Firstly, we found that SopB-dependent membrane translocation and phosphorylation of Akt are insensitive to the PI3K inhibitor wortmannin. Similarly, depletion of the PI3K regulatory subunits p85α and p85ß by RNAi had no inhibitory effect on SopB-dependent Akt phosphorylation. Nevertheless, SopB-dependent phosphorylation does depend on the Akt kinases, PDK1 and rictor-mTOR. Membrane translocation assays revealed a dependence on SopB for Akt recruitment to Salmonella ruffles and suggest that this is mediated by phosphoinositide (3,4) P2 rather than phosphoinositide (3,4,5) P3. Altogether these data demonstrate that Salmonella activates Akt via a wortmannin insensitive mechanism that is likely a class I PI3K-independent process that incorporates some essential elements of the canonical pathway

    Title: Evaluation of a Domestic Violence Chat Helpline Background: Domestic violence (DV) is a serious and prevalent public health issue in the United States. Victims and survivors of DV can suffer from negative physical, mental, sexual, and reproductive health problems, and have negative social and economic complications. Although DV is a known problem, there are many gaps in knowledge, research, and program evaluation. This cross-sectional research project seeks to evaluate the effectiveness of The National Domestic Violence Hotline (NDVH) and its younger counterpart Love is Respect (LIR). NDVH and LIR provide crisis intervention, resources, referrals, and healthy and abusive relationship information nationwide. These services have traditionally been offered via phone lines, but as the technology improves these services are being utilized more via chat based modes. Methods: This study incorporates peer reviewed observational listening techniques used by other hotlines to examine 217 chats from a single day of the chatline. Each chat is analyzed using a conceptual framework to assess the provision of quality and responsive services by the advocate to the chatter. Variables to assess our evaluation are both qualitative and quantitative in format. Results: This study is ongoing and our presentation will provide quantitative metrics on performance towards providing information and resources to chatters and will assess how often expressed needs by the chatter are met within the chat session. Qualitative data will be presented to provide the context of the situation and to illustrate the quantitative data presented. Conclusion: This evaluation is the first of its kind and will provide helplines working with youth and adults a better understanding of the needs of their client population in addition to their advocate and program effectiveness
