    Crab body pyrolysis: Characterization and applications of crab biochar: A crabby solution

    Nova Scotia crab harvesters sell over 5 million lbs of Snow Crab (Chionoecetes opilio) annually. The commercially desired product are the legs and shoulders generating resultant waste streams from bodies of the snow crabs (approximately 1/3 of the crab). Currently this waste is landfilled which is costly and fossil fuel intensive. There is a desire to find a more environmentally sustainable practice to divert this organic animal waste from NS landfills. In a landfill, snow crab residues will decompose and generate some small amount of fixed carbon, however much of the carbon is released into the environment as CO2 during decomposition and aside from some microbial benefits none of the remaining interesting chemicals are utilized during landfill decomposition. The chemical composition of the snow crab includes a high content of protein (34.2% dw) and essential amino acids; they also have fat (17.1% dw), with a high proportion of ω3 polyunsaturated fatty acids and approximately 28.5% dw minerals (calcium, phosphorous, and magnesium) making this waste stream very intriguing as a starting biomass for the generation of biochar. In this paper we have determined the optimal pyrolysis conditions and highest yield for the char generated from the crab body waste stream. The chars have been fully characterized and we have investigated several applications ranging from neutralization material for acidic waters to concrete additives and catalysis.     Functional Movement Screen Task Scores and Joint Range-of-motion: A Construct Validity Study

    Little is known about the construct validity of the Functional Movement Screen (FMS). We aimed to assess associations between FMS task scores and measures of maximum joint range-of-motion (ROM) among university varsity student-athletes from 4 sports (volleyball, basketball, ice hockey, and soccer). Athletes performed FMS tasks and had their maximum ankle, hip and shoulder ROM measured. Multivariable linear regression was used to estimate associations between FMS task scores and ROM measurements. 101 university student-athletes were recruited (52 W/49 M; mean age 20.4±1.9 years). In general, athletes with higher FMS task scores had greater ROM compared to those with lower task scores. For example, athletes who scored 2 on the FMS squat task had 4° (95% CI, 1° to 7°) more uni-articular ankle dorsiflexion ROM compared with those who scored 1, while those who scored 3 on the FMS squat task had 10° (4° to 17°) more uni-articular ankle dorsiflexion ROM compared with those who scored 1. Large variation in ROM measurements was observed. In sum, substantial overlap in joint ROM between groups of athletes with different FMS task scores weakens the construct validity of the FMS as an indicator of specific joint ROM

In the process, they resolved many learning, logistical, and legal problems and moved this technology forward by an order of magnitude. Principal goals include: nationwide interoperability, network of repositories, linked servers, repository software programs, national and international standards, digital rights management, business and management models, evaluation and feedback, dissemination of results, and bilingual access to all Canadians, particularly learners with disabilities.     A critical reassessment of the role of mitochondria in tumorigenesis

    <p><b>Background:</b> Mitochondrial DNA (mtDNA) is being analyzed by an increasing number of laboratories in order to investigate its potential role as an active marker of tumorigenesis in various types of cancer. Here we question the conclusions drawn in most of these investigations, especially those published in high-rank cancer research journals, under the evidence that a significant number of these medical mtDNA studies are based on obviously flawed sequencing results.</p> <p><b>Methods and Findings:</b> In our analyses, we take a phylogenetic approach and employ thorough database searches, which together have proven successful for detecting erroneous sequences in the fields of human population genetics and forensics. Apart from conceptual problems concerning the interpretation of mtDNA variation in tumorigenesis, in most cases, blocks of seemingly somatic mutations clearly point to contamination or sample mix-up and, therefore, have nothing to do with tumorigenesis.</p> <p><b>Conclusion:</b> The role of mitochondria in tumorigenesis remains unclarified. Our findings of laboratory errors in many contributions would represent only the tip of the iceberg since most published studies do not provide the raw sequence data for inspection, thus hindering a posteriori evaluation of the results. There is no precedent for such a concatenation of errors and misconceptions affecting a whole subfield of medical research.</p&gt

    A longitudinal study looking at the impact of COVID-19 restrictions and transitions on psychological distress in caregivers of children with Intellectual Disabilities in the UK

    Introduction: The current study explored longitudinally whether child behaviours that challenge and caregiver coping strategies was associated with psychological distress in caregivers of children with and without intellectual disability during and after lockdown. Method: An online survey was completed by caregivers who had children with and without intellectual disability during Time Period 1 (T1; August-December 2021, n = 171) and then again during Time Period 2 (T2; January-March 2022, n = 109). Results: Child behaviours that challenge and caregiver psychological distress reduced in T2 compared to T1. Child behaviours that challenge, emotion focused coping and avoidant coping was associated distress at both time points in caregivers of children with and without intellectual disabilities. Conclusions: The study shows that both child behaviours that challenge and caregiver psychological distress reduced as lockdown ended. However, caregiver coping strategies may have contributed to psychological distress, which has implications for interventions and support for caregivers

    Pharmacologic inhibition of S-nitrosoglutathione reductase protects against experimental asthma in BALB/c mice through attenuation of both bronchoconstriction and inflammation

    BACKGROUND: S-nitrosoglutathione (GSNO) serves as a reservoir for nitric oxide (NO) and thus is a key homeostatic regulator of airway smooth muscle tone and inflammation. Decreased levels of GSNO in the lungs of asthmatics have been attributed to increased GSNO catabolism via GSNO reductase (GSNOR) leading to loss of GSNO- and NO- mediated bronchodilatory and anti-inflammatory actions. GSNOR inhibition with the novel small molecule, N6022, was explored as a therapeutic approach in an experimental model of asthma. METHODS: Female BALB/c mice were sensitized and subsequently challenged with ovalbumin (OVA). Efficacy was determined by measuring both airway hyper-responsiveness (AHR) upon methacholine (MCh) challenge using whole body plethysmography and pulmonary eosinophilia by quantifying the numbers of these cells in the bronchoalveolar lavage fluid (BALF). Several other potential biomarkers of GSNOR inhibition were measured including levels of nitrite, cyclic guanosine monophosphate (cGMP), and inflammatory cytokines, as well as DNA binding activity of nuclear factor kappa B (NFκB). The dose response, onset of action, and duration of action of a single intravenous dose of N6022 given from 30 min to 48 h prior to MCh challenge were determined and compared to effects in mice not sensitized to OVA. The direct effect of N6022 on airway smooth muscle tone also was assessed in isolated rat tracheal rings. RESULTS: N6022 attenuated AHR (ED(50) of 0.015 ± 0.002 mg/kg; Mean ± SEM) and eosinophilia. Effects were observed from 30 min to 48 h after treatment and were comparable to those achieved with three inhaled doses of ipratropium plus albuterol used as the positive control. N6022 increased BALF nitrite and plasma cGMP, while restoring BALF and plasma inflammatory markers toward baseline values. N6022 treatment also attenuated the OVA-induced increase in NFκB activation. In rat tracheal rings, N6022 decreased contractile responses to MCh. CONCLUSIONS: The significant bronchodilatory and anti-inflammatory actions of N6022 in the airways are consistent with restoration of GSNO levels through GSNOR inhibition. GSNOR inhibition may offer a therapeutic approach for the treatment of asthma and other inflammatory lung diseases. N6022 is currently being evaluated in clinical trials for the treatment of inflammatory lung disease
