    The evolution of the dialogue between Russian and Italian Сinema

    The article deals with the issues of cultural interaction between Russia and Italy in the sphere of cinematography. Cultural dialogue between Russian and Italian cinema can be viewed from different angles. The first level is intercultural communication as a tool of communicational system, the second is cultural interaction as a process of cultural practices of the two countries, and the third level is cultural diplomacy where intercultural interaction with its various forms of communication and contacts is regarded as the implementation of soft power. Based on academic research and Pavel Parshin’s theory, the following main tools of soft power can be named: mutual attraction, steady positive attitude, and permanent exchange of cultural experience. Thus cultural interaction is both the search for artistic unity and the embodiment of the dynamics of cultural dialogue analyzed through the chronotope theory of Mikhail Bakhtin within big and small time. It should be mentioned that it was M. Bakhtin who introduced the concept of dialogical nature of culture and the term of cultural dialogue. This approach makes it possible to follow the evolution of cultural dialogue between Russia and Italy over the years and define and characterize the types and stages of cultural dialogue between Russia and Italy within a broad system of cultural diplomatic relations between the countries. This paper analyses specificity of the following stages: the stage of indirect dialogue; the stage of adoption of cultural practices; the stage of suspended and re-established dialogue; the stage of activization of cultural dialogue; the stage of cultural intercourse in the era of globalization and digitalization; the modern pre-pandemic stage of cultural dialogue between Russia and Italy.     The operational value of inlet monitoring at service reservoirs

    Drinking water quality leaving water treatment works is known to deteriorate as it makes its way through distribution networks. As deterioration rates are related to the condition of the network and how it is operated, it is important to determine the location and magnitude so that causes can be determined and effective maintenance implemented. Water quality is typically monitored at outlets of service reservoirs to help track changing water quality. However, these results do not confirm whether the issue is linked to the network between treatment and the service reservoir, the service reservoir itself, or both. The work in this paper investigates the value of using inlet monitoring at service reservoirs to overcome this limitation. Results show that monitoring at both the inlet and outlet of service reservoirs provides valuable information on asset performance and highlights the location and extent of deterioration helping inform cost-efficient resource provision

    Influence of service reservoir construction age on the likelihood of bacteriological failure occurrence

    Service reservoirs (SRs) are crucial components in drinking water distribution systems (DWDSs). These assets are often associated with water quality issues, including significant waterborne disease outbreaks. To ensure safety of supply, monitoring is conducted at SR outlets for disinfectant residual and bacteriological indicators. Older SRs are often believed to be a source of bacteriological failures as old or poorly constructed SRs can be prone to structural issues, which can lead to ingress and contamination of the stored water. However, there is a lack of evidence to support the existence of a genuine relationship between SR age and risk of failure. In this study, historical bacteriological failure data at SR outlets and company-wide SR construction age data collected from three large UK water utilities was used to assess the potential relationship between the duration of time a SR has been in service and the likelihood of bacteriological failure. Findings showed that there was no relationship between the two parameters. Analysis also revealed instances where newer SRs, no older than twenty years old, experienced one or more bacteriological failures, challenging the ingrained association that older SRs are at a higher risk of ingress and bacteriological contamination

    Cloning and Characterization of Polyhydroxybutyrate Synthase from Methylobacterium extorquens AM1

    В результате поиска генов, кодирующих вероятные ПГБ-синтазы в геномах бактерий рода Methylobacterium, выявлены множественные (до пяти у одного штамма) гены ПГБ-синтаз. Филогенетическим анализом показано, что белки PhaC1, PhaC2, PhaC3 относятся к I классу ПГБ-синтаз, белки PhaC4 – к ПГБ-синтазам III класса, тогда как PhaC5, по-видимому, представляет неохарактеризованный класс ПГБ-синтаз. Впервые выделена и охарактеризована рекомбинантная ПГБ-синтаза I класса (КФ 2.3.1.B2) из Methylobacterium extorquens AM1, кодируемая геном phaC1. Молекулярная масса мономера фермента составила 78 кДа. Константа Михаэлиса (Km) для PhaC1 из штамма AM1 составила 1,3 мМ, а максимальная скорость реакции (Vmax) – 0,1 мкмоль∙мин-1∙мг-1. Получен делеционный мутант Methylobacterium extorquens по гену phaC, перспективный для дальнейшего исследования особенностей биосинтеза ПГБ метилобактериямиMultiple genes encoding putative PHB synthases (up to 5 in single strain) were found in Methylobacterium genomes. As a result of phylogenetic analysis proteins PhaC1, PhaC2, PhaC3 were identified as class I PHB synthases, PhaC4 proteins were identified as class III PHB synthases, while PhaC5 apparently belongs to uncharacterized class of PHB synthases. Firstly, the recombinant class I PBH synthase (EC 2.3.1.B2) encoded by phaC1 gene from Methylobacterium extorquens AM1 was purified and characterized. Molecular mass of enzyme monomer was 78 kDa. Michaelis constant (Km) for PhaC1 was 1,3 mM and maximal reaction rate (Vmax) was 0,1 μmol∙min–1∙mg–1. The deletion mutant of Methylobacterium extorquens in the phaC gene was obtained which is promising for further study of peculiarities of methylobacteria’s PHB biosynthesi

    Настоящая статья посвящена рассмотрению вопросов совершенствования метрологического обеспечения средств и методов неразрушающего контроля. Авторами проанализирована проблема отс3утствия единого подхода к вопросам систематизации и структуризации информации об оборудовании неразрушающего контроля, обоснована ее актуальность, а также необходимость и важность ее решения на законодательном уровне путем стандартизации. Изложены способы решения существующих задач посредством разработки универсального многопараметрического классификатора средств и методов неразрушающего контроля и создания цифровых паспортов приборов в качестве его основной составной части. Описаны результаты по выполненным на данный момент мероприятиям в рамках поставленных задач и траектория их дальнейшей реализации.This article is devoted to the consideration of the issues of improving the metrological support of instruments and methods of non-destructive testing. The authors analyzed the problem of the lack of a unified approach to the systematization and structuring of information about non-destructive testing equipment, justified its relevance, as well as the need for and importance of its solution at the legislative level through standardization. The proposals of possible ways to solve existing problems through the development of a universal multiparametric classifier of instruments and methods of non-destructive testing and the creation of digital passports of devices as its main component are presented. The results of the activities carried out now within the framework of the tasks and the trajectory of their further implementation are described
