    Exploración de alternativas para el desarrollo sostenible de sistemas de producción hortícola-ganaderos familiares en el sur de Uruguay

    La sostenibilidad de la mayoría de los predios hortícolas familiares en el sur de Uruguay está amenazada por ingresosinsuficientes y por el deterioro de los recursos naturales. En este estudio se cuantificó el efecto de la inclusión de diferentesactividades de producción ganadera en la estructura y en los resultados económico-productivos y ambientales de diferentestipos de sistemas de producción (predios) hortícolas. Se realizó un estudio exploratorio a escala predial con modelos desimulación utilizando dos predios como estudio de caso. Los resultados sugieren que la inclusión de actividades de producciónganadera en los sistemas de producción hortícolas es un factor determinante para reducir la erosión promedio del áreacultivada. Para reducir la erosión el modelo diseñó predios que reducen el área de hortalizas al tiempo que expanden larotación forrajera. Como consecuencia de esto, también se redujo el ingreso familiar (IF) entre 12 % y 31 % según el casoestudiado. Sin embargo, pudimos diseñar sistemas de producción con erosión menor a la tolerable para el tipo de suelo e IFmayor al objetivo, en los dos predios. De las actividades ganaderas evaluadas las más apropiadas para incluir en predios hortícolasson el engorde de novillos o vaquillonas en ciclos de 14 a 18 meses, debido al menor costo para reposición de animales y al menoruso de concentrados, lo que disminuye las necesidades de capital y la dependencia de insumos externos

    Molecular basis of anti-inflammatory action of platelet rich plasma on human chondrocytes : mechanisms of NF-kB inhibition via HGF

    Loss of articular cartilage through injury or disease presents major clinical challenges also because cartilage has very poor regenerative capacity, giving rise to the development of biological approaches. As autologous blood product, platelet-rich plasma (PRP) provides a promising alternative to surgery by promoting safe and natural healing. Here we tested the possibility that PRP might be effective as an anti-inflammatory agent, providing an attractive basis for regeneration of articular cartilage, and two principal observations were done. First, activated PRP in chondrocytes reduced the transactivating activity of NF-kB, critical regulator of the inflammatory process, and decreased the expression of COX2 and CXCR4 target genes. By analyzing a panel of cytokines with different biological significance, in activated PRP we observed increases in hepatocyte growth factor (HGF), interleukin-4 and tumor necrosis factor-alpha (TNF-alpha). HGF and TNF-alpha, by disrupting NF-kB-transactivating activity, were important for the anti-inflammatory function of activated PRP. The key molecular mechanisms involved in PRP-inhibitory effects on NF-kB activity were for HGF the enhanced cellular IkBalpha expression, that contributed to NF-kB-p65 subunit retention in the cytosol and nucleo-cytoplasmic shuttling, and for TNF-alpha the p50/50 DNA-binding causing inhibition of target-gene expression. Second, activated PRP in U937-monocytic cells reduced chemotaxis by inhibiting chemokine transactivation and CXCR4-receptor expression, thus possibly controlling local inflammation in cartilage. In conclusion, activated PRP is a promising biological therapeutic agent, as a scaffold in micro-invasive articular cartilage regeneration, not only for its content of proliferative/differentiative growth factors, but also for the presence of anti-inflammatory agents including HGF

    The application of ecologically intensive principles to the systemic redesign of livestock farms on native grasslands: A case of co-innovation in Rocha, Uruguay

    CONTEXT: Family-run cow-calf farms based on native grasslands exhibit low economic and social sustainability, as reflected in low family incomes and high workloads. Experimental results have shown that pasture–herd interaction management could improve native grasslands and animal productivity OBJECTIVE: This paper analyzes the extent to which the sustainability of family-run livestock farms based on native grasslands could be enhanced by a systemic redesign informed by ecological intensification practices. The research questions address the initial state of farm sustainability, key bottlenecks to improving farm sustainability, and changes in sustainability criteria achieved over three years of farm redesign. METHODS: The study was executed as part of a multi-level co-innovation project in Uruguay in which a team of scientist-practitioners and seven farm families participated in farm characterization, diagnosis, and redesign. The farm characterization took the form of indicators to describe the farms’ management and bio-physical subsystems. Redesign plans were negotiated between the research team and the farmers. Frequent monitoring and evaluation cycles enabled finetuning across the years of implementation. RESULTS AND CONCLUSION: Improvements were observed in the economic indicators gross margin (+55%), return to labor (+71%), and family income (+53%) and in the social indicator workload ( 22%), and the environmental indicators bird diversity and ecosystem integrity index were maintained or increased slightly. These changes were explained by the uptake of coherent sets of ecological intensification practices causing changes in forage height (+30%), forage allowance (+69%), pregnancy (+22), weight of weaning calf per mating cow (+32%), and presence of tussocks (+65%). Ecological intensification principles resulted in synergistic positive effects between productivity–biodiversity tradeoffs and the scope for enhanced farm resilience and stability. SIGNIFICANCE: Cow-calf family-run farms can be transformed to produce positive environmental and social effects and viable economic results. The implementation of projects in a co-innovation context may be taken as a guide to scaling up and scaling out the ecological intensification of livestock production on native grasslands, contributing to an extension system at the national level with the aim of improving cow-calf systems sustainability.Estación Experimental Agropecuaria BarilocheFil: Ruggia, Andrea.     Relationship between tumour shrinkage and reduction in Ki67 expression after primary chemotherapy in human breast cancer

    The association between tumour shrinkage and reduction in kinetic cell activity after primary chemotherapy in human breast cancer is still a matter of investigation. 157 patients with T2-4, N0-1, M0 breast cancer received primary chemotherapy consisting of either the CMF regimen + tamoxifen (the first consecutive 76 cases) or the single agent epirubicin (the subsequent 81). Ki67, p53, bcl2, c-erbB2 and steroid hormone receptors were evaluated immunohistochemically in tumour specimens obtained before chemotherapy and at surgery. Tumour shrinkage of >50% occurred in 72.4% of patients. Ki67 expression significantly decreased after chemotherapy; the reduction correlated with tumour response in both univariate (P < 0.005) and multivariate analysis (P = 0.02). p53, bcl-2, steroid hormone receptor and c-erbB2 immunostaining were scarcely affected. Baseline bcl2 (P = 0.04) and c-erbB2 (P = 0.02) were directly and inversely associated with the reduction in Ki67 immunostaining, respectively. Baseline p53 expression (P < 0.01) was directly related with Ki67 expression at residual tumour, whereas oestrogen receptor expression (P < 0.001) was inversely related. Ki67 at residual tumour was a better predictor for relapse-free survival (RFS) than baseline Ki67. Clinical response (P < 0.03), but not reduction in Ki67, was a significant independent predictor for disease recurrence. Chemotherapy was found to induce tumour shrinkage and to reduce the number of cells in the cell cycle, but its effect on tumour biology/aggressiveness was minimal. Reduction in Ki67 immunostaining correlated with clinical response but failed to be related to RFS. Ki67 expression at surgery rather than at baseline appears to be a better predictor for disease relapse. © 2001 Cancer Research Campaign  http://www.bjcancer.co

    Adjuvant sequential methotrexate → 5-fluorouracil vs 5-fluorouracil plus leucovorin in radically resected stage III and high-risk stage II colon cancer

    The aim of the study was to determine whether modulation of 5-fluorouracil (FU) by methotrexate (MTX) improves survival compared to FU+6-s-leucovorin (LV) following potentially curative resection of stage II and III colon cancer. Within 8 weeks from surgery, 1945 patients with stage III (44%) or high-risk stage II (55%) colon cancer were randomly assigned to receive either 6 monthly cycles of FU 370 mg m−2 i.v. bolus preceded by LV 100 mg m−2 i.v. bolus on days 1–5, or 6 monthly cycles of sequential MTX 200 mg m−2 i.v. days 1 and 15 and FU 600 mg m−2 i.v. on days 2 and 16 followed by LV rescue (15 mg given p.o. q 6 h × 6 doses). Levamisole 50 mg p.o. t.i.d. on days 1–3, every 14 days for 6 months, was planned to be given in both arms. After a median follow-up of 4.2 years, 568 patients have relapsed and 403 have died. Survival was similar with MTX → FU and FU+LV (77 vs 77% at 5 years; P=0.90), as were 5-year disease-free survivals (67 vs 63%; P=0.44). Efficacy results were similar for both stage III and II patients. There were two toxic deaths, two in the MTX → FU arm (0.2%) and zero in the control arm. We conclude that biochemical modulation of FU with LV or with MTX produces similar results in the adjuvant setting of colon cancer

    Primary chemotherapy with adriamycin, cisplatin, vincristine and cyclophosphamide in locally advanced thymomas: a single institution experience

    From 1990 to 1997, 16 consecutive patients with stage III and IVa invasive thymoma were treated in a single institution with primary chemotherapy consisting in adriamycin (40 mg m–2), cisplatin (50 mg m–2) administered intravenously on day 1, vincristine (0.6 mg m–2) on day 2 and cyclophosphamide (700 mg m–2) on day 4 (ADOC). The courses were repeated every 3 weeks. The aim was to evaluate the impact of this cytotoxic regimen with respect to response rate, per cent of patients radically resected, time to progression and overall survival. Two complete responses (one clinical and one pathological) and 11 partial responses were observed (overall response rate 81.2%); two patients had stable disease and one progressed. Toxicity was mild as only two patients developed grade III/IV neutropenia and one patient grade III nausea/vomiting. Nine patients were radically resected (five out of ten with stage III, and four out of six with stage IVa). Median time to progression and overall survival was 33.2 and 47.5 months respectively. Three patients were alive and disease free after more than 5 years. The ADOC scheme is highly active and manageable in the treatment of locally advanced thymoma. As a preoperative approach it should be offered to patients not amenable to surgery or to those surgically resectable but with a great deal of morbidity. © 1999 Cancer Research Campaig
