A deletion in GDF7 is associated with a heritable forebrain commissural malformation concurrent with ventriculomegaly and interhemispheric cysts in cats
Publisher Copyright: © 2020 by the authors.An inherited neurologic syndrome in a family of mixed-breed Oriental cats has been characterized as forebrain commissural malformation, concurrent with ventriculomegaly and interhemispheric cysts. However, the genetic basis for this autosomal recessive syndrome in cats is unknown. Forty-three cats were genotyped on the Illumina Infinium Feline 63K iSelect DNA Array and used for analyses. Genome-wide association studies, including a sib-transmission disequilibrium test and a case-control association analysis, and homozygosity mapping, identified a critical region on cat chromosome A3. Short-read whole genome sequencing was completed for a cat trio segregating with the syndrome. A homozygous 7 bp deletion in growth differentiation factor 7 (GDF7) (c.221_227delGCCGCGC [p.Arg74Profs]) was identified in affected cats, by comparison to the 99 Lives Cat variant dataset, validated using Sanger sequencing and genotyped by fragment analyses. This variant was not identified in 192 unaffected cats in the 99 Lives dataset. The variant segregated concordantly in an extended pedigree. In mice, GDF7 mRNA is expressed within the roof plate when commissural axons initiate ventrally-directed growth. This finding emphasized the importance of GDF7 in the neurodevelopmental process in the mammalian brain. A genetic test can be developed for use by cat breeders to eradicate this variant.Peer reviewe
Werewolf, there wolf : Variants in hairless associated with hypotrichia and roaning in the lykoi cat breed

Licensee MDPI, Basel, Switzerland.A variety of cat breeds have been developed via novelty selection on aesthetic, dermatological traits, such as coat colors and fur types. A recently developed breed, the lykoi (a.k.a. werewolf cat), was bred from cats with a sparse hair coat with roaning, implying full color and all white hairs. The lykoi phenotype is a form of hypotrichia, presenting as a significant reduction in the average numbers of follicles per hair follicle group as compared to domestic shorthair cats, a mild to severe perifollicular to mural lymphocytic infiltration in 77% of observed hair follicle groups, and the follicles are often miniaturized, dilated, and dysplastic. Whole genome sequencing was conducted on a single lykoi cat that was a cross between two independently ascertained lineages. Comparison to the 99 Lives dataset of 194 nonâlykoi cats suggested two variants in the cat homolog for Hairless (HR) (HR lysine demethylase and nuclear receptor corepressor) as candidate causal gene variants. The lykoi cat was a compound heterozygote for two loss of function variants in HR, an exon 3 c.1255_1256dupGT (chrB1:36040783), which should produce a stop codon at amino acid 420 (p.Gln420Serfs*100) and, an exon 18 c.3389insGACA (chrB1:36051555), which should produce a stop codon at amino acid position 1130 (p.Ser1130Argfs*29). Ascertainment of 14 additional cats from founder lineages from Canada, France and different areas of the USA identified four additional loss of function HR variants likely causing the highly similar phenotypic hair coat across the diverse cats. The novel variants in HR for cat hypotrichia can now be established between minor differences in the phenotypic presentations.Peer reviewe
Applications and efficiencies of the first cat 63K DNA array
Correction, Volume: 8 Article Number: 8746 DOI: 10.1038/s41598-018-26885-5 Published: JUN 4 2018The development of high throughput SNP genotyping technologies has improved the genetic dissection of simple and complex traits in many species including cats. The properties of feline 62,897 SNPs Illumina Infinium iSelect DNA array are described using a dataset of over 2,000 feline samples, the most extensive to date, representing 41 cat breeds, a random bred population, and four wild felid species. Accuracy and efficiency of the array's genotypes and its utility in performing population-based analyses were evaluated. Average marker distance across the array was 37,741 Kb, and across the dataset, only 1% (625) of the markers exhibited poor genotyping and only 0.35% (221) showed Mendelian errors. Marker polymorphism varied across cat breeds and the average minor allele frequency (MAF) of all markers across domestic cats was 0.21. Population structure analysis confirmed a Western to Eastern structural continuum of cat breeds. Genome-wide linkage disequilibrium ranged from 50-1,500 Kb for domestic cats and 750 Kb for European wildcats (Felis silvestris silvestris). Array use in trait association mapping was investigated under different modes of inheritance, selection and population sizes. The efficient array design and cat genotype dataset continues to advance the understanding of cat breeds and will support monogenic health studies across feline breeds and populations.Peer reviewe
An Investigation into the Impact of Pre-Adolescent Training on Canine Behavior
An online survey about puppy training was sent to members of the Center for Canine Behavior Studies and posted on our social media platforms. Six hundred forty-one (641) qualifying owners provided information on 1023 dogs. About half (48%) of the dogs involved in the study attended puppy training and the balance (52%) did not. The goal of the study was to find out whether puppy training at various ages (1â3 months, 4 months, 5â6 months) helped prevent behavior problems later in life (â„1 year). Attending training at 6 months of age or younger resulted in 0.71 the odds of developing aggressive behavior (95% CI: 0.53â0.97; p = 0.030), 0.64 the odds of having a compulsive behavior (95% CI: 0.45â0.92; p = 0.015), 0.60 the odds of exhibiting destructive behavior (95% CI: 0.37â0.96; p = 0.035), 0.68 the odds of excessive barking (95% CI: 0.47â0.99; p = 0.043), and 1.56 the odds of house soiling (95% CI: 1.08â2.27; p = 0.019). Ancillary findings about the entire study population were that dogs acquired at 12 weeks of age or younger were found to have 0.65 the odds of fear/anxiety (95% CI: 0.46â0.92; p = 0.016) and 0.50 the odds of exhibiting destructive behavior (95% CI: 0.31â0.79; p = 0.003). In addition, male dogs were found to have 0.68 the odds of developing aggressive behavior (95% CI: 0.53â0.88; p = 0.003), 0.66 the odds of developing compulsive behavior (95% CI: 0.49â0.88; p = 0.006), 0.37 the odds of mounting/humping (95% CI: 0.26â0.52; p < 0.001), and 1.53 the odds of rolling in repulsive materials (95% CI: 1.18â1.97; p = 0.001). Neutered dogs of either sex were found to have 3.10 the odds of fear/anxiety (95% CI: 2.05â4.72; p < 0.001), 1.97 the odds of escaping/running away (95% CI: 1.12â3.69; p = 0.025), 2.01 the odds of exhibiting coprophagia (95% CI 1.30â3.19; p = 0.002), and 1.72 the odds of rolling in repulsive materials (95% CI: 1.12â2.66; p = 0.014). The odds of problematic jumping deceased by 0.84 for each 1-year increase in age (95% CI: 0.80â0.88; p < 0.001)
Selection Factors Influencing Eventual Owner Satisfaction about Pet Dog Adoption
Personal likes, experience, and deep-rooted interests to satisfy emotional needs such as companionship, affection, empathy, and security are some of the underlying human motivations for acquiring a pet companion. In this study, we asked how long the owner took to decide whether to adopt a dog, who their dog was adopted from, their primary motivation for adoption, a ranking of characteristics considered during the adoption process, and how satisfied they were with the eventual outcome. Participants (n = 933) to this Center for Canine Behavior Studies survey completed an online questionnaire with responses representing 1537 dog/owner pairs. A majority of participants reported satisfaction with at least one of their adopted dogs. Odds of eventual satisfaction are higher for participants who spent less than a week considering an adoption or were seeking a pet to provide companionship and affection. Participants that prioritized personality as an adoption criteria were more likely to be satisfied with their adopted dogs. A mast majority (91%) of participants reported they would consider adopting another dog in the future. Selection criteria rankings that participants indicated they would employ for future adoptions tended to shift away from physical to behavior characteristics when compared to selection criteria priorities of prior adoptions
Associations between owner personality and psychological status and the prevalence of canine behavior problems
<div><p>Behavioral problems are a major source of poor welfare and premature mortality in companion dogs. Previous studies have demonstrated associations between ownersâ personality and psychological status and the prevalence and/or severity of their dogsâ behavior problems. However, the mechanisms responsible for these associations are currently unknown. Other studies have detected links between the tendency of dogs to display behavior problems and their ownersâ use of aversive or confrontational training methods. This raises the possibility that the effects of owner personality and psychological status on dog behavior are mediated via their influence on the ownerâs choice of training methods. We investigated this hypothesis in a self-selected, convenience sample of 1564 current dog owners using an online battery of questionnaires designed to measure, respectively, owner personality, depression, emotion regulation, use of aversive/confrontational training methods, and owner-reported dog behavior. Multivariate linear and logistic regression analyses identified modest, positive associations between ownersâ use of aversive/confrontational training methods and the prevalence/severity of the following dog behavior problems: owner-directed aggression, stranger-directed aggression, separation problems, chasing, persistent barking, and house-soiling (urination and defecation when left alone). The regression models also detected modest associations between ownersâ low scores on four of the âBig Fiveâ personality dimensions (Agreeableness, Emotional Stability, Extraversion & Conscientiousness) and their dogsâ tendency to display higher rates of owner-directed aggression, stranger-directed fear, and/or urination when left alone. The study found only weak evidence to support the hypothesis that these relationships between owner personality and dog behavior were mediated via the ownersâ use of punitive training methods, but it did detect a more than five-fold increase in the use of aversive/confrontational training techniques among men with moderate depression. Further research is needed to clarify the causal relationship between owner personality and psychological status and the behavioral problems of companion dogs.</p></div
