Regenerasi Pepaya Melalui Kultur in Vitro
A study was conducted in the Indonesian Center forAgricultural Biotechnology and Genetic Resources Researchand Development to optimize papaya regeneration systemsthrough in vitro culture. Four steps were done, i.e., callusinduction, callus regeneration, root formation, and acclimatization.Explant materials used were immature embryos ofpapaya cv. Burung. Immature papaya embryos were culturedon different media. The best medium for embryogeniccallus development was ½ MS + 10 mg/l 2.4-D + 60% sucrose+ 143 mg/l adenine sulphate + 50 mg/l myo inositol +400 mg/l glutamine, while that for callus embryo regenerationwas MS + 0.5 mg/l GA3 + 0.1 mg/l kinetin + Morel andWetmore Vitamin. Using this medium, the average of shootformation was three shoots per explant of embriogeniccallus, and the percentage of regenerated callus was 80%.The color of shoot derived from this treatment was green.Eighty percent of plants formed a complete root developmentusing ½ MS + 0.5 mg/l paclobutrazol media. Media hullof rice and compost was the best medium for papaya plantacclimatization. The percentage of survival on that acclimatizationstep was 65%
Transformasi Gen Antisens ACC Oksidase Pada Pepaya Dengan Teknik Penembakan Partikel
Papaya (Carica papaya L.) is a climacteric fruit that exhibit avery fast ripening rate. Ethylene controls the ripening eventin the papaya fruit. 1-aminocyclopropane-1-carbocxylic acid(ACC) oxidase gene encodes a specific enzyme for ethylenebiosynthesis. The gene had become a target for manipulationto make a gene construct of an antisense ACC oxidaseto regenerate transgenic papaya that has a characteristic ofdelayed ripening. The objective of the experiment is to engineer transgenic papaya that has a delayed ripening characteristic by transforming papaya with the antisense ACC oxidase gene through particle bombardment technique. The immature embryos of papaya variety Burungwere used for the explants. Antisense ACC oxidase and reporter (gus) genes were co-transformed to papaya calli. Four hundreds eighteen calli were bombarded by the antisense ACC oxidase gene. The transformation experiment resulted 25 putatives transgenic plants out of fifty plantsacclimatized in a greenhouse. Gus gene expression assay observed at 9 days after bombardment showed that the papaya explants bombarded twice at 9 cm shoot distance had 53.3% transformation rate of gus positive and 5.25 blue spots number in average. The results of PCR analysis showed that four out of 25 transgenic putative papaya plants (TR6, TR9, TR20, dan TR24), indicated a positive PCR of the antisense ACC oxidase gene with the amplified fragment DNA size of 800 base pair

Logistic Management and Organizational Consumer's Perception on Asparagus Marketing by Farmer CooperativeAsparagus is a commodity that is commonly sold for modern markets. Mertanadi farmer cooperative is the only cooperative in Bali that supply asparagus to many destinations including supermarkets, hotels, restaurants and other suppliers. The commodity is supplied by the cooperative members at Plaga Village, Petang district, Badung regency. The aims of this study are to find out logistic management of asparagus by Mertanadi farmer cooperative to the organizational consumers. This research was carried out to 17 organizational consumers of Mertanadi cooperative located in Denpasar city, Badung regency and Gianyar regency. Logistic management by the cooperative was analyzed using descriptive qualitative method, while the perceptions of the consumers to the commodity were analyzed using Likert scale. Result of the study shows that the Mertanadi farmer coopertaive has implemented the seven basic concepts of logistic management including planning, budgeting, procurement, warehousing and distribution, maintenance and deletion as well as controlling. Perceptions of the organizational consumers seen from product availability, product quality and price are categorized good. However there is one weakness, according to the consumers, that is the cooperative is not able to fulfil the entire demand. It is expected that the cooperative needs to manage to fulfil the whole consumer's demand
In-situ surface technique analyses and ex-situ characterization of Si1-xGex epilayers grown on Si(001)-2 ×1 by molecular beam epitaxy
Si1-xGex epilayers grown by Molecular Beam Epitaxy on Si(001) at 400 ○C have been analyzed in-situ by surface techniques such as X-ray and Ultraviolet Photoelectron Spectroscopies (XPS and UPS), Low Energy Electron Diffraction (LEED) and photoelectron diffraction (XPD). The Ge surface concentrations (x) obtained from the ratios of Ge and Si core level intensities are systematically higher than those obtained by the respective evaporation fluxes. This indicates a Ge enrichment in the first overlayers confirmed by Ge-like UPS valence band spectra. The structured crystallographic character of the epilayers is ascertained by LEED and XPD polar scans in the (100) plane since the Ge Auger LMM and the Si 2p XPD intensity patterns from the Si1-xGex epilayers are identical to those of the Si substrate. The residual stress in the epilayer is determined by ex-situ X-ray diffraction (XRD) which also allows, as Rutherford Back Scattering (RBS), Ge concentration determinations

IFI16 reduced expression is correlated with unfavorable outcome in chronic lymphocytic leukemia
Chronic lymphocytic leukemia (CLL) is the most common leukemia in adults. Its clinical course is typically indolent; however, based on a series of pathobiological, clinical, genetic, and phenotypic parameters, patient survival varies from less than 5 to more than 20 years. In this paper, we show for the first time that the expression of the interferon-inducible DNA sensor IFI16, a member of the PYHIN protein family involved in proliferation inhibition and apoptosis regulation, is associated with the clinical outcome in CLL. We studied 99 CLLs cases by immunohistochemistry and 10 CLLs cases by gene expression profiling. We found quite variable degrees of IFI16 expression among CLLs cases. Noteworthy, we observed that a reduced IFI16 expression was associated with a very poor survival, but only in cases with ZAP70/CD38 expression. Furthermore, we found that IFI16 expression was associated with a specific gene expression signature. As IFI16 can be easily detected by immunohistochemistry or flow cytometry, it may become a part of phenotypic screening in CLL patients if its prognostic role is confirmed in independent series
Temperature - Emissivity separation assessment in a sub-urban scenario
In this paper, a methodology that aims at evaluating the effectiveness of different TES strategies is presented. The methodology takes into account the specific material of interest in the monitored scenario, sensor characteristics, and errors in the atmospheric compensation step. The methodology is proposed in order to predict and analyse algorithms performances during the planning of a remote sensing mission, aimed to discover specific materials of interest in the monitored scenario. As case study, the proposed methodology is applied to a real airborne data set of a suburban scenario. In order to perform the TES problem, three state-of-the-art algorithms, and a recently proposed one, are investigated: Temperature-Emissivity Separation'98 (TES-98) algorithm, Stepwise Refining TES (SRTES) algorithm, Linear piecewise TES (LTES) algorithm, and Optimized Smoothing TES (OSTES) algorithm. At the end, the accuracy obtained with real data, and the ones predicted by means of the proposed methodology are compared and discussed
