    Molecular characterisation, evolution and expression analysis of g-type lysozymes in Ciona intestinalis

    Lysozyme is an important defense molecule of the innate immune system. Known for its bactericidal properties, lysozyme catalyzes the hydrolysis of b-(1,4)-glycosidic bonds between the N-acetyl glucosamine and N-acetyl muramic acid in the peptidoglycan layer of bacterial cell walls. In this study, the complete coding sequence of four g-type lysozymes were identified in Ciona intestinalis. Phylogenetic analysis and modelling supported the hypothesis of a close relationship with the vertebrate g-type lysozymes suggesting that the C. intestinalis g-type lysozyme genes (CiLys-g1, Cilys-g2, CiLys-g3, CiLys-g4) share a common ancestor in the chordate lineage. Protein motif searches indicated that C. intestinalis gtype lysozymes contain a GEWL domain with a GXXQ signature, typical of goose lysozymes. Quantitative Real-Time PCR analysis results showed that transcripts are expressed in various tissues from C. intestinalis. In order to determine the involvement of C. intestinalis g-type lysozymes in immunity, their expression was analyzed in the pharynx, showing that transcripts were significantly up-regulated in response to a challenge with lipopolysaccharide (LPS). These data support the view that CiLys g-type are molecules with potential for immune defense system against bacterial infection

    Interference of dibutylphthalate on human prostate cell viability

    Dibutylphthalate (DBP) is an environmental pollutant widely used as plasticizer in a variety of industrial applications worldwide. This agent can be found in personal-care products, children's toy, pharmaceuticals, food products. Exposure to DBP can occur via ingestion and inhalation as well as intravenous or skin contact. DBP belongs to the family of endocrine disrupting chemicals (EDCs) and its effects on reproductive system were demonstrated both in vivo and in vitro. In the present study we evaluated the effects of DBP on human prostate adenocarcinoma epithelial cells (LNCaP) in order to highlight xenoestrogens influence on human prostate. Moreover, we have compared DBP effects with 17β-estradiol action in order to investigate possible mimetical behaviour. We have assessed the effects of both compounds on the cell viability. After then, we have evaluated the expression of genes and proteins involved in cell cycle regulation. Furthermore, we have observed the expression and the cell localization of estrogen (ERs) and androgen (AR) receptors. In conclusion, we have demonstrated that DBP interacts with estrogen hormonal receptor pathway but differently from E2. DBP alters the normal gland physiology and it is involved in the deregulation of prostate cell cycle

    An Integral Formulation for Rectangular Wires in a 3D Magneto-Quasi-Static Field

    This paper proposes an integral formulation for calculating the magnetic and ohmic losses in rectangular wires immersed in an external 3D magneto-quasi-static field. The formulation is based on some simplifying assumptions that allow to employ a collocation method with constant elements for the discretization, reducing the computational burden. Even if the assumptions introduce an approximation, for the application at hand the numerical tests have shown that the accuracy is still acceptable. Moreover, the computational time is drastically reduced with respect to other approaches based on the finite element method

    Alkyphenol exposure alters steroidogenesis in male lizard podarcis siculus

    Background: Nonylphenol (NP) and Octylphenol (OP) are persistent and non-biodegradable environmental contaminants classified as endocrine disruptor chemicals (EDCs). These compounds are widely used in several industrial applications and present estrogen-like properties, which have extensively been studied in aquatic organisms. The present study aimed to verify the interference of these compounds alone, and in mixture, on the reproductive cycle of the male terrestrial vertebrate Podarcis siculus, focusing mainly on the steroidogenesis process. Methods: Male lizards have been treated with different injections of both NP and OP alone and in mixture, and evaluation has been carried out using a histological approach. Results: Results obtained showed that both substances are able to alter both testis histology and localization of key steroidogenic enzymes, such as 3β-hydroxysteroid dehydrogenase (3β-HSD), 17β-hydroxysteroid dehydrogenase (17β-HSD) and P450 aromatase. Moreover, OP exerts a preponderant effect, and the P450 aromatase represents the major target of both chemicals. Conclusions: In conclusion, NP and OP inhibit steroidogenesis, which in turn may reduce the reproductive capacity of the specimens

    OctylPhenol (OP) Alone and in Combination with NonylPhenol (NP) Alters the Structure and the Function of Thyroid Gland of the Lizard Podarcis siculus

    Abstract: Different environmental contaminants disturb the thyroid system at many levels. AlkylPhenols (APs), by-products of microbial degradation of AlkylPhenol Polyethoxylates (APEOs), constitute an important class of Endocrine Disrupting Chemicals (EDCs), the two most often used environmental APs being 4-nonylphenol (4-NP) and 4-tert-octylphenol (4-t-OP). The purpose of the present study was to investigate the effects on the thyroid gland of the bioindicator Podarcis siculus of OP alone and in combination with NP. We used radioimmunoassay to determine their effects on plasma 3,3′,5-triiodo-L-thyronine (T3), 3,3′,5,5′-L-thyroxine (T4), thyroid-stimulating hormone (TSH), and thyrotropin-releasing hormone (TRH) levels in adult male lizards. We also investigated the impacts of AP treatments on hepatic 5′ORD (type II) deiodinase and hepatic content of T3 and T4. After OP and OP + NP administration, TRH levels increased, whereas TSH, T3, and T4 levels decreased. Lizards treated with OP and OP + NP had a higher concentration of T3 in the liver and 5′ORD (type II) activity, whereas T4 concentrations were lower than that observed in the control group. Moreover, histological examination showed that the volume of the thyroid follicles became smaller in treated lizards suggesting that that thyroid follicular epithelial cells were not functionally active following treatment. This data collectively suggest a severe interference with hypothalamus–pituitary–thyroid axis and a systemic imbalance of thyroid hormones. Graphic Abstract: [Figure not available: see fulltext.

Different inorganic semiconductor materials have been used for the fabrication of such biocompatible microlasers, but often operate at visible wavelengths ill-suited for imaging through tissue. Here, we report on whispering gallery mode microdisk lasers made from a range of GaInP/AlGaInP multi-quantum well structures with compositions tailored to red-shifted excitation and emission. The selected semiconductor alloys show minimal toxicity and allow fabrication of lasers with stable single-mode emission in the NIR (675 – 720 nm) and sub-pJ thresholds. The microlasers operate in the first therapeutic window under direct excitation by a conventional diode laser and can also be pumped in the second therapeutic window using two-photon excitation at pulse energies compatible with standard multiphoton microscopy. Stable performance is observed under cell culturing conditions for five days without any device encapsulation. With their bio-optimized spectral characteristics, low lasing threshold and compatibility with two-photon pumping, AlGaInP-based microlasers are ideally suited for novel cell tagging and in vivo sensing applications.Publisher PDFPeer reviewe

    Flood hazard of the Somma-Vesuvius region based on historical (19-20th century) and geomorphological data

    This paper presents a preliminary susceptibility map of the flood hazard for the Somma-Vesuvius volcanic district, worked out by means of multidisciplinary historical, geological, geomorphological and rainfall data processing. It is well known that the Somma-Vesuvius volcano, due to its explosive volcanism and the dense urbanization of the surrounding area, with a population exceeding 650,000 is one of the most dangerous active volcanoes of the world. Although this area has been extensively studied from the volcanological point of view with regards to its volcanic hazard, there are currently not many detailed studies about its flood hazard factors, despite the fact that, in the last century, many intense rainfall events in this area have produced several floods that invaded the surrounding plains affecting towns and roads, and causing much damages and loss of lives. Accordingly, in this paper high-resolution DEM (5×5 m pixel) and detailed geomorphological maps of the whole area have been analyzed and processed in GIS environment, carrying out a comparative study of the present-day morphology and the morphology of the 1900’s volcanic edifice, including changes of infrastructures and buildings throughout the last century. These results, together with historical chronicles data and the rainfall accurate data for all flood events, have been processed in this paper for highlighting the drainage basins areas of Somma-Vesuvius where the flood phenomena could be more probable in the future, working out a preliminary zoning map, also suggesting in which sectors interventions useful for mitigation of flood risk should be implemented
