    Fertility preservation in ovarian tumours

    A considerable number of patients with a cancer diagnosis are of childbearing age and have not satisfied their desire for a family. Despite ovarian cancer (OC) usually occurring in older patients, 3%–14% are diagnosed at a fertile age with the overall 5-year survival rate being 91.2% in women ≀44 years of age when it is found at 1A–B stage. In this scenario, testing the safety and the efficacy of fertility sparing strategies in OC patients is very important overall in terms of quality of life. Unfortunately, the lack of randomised trials to validate conservative approaches does not guarantee the safety of fertility preservation strategies. However, evidence-based data from descriptive series suggest that in selected cases, the preservation of the uterus and at least one part of the ovary does not lead to a high risk of relapse. This conservative surgery helps to maintain organ function, giving patients of childbearing age the possibility to preserve their fertility. We hereby analysed the main evidence from the international literature on this topic in order to highlight the selected criteria for conservative management of OC patients, including healthy BRCA mutations carriers

    Fast Ultrahigh-Density Writing of Low Conductivity Patterns on Semiconducting Polymers

    The exceptional interest in improving the limitations of data storage, molecular electronics, and optoelectronics has promoted the development of an ever increasing number of techniques used to pattern polymers at micro and nanoscale. Most of them rely on Atomic Force Microscopy to thermally or electrostatically induce mass transport, thereby creating topographic features. Here we show that the mechanical interaction of the tip of the Atomic Force Microscope with the surface of a class of conjugate polymers produces a local increase of molecular disorder, inducing a localized lowering of the semiconductor conductivity, not associated to detectable modifications in the surface topography. This phenomenon allows for the swift production of low conductivity patterns on the polymer surface at an unprecedented speed exceeding 20 ÎŒms−1\mu m s^{-1}; paths have a resolution in the order of the tip size (20 nm) and are detected by a Conducting-Atomic Force Microscopy tip in the conductivity maps.Comment: 22 pages, 6 figures, published in Nature Communications as Article (8 pages

    Search for dark matter and other new phenomena in events with an energetic jet and large missing transverse momentum using the ATLAS detector

    Results of a search for new phenomena in final states with an energetic jet and large missing transverse momentum are reported. The search uses proton-proton collision data corresponding to an integrated luminosity of 36.1 fbĂą1at a centre-of-mass energy of 13 TeV collected in 2015 and 2016 with the ATLAS detector at the Large Hadron Collider. Events are required to have at least one jet with a transverse momentum above 250 GeV and no leptons (e or ĂŽÂŒ). Several signal regions are considered with increasing requirements on the missing transverse momentum above 250 GeV. Good agreement is observed between the number of events in data and Standard Model predictions. The results are translated into exclusion limits in models with pair-produced weakly interacting dark-matter candidates, large extra spatial dimensions, and supersymmetric particles in several compressed scenarios.[Figure not available: see fulltext.]

    Seasonality of Marriages and Ecological Contexts in Rural Communities of Central-Southern Italy (Abruzzo), 1500–1871

    The seasonality of 27,705 marriages celebrated in a four century span in the Province of Teramo (Abruzzo, Italy) was analyzed to identify the presence of a long-term pattern related to the prevailing subsistence activity and the main factors affecting it. The results show general agreement in all centuries with the agricultural patterns of other lowland or south-central Italian groups (Piedmont, Veneto, Liguria, Emilia Romagna, Tuscany, Latium, Campania, Apulia) and Mediterranean regions (Spain, France), although with some differences. Religious factors strongly affected the timing of marriage only during Lent. Of interest is the progressive increase through the centuries of marriages in the summer-autumn months, associated with a decrease in January and February. This suggests the passage from a summer migration system to a rural sedentary system with occasional seasonal work

    Measurements of Higgs boson production and couplings in the four-lepton channel in pp collisions at center-of-mass energies of 7 and 8 TeV with the ATLAS detector

    The final ATLAS Run 1 measurements of Higgs boson production and couplings in the decay channel H → ZZ∗ → l+l-l'+l'-, where l, l' = e or ÎŒ, are presented. These measurements were performed using pp collision data corresponding to integrated luminosities of 4.5 and 20.3 fb-1 at center-of-mass energies of 7 and 8 TeV, respectively, recorded with the ATLAS detector at the LHC. The H → ZZ∗ → 4l signal is observed with a significance of 8.1 standard deviations, with an expectation of 6.2 standard deviations, at mH = 125.36 GeV, the combined ATLAS measurement of the Higgs boson mass from the H → γγ and H → ZZ∗ → 4l channels. The production rate relative to the Standard Model expectation, the signal strength, is measured in four different production categories in the H → ZZ∗ → 4l channel. The measured signal strength, at this mass, and with all categories combined, is 1.44+0.40-0.33. The signal strength for Higgs boson production in gluon fusion or in association with tt or bb pairs is found to be 1.7+0.5-0.4, while the signal strength for vector-boson fusion combined with WH/ZH associated production is found to be 0.3+1.6-0.9

    Cyathostomine egg reappearance period following ivermectin treatment in a cohort of UK Thoroughbreds.

    Background: In spite of the emergence of populations of drug-resistant cyathostomins worldwide, little is known of parasite species responsible for 'early egg shedding' in cohorts of horses subjected to treatment with widely used anthelmintics (e.g. ivermectin [IVM]). In this study, we determined the cyathostomin egg reappearance period (ERP) after IVM treatment of a cohort of yearlings from a large Thoroughbred (TB) stud farm in the United Kingdom, and identified species of IVM-'resistant' cyathostomins using a combination of fundamental parasitology techniques coupled with advanced molecular tools. Methods: Individual faecal samples were collected from TB yearlings with cyathostomin infection prior to IVM treatment, as well as at 2, 14, 21, 28, 35, 42 and 49 days posttreatment. Faecal egg counts (FEC) were performed for each individual sample for determination of ERPs. In addition, individual larval cultures were performed and representative numbers of third stage larvae (L3s) harvested from each culture were subjected to molecular species identification via PCR-Reverse Line Blot (RLB). Results: Prior to IVM treatment, 11 cyathostomin species were detected in faecal samples from TB horses enrolled in this study, i.e. Cyathostomum (Cya.) catinatum, Cylicostephanus (Cys.) longibursatus, Cys. goldi, Cylicocyclus (Cyc.) nassatus, Cys. calicatus, Cya, pateratum, Cyc. radiatus, Paraposteriostomum mettami, Coronocyclus (Cor.) labratus, Cyc. insigne and Cyc. radiatus variant A. Of these, eggs of Cya. catinatum, Cys. longibursatus, Cyc. nassatus and Cyc. radiatus could be detected at 28 days post-treatment, while from day 42 onwards, cyathostomin species composition reflected data obtained pre-IVM treatment, with the exception of eggs of Cor. labratus and Cyc. insigne that could no longer be detected post-IVM administration. Conclusions: This study provides valuable data on the occurrence of IVM-resistance in cyathostomins in the UK. Nevertheless, further investigations are needed to shed light on the prevalence and incidence of drug-resistance in this country as well as other areas of the world where equine trade is substantial

    Genotyping and antibiotic resistance traits in Campylobacter jejuni and coli from pigs and wild boars in Italy

    The present study investigated the genomic constitution and antimicrobial resistance (AMR) of 238 Campylobacter from pigs and wild boars in Italy between 2012 and 2019. Campylobacter strains were genotyped using multilocus sequence typing (MLST) and whole genome MLST (wgMLST), screened for antimicrobial resistance genes, and tested for phenotypic susceptibility to six different antibiotics. C. coli was detected in 98.31% and 91.66% of pigs and wild boars, while C. jejuni was isolated in the remaining cases. MLST assigned 73 STs and 13 STs in pigs and wild boars, respectively, including 44 novel STs. The predominant ST in pigs was ST-854 (12.36%), followed by ST-9264 (6.18%). ST-1055 and ST-1417 were predominant in wild boars (30% and 13.33%, respectively). The minimum spanning tree using 1,121 global MLST profiles showed specific Italian clusters and a clear separation between pig and wild boar profiles. The wgMLST confirmed the MLST clustering and revealed a high genetic diversity within C. coli population in Italy. Minimum inhibitory concentrations (MIC) of six antibiotics revealed higher resistance in pigs to ciprofloxacin, nalidixic acid, streptomycin and tetracycline, compared to wild boar. In contrast, most strains were susceptible to gentamicin. Worrying levels of multidrug resistance (MDR) were observed mostly in pig isolates. Molecular screening of AMR mechanisms revealed the predominance of gyrA T86I substitution among fluoroquinolone- and quinolone-resistant isolates, and the 23S rRNA A2075G mutation among macrolide-resistant isolates. Other resistance determinants were observed: (i) tet(O) gene was present among tetracycline-resistant isolates; (ii) rpsL and aph(3’)-III genes conferring resistance to aminoglycosides, were identified only in streptomycin or gentamicin-resistant pig isolates; (iii) cmeA, cmeB, cmeC, cmeR genes responsible of pump efflux mechanisms, were observed in almost all the strains; (iv) OXA-61, encoding ÎČ-lactamase, was found in the half of the strains. Genotypic and phenotypic AMR profiling was fairly correlated for quinolones/fluoroquinolones. Campylobacter infection is common also in wild boar populations in Italy, suggesting that wild boars could be a reservoir of resistant and multi-resistant Campylobacter species, which may be of public health concern. The present study adds to our knowledge on the epidemiological and ecological traits of this pathogen in domesticated and wild swine

    ROS in cancer therapy: the bright side of the moon.

    Reactive oxygen species (ROS) constitute a group of highly reactive molecules that have evolved as regulators of important signaling pathways. It is now well accepted that moderate levels of ROS are required for several cellular functions, including gene expression. The production of ROS is elevated in tumor cells as a consequence of increased metabolic rate, gene mutation and relative hypoxia, and excess ROS are quenched by increased antioxidant enzymatic and nonenzymatic pathways in the same cells. Moderate increases of ROS contribute to several pathologic conditions, among which are tumor promotion and progression, as they are involved in different signaling pathways and induce DNA mutation. However, ROS are also able to trigger programmed cell death (PCD). Our review will emphasize the molecular mechanisms useful for the development of therapeutic strategies that are based on modulating ROS levels to treat cancer. Specifically, we will report on the growing data that highlight the role of ROS generated by different metabolic pathways as Trojan horses to eliminate cancer cells
