The objectives of this report are to describe (1) the internship activities done by the writer as an intern at Sahid Jaya Solo Hotel for a month from February 20th to March 19th 2017; (2) the steps that the writer did to do the activities, and (3) the problem that the writer met during doing the activities. There are two kinds of activities done by the writer, namely: main activities and secondary activities. The main activities the writer did were translating wedding packages brochures and translating menu of RatuRatih Restaurant in Sahid Jaya Solo Hotel. In doing the first activity the writer did: some preparations before translating, analyzing the source text, transferring ideas or information, and restructuring. The secondary activities the writer did were making promotion content on social media using English, collecting news of Sahid Jaya Solo Hotel, clipping the news of Sahid Jaya Solo Hotel and clipping news of competitor resume. In translating wedding packages brochures and menu of RatuRatih Restaurant, the writer was always careful in selecting word so that it can be understood easily by guests. The strategies the writer did to solve the problems were using dictionary, searching on Google to know the meaning, and asking the Public Relations Manager for discussion

    Impact of Online Education and Sentiment Analysis from Twitter Data using Topic Modeling Algorithms

    During a pandemic, all industries suffer greatly, and every sector of the world suffers in some way, including the education sector. Internet expressions reflect users' feelings about a product or service. The polarity of information in source data toward a subject under investigation is determined by sentiment analysis processes. The goal of this study is to examine social media expressions about online teaching and learning, as online education will become a part of everyday life in the future. We collected data from Twitter using keywords related to online education and Google form from engineering undergraduate students for prototype implementation. This analysis will assist teachers, parents, and the student community in understanding the benefits and drawbacks of the education industry, allowing for further improvement in educational outcomes. We used aspect-based sentiment analysis and topic modeling to determine sentiment polarity and important topics for education sector stakeholders. To begin, we used TextBlob Python package to determine sentiment polarity, and Bag of Words, LDA and LSA model for discovering topics. After modeling topics from the collected data, topic Coherence is used to assess the degree of semantic similarity between high-scoring words in the topic. The word cloud and LDAvis are used to visualize data. The experimental results are promising and it will assist education stakeholders in addressing the concerns that have been identified as social media expressions to work on

    Tailormade Drug Delivery System: A Novel Trio Concept of 3DP+ Hydrogel+ SLA

    Hydrogels possess three-dimensional polymeric network structure and equipped for retaining extensive measure of water or organic fluids. This quality makes them as exceptional candidate for the simulation of extra cellular matrixes. For this the three dimensional printing (3DP) has evolved as the technique for the formation of the digital models. The 3DP is capable for processing the prescriptions and the therapeutic gadgets. One of the technique known as stereolithographic (SLA) printing has shown promising results in formulating the hydrogel based system for fabrication. The SLA acts by cross connecting the saps to shape the polymer matrices. Due to water captured in the gels it is conceivable to create the pre- wetted, medicate hydrogels and gadgets. The 3DP helps in formation of tailor made drug delivery system as per needs of patients.     For the last time: temporal sensitivity and perceived timing of the final stimulus in an isochronous sequence

    An isochronous sequence is a series of repeating events with the same inter-onset-interval. A common finding is that as the length of a sequence increases, so does temporal sensitivity to irregularities - that is, the detection of deviations from isochrony is better with a longer sequence. Several theoretical accounts exist in the literature as to how the brain processes sequences for the detection of irregularities, yet there remains to be a systematic comparison of the predictions that such accounts make. To compare the predictions of these accounts, we asked participants to report whether the last stimulus of a regularly-timed sequence appeared ‘earlier’ or ‘later’ than expected. Such task allowed us to separately analyse bias and performance. Sequences lengths (3, 4, 5 or 6 beeps) were either randomly interleaved or presented in separate blocks. We replicate previous findings showing that temporal sensitivity increases with longer sequence in the interleaved condition but not in the blocked condition (where performance is higher overall). Results also indicate that there is a consistent bias in reporting whether the last stimulus is isochronous (irrespectively of how many stimuli the sequence is composed of). Such result is consistent with a perceptual acceleration of stimuli embedded in isochronous sequences. From the comparison of the models’ predictions we determine that the improvement in sensitivity is best captured by an averaging of successive estimates, but with an element that limits performance improvement below statistical optimality. None of the models considered, however, provides an exhaustive explanation for the pattern of results found

    Diverse Clinical Isolates of Mycobacterium tuberculosis Develop Macrophage-Induced Rifampin Tolerance.

    The Mycobacterium tuberculosis lineage 4 strains CDC1551 and H37Rv develop tolerance to multiple antibiotics upon macrophage residence. To determine whether macrophage-induced tolerance is a general feature of clinical M. tuberculosis isolates, we assessed macrophage-induced drug tolerance in strains from lineages 1-3, representing the other predominant M. tuberculosis strains responsible for tuberculosis globally. All 3 lineages developed isoniazid tolerance. While lineage 1, 3, and 4 strains developed rifampin tolerance, lineage 2 Beijing strains did not. Their failure to develop tolerance may be explained by their harboring of a loss-of-function mutation in the Rv1258c efflux pump that is linked to macrophage-induced rifampicin tolerance

    Abstract Background The success of hematopoietic stem cell (HSC) transplantation is dependent on the quality of the donor HSCs. Some sources of HSCs display reduced engraftment efficiency either because of inadequate number (e.g., fetal liver and cord blood), or age-related dysfunction (e.g. in older individuals). Therefore, use of pharmacological compounds to improve functionality of HSCs is a forefront research area in hematology. Methods Lineage negative (Lin−) cells isolated from murine bone marrow or sort-purified Lin−Sca-1+c-Kit+CD34− (LSK-CD34−) were treated with a nitric oxide donor, sodium nitroprusside (SNP). The cells were subjected to various phenotypic and functional assays. Results We found that SNP treatment of Lin− cells leads to an increase in the numbers of LSK-CD34+ cells in them. Using sort-purified LSK CD34− HSCs, we show that this is related to acquisition of CD34 expression by LSK-CD34− cells, rather than proliferation of LSK-CD34+ cells. Most importantly, this upregulated expression of CD34 had age-dependent contrasting effects on HSC functionality. Increased CD34 expression significantly improved the engraftment of juvenile HSCs (6–8 weeks); in sharp contrast, it reduced the engraftment of adult HSCs (10–12 weeks). The molecular mechanism behind this phenomenon involved nitric oxide (NO)-mediated differential induction of various transcription factors involved in commitment with regard to self-renewal in adult and juvenile HSCs, respectively. Preliminary experiments performed on cord blood-derived and mobilized peripheral blood-derived cells revealed that NO exerts age-dependent contrasting effects on human HSCs as well. Conclusions This study demonstrates novel age-dependent contrasting effects of NO on HSC functionality and suggests that HSC age may be an important parameter in screening of various compounds for their use in manipulation of HSCs
