    Les Ă©leveurs dans la gestion des menaces sanitaires globales : les Ă©leveurs vietnamiens et la grippe aviaire

    This paper documents the logics underpinning Vietnamese farmers' management practices of an emerging disease, the avian flu (H5N1). In the area of our survey, one the front line of the fight against H5N1 according to international organizations (WHO, OIE, FAO), farmers and their poultry are supposed to be highly impacted by the epizootic caused by the virus. Moreover, farmers are called upon to collaborate to the global fight against the virus before it mutates in a pandemic virus. Our study shows that direct (poultry mortality) and indirect impacts (consequence of the measures imposed by the government to contain the virus, fluctuation of consumers' demand,...) show to be relatively limited when compared to the permanent state of instability which characterizes the context of poultry production in the surveyed village. This instability is mainly related to numerous and regular poultry infectious diseases and market instability. If international community considers H5N1 as a zoonotic risk and a pandemic threat which asks for emergency tools, H5N1 is framed by the farmers of our study as an epizootic problem manageable through routinized measures. These measures aim at minimizing the economic impact of the disease rather than preventing poultry from the disease. Consequently, local management of the disease cannot fit with the precautionary approach promoted by the international community. ...French Abstract : Cet article analyse la logique de gestion d'une émergence sanitaire, la grippe aviaire (H5N1) dans l'espace confiné d'une communauté d'éleveurs vietnamiens, et la confronte à celle de l'espace public des organisations internationales (OMS, OIE, FAO,...). La zone d'étude appartient à ce que les organisations internationales considèrent comme la ligne de front de la lutte internationale contre la grippe aviaire. Les éleveurs, à travers leurs volailles, y sont considérés comme d'importantes victimes de l'épizootie provoquée par le virus. Ils sont en outre très largement sollicités pour participer à la lutte contre le virus, afin d'éviter que celui-ci ne mute en un virus capable de provoquer une pandémie mondiale de grippe. Cependant, notre travail montre que l'impact du virus dans le village étudié se révèle relativement limité dans ses effets directs (mortalités) et indirects (mesures d'urgence, conséquences économiques et réglementaires), compte tenu du contexte permanant de pression infectieuse (maladie de Newcastle et autres), et d'instabilité des marchés que connaissent les éleveurs. La comparaison avec la logique de la communauté internationale montre que la grippe aviaire a été appréhendée par les éleveurs essentiellement dans sa dimension épizootique (au détriment des dimensions zoonotique et pandémique) et gérée par des stratégies routinières de minimisation des effets plutôt que de contrôle des causes. En ce sens, le virus s'il a posé problème aux éleveurs du village étudié n'a cependant pas été appréhendé comme un risque et encore moins vécu comme une crise sanitaire.     Magnetoresistance and collective Coulomb blockade in super-lattices of ferromagnetic CoFe nanoparticles

    We report on transport properties of millimetric super-lattices of CoFe nanoparticles surrounded by organic ligands. R(T)s follow R(T) = R_0.exp(T/T_0)^0.5 with T_0 ranging from 13 to 256 K. At low temperature I(V)s follow I=K[(V-V_T)/V_T]^ksi with ksi ranging 3.5 to 5.2. I(V) superpose on a universal curve when shifted by a voltage proportional to the temperature. Between 1.8 and 10 K a high-field magnetoresistance with large amplitude and a strong voltage-dependence is observed. Its amplitude only depends on the magnetic field/temperature ratio. Its origin is attributed to the presence of paramagnetic states present at the surface or between the nanoparticles. Below 1.8 K, this high-field magnetoresistance abruptly disappears and inverse tunnelling magnetoresistance is observed, the amplitude of which does not exceed 1%. At this low temperature, some samples display in their I(V) characteristics abrupt and hysteretic transitions between the Coulomb blockade regime and the conductive regime. The increase of the current during these transitions can be as high as a factor 30. The electrical noise increases when the sample is near the transition. The application of a magnetic field decreases the voltage at which these transitions occur so magnetic-field induced transitions are also observed. Depending on the applied voltage, the temperature and the amplitude of the magnetic field, the magnetic-field induced transitions are either reversible or irreversible. These abrupt and hysteretic transitions are also observed in resistance-temperature measurements. They could be the soliton avalanches predicted by Sverdlov et al. [Phys. Rev. B 64, 041302 (R), 2001] or could also be interpreted as a true phase transition between a Coulomb glass phase to a liquid phase of electrons

    Economic factors influencing zoonotic disease dynamics: demand for poultry meat and seasonal transmission of avian influenza in Vietnam

    While climate is often presented as a key factor influencing the seasonality of diseases, the importance of anthropogenic factors is less commonly evaluated. Using a combination of methods-wavelet analysis, economic analysis, statistical and disease transmission modelling-we aimed to explore the influence of climatic and economic factors on the seasonality of H5N1 Highly Pathogenic Avian Influenza in the domestic poultry population of Vietnam. We found that while climatic variables are associated with seasonal variation in the incidence of avian influenza outbreaks in the North of the country, this is not the case in the Centre and the South. In contrast, temporal patterns of H5N1 incidence are similar across these 3 regions: periods of high H5N1 incidence coincide with Lunar New Year festival, occurring in January-February, in the 3 climatic regions for 5 out of the 8 study years. Yet, daily poultry meat consumption drastically increases during Lunar New Year festival throughout the country. To meet this rise in demand, poultry production and trade are expected to peak around the festival period, promoting viral spread, which we demonstrated using a stochastic disease transmission model.     Whole proteome analyses on Ruminiclostridium cellulolyticum show a modulation of the cellulolysis machinery in response to cellulosic materials with subtle differences in chemical and structural properties

    Lignocellulosic materials from municipal solid waste emerge as attractive resources for anaerobic digestion biorefinery. To increase the knowledge required for establishing efficient bioprocesses, dynamics of batch fermentation by the cellulolytic bacterium Ruminiclostridium cellulolyticum were compared using three cellulosic materials, paper handkerchief, cotton discs and Whatman filter paper. Fermentation of paper handkerchief occurred the fastest and resulted in a specific metabolic profile: it resulted in the lowest acetate-to-lactate and acetate-to-ethanol ratios. By shotgun proteomic analyses of paper handkerchief and Whatman paper incubations, 151 proteins with significantly different levels were detected, including 20 of the 65 cellulosomal components, 8 non-cellulosomal CAZymes and 44 distinct extracytoplasmic proteins. Consistent with the specific metabolic profile observed, many enzymes from the central carbon catabolic pathways had higher levels in paper handkerchief incubations. Among the quantified CAZymes and cellulosomal components, 10 endoglucanases mainly from the GH9 families and 7 other cellulosomal subunits had lower levels in paper handkerchief incubations. An in-depth characterization of the materials used showed that the lower levels of endoglucanases in paper handkerchief incubations could hypothetically result from its lower crystallinity index (50%) and degree of polymerization (970). By contrast, the higher hemicellulose rate in paper handkerchief (13.87%) did not result in the enhanced expression of enzyme with xylanase as primary activity, including enzymes from the xyl-doc cluster. It suggests the absence, in this material, of molecular structures that specifically lead to xylanase induction. The integrated approach developed in this work shows that subtle differences among cellulosic materials regarding chemical and structural characteristics have significant effects on expressed bacterial functions, in particular the cellulolysis machinery, resulting in different metabolic patterns and degradation dynamics.This work was supported by a grant [R2DS 2010-08] from Conseil Regional d'Ile-de-France through DIM R2DS programs (http://www.r2ds-ile-de-france.com/).     Secretion of Clostridium difficile Toxins A and B Requires the Holin-like Protein TcdE

    The pathogenesis of Clostridium difficile, the major cause of antibiotic-associated diarrhea, is mainly associated with the production and activities of two major toxins. In many bacteria, toxins are released into the extracellular environment via the general secretion pathways. C. difficile toxins A and B have no export signature and their secretion is not explainable by cell lysis, suggesting that they might be secreted by an unusual mechanism. The TcdE protein encoded within the C. difficile pathogenicity locus (PaLoc) has predicted structural features similar to those of bacteriophage holin proteins. During many types of phage infection, host lysis is driven by an endolysin that crosses the cytoplasmic membrane through a pore formed by holin oligomerization. We demonstrated that TcdE has a holin-like activity by functionally complementing a λ phage deprived of its holin. Similar to λ holin, TcdE expressed in Escherichia coli and C. difficile formed oligomers in the cytoplamic membrane. A C. difficile tcdE mutant strain grew at the same rate as the wild-type strain, but accumulated a dramatically reduced amount of toxin proteins in the medium. However, the complemented tcdE mutant released the toxins efficiently. There was no difference in the abundance of tcdA and tcdB transcripts or of several cytoplasmic proteins in the mutant and the wild-type strains. In addition, TcdE did not overtly affect membrane integrity of C. difficile in the presence of TcdA/TcdB. Thus, TcdE acts as a holin-like protein to facilitate the release of C. difficile toxins to the extracellular environment, but, unlike the phage holins, does not cause the non-specific release of cytosolic contents. TcdE appears to be the first example of a bacterial protein that releases toxins into the environment by a phage-like system

    Horseplay, care and hands on hard work: gendered strategies of a project manager on a construction site

    The discourse of managerial expertise favours rational analysis and masculine ideals but contemporary management literature also recognises the value of well-being and employee voice in the workplace. Drawing upon narrative analysis of interview data, we share unique insights into the lived experiences of Laura, one female project manager who recently managed a construction site in the Midlands in the UK. In contrast to previous research which indicates that female managers tend to conform to quite a traditional set of gender behaviours, Laura embraces a range of workplace appropriate gendered strategies, such as hard work and horseplay, together with sensitivity and caring. She draws from this mix of gendered strategies in negotiating between two different discourses of construction; one professional and one tough and practical. Her behaviour both reproduces the masculine ideals (through horseplay and heroic management) and opens up possibilities for modernising construction management (by caring). It is this combination of strategies that is at the heart of tacit expertise for Laura. Theoretically, the discussion adds to the development of a more nuanced understanding of management expertise as situated and person specific knowledge that draws on both the explicit and tacit. Specifically, the centrality of gendered strategies beyond the masculine ideals to success on site is highlighted
