    Protective effects of flavonoids from corn silk on oxidative stress induced by exhaustive exercise in mice

    The present study aims at exploring the effects of flavonoids from corn silk (FCS) on oxidative stress induced by exhaustive exercise in mice. Sixty mice were randomized into 3 groups: Low-dose FCS treatment group (LFG), high-dose FCS treatment group (HFG) and control group (CG). The mice were treated with FCS (100 and 400 mg/kg) or placebo (distilled water) by daily oral gavage for 28 days. After the last treatment, animals were submitted to treadmill for exhaustion and running time, malondialdehyde (MDA), superoxide dismutase (SOD) and glutathione peroxidase (GPX) levels were measured. The results suggested that FCS could elevate the exercise tolerance of mice, and provide protection against oxidative stress induced by exhaustive exercise in mice, by inhibiting lipid per-oxidation and increasing anti-oxidant enzymes levels.Key words: Flavonoids from corn silk, oxidative stress, exhaustive exercise, mic

    Effectively predicting whether and when a topic will become prevalent in a social network

Effective forecasting of future prevalent topics plays an important role in social network business development. It involves two challenging aspects: predicting whether a topic will become prevalent, and when. This cannot be directly handled by the existing algorithms in topic modeling, item recommendation and action forecasting. The classic forecasting framework based on time series models may be able to predict a hot topic when a series of periodical changes to user-addressed frequency in a systematic way. However, the frequency of topics discussed by users often changes irregularly in social networks. In this paper, a generic probabilistic framework is proposed for hot topic prediction, and machine learning methods are explored to predict hot topic patterns. Two effective models, PreWHether and PreWHen, are introduced to predict whether and when a topic will become prevalent. In the PreWHether model, we simulate the constructed features of previously observed frequency changes for better prediction. In the PreWHen model, distributions of time intervals associated with the emergence to prevalence of a topic are modeled. Extensive experiments on real dataseis demonstrate that our method outperforms the baselines and generates more effective predictions

    Characterization and phylogenetic analysis of the chitinase gene from the Helicoverpa armigera single nucleocapsid nucleopolyhedrovirus

    A putative chitinase gene was identified within the fragment EcoRI-K of the Helicoverpa armigera single-nucleocapsid nucleopolyhedrovirus (HearNPV, also called HaSNPIV) genome. The open reading frame (ORF) contains 1713 nucleotides (nt) and encodes a protein of 570 amino acids (aa) with a predicted molecular weight of 63.6 kDa. Transcription started at about 18 h post infection (p.i.) and the protein was first detected at 20 h p.i. The times of transcription and expression are characteristic of a late baculovirus gene. 5' and 3' RACE indicated that transcription was initiated from the adenine residue located at -246 nt upstream from the ATG start site and the poly (A) tail was added at 267 nt downstream from the stop codon. This is the first report on the molecular characterization of a chitinase from a single nucleocapsid NPV. The phylogeny of baculoviral chitinase genes were extensively examined in comparison with chitinases derived from bacteria, fungi, nematode, actinomycetes, viruses, insects and mammals. Neighbor-joining and most parsimony analyses showed that the baculoviral chitinases were clustered exclusively within gamma-proteobacteria. Our results strongly suggest that baculoviruses acquired their chitinase genes from bacteria. (C) 2004 Elsevier B.V.     食管癌原發灶與淋巴結轉移灶細胞染色體變化特征的比較

    BACKGROUND & OBJECTIVE: Local lymph node and blood metastasis could occur at early stage of esophageal squamous cell carcinoma (ESCC), which may be the key factors of its recurrence and poor prognosis. However, the mechanism of ESCC metastasis is unclear. This study was to analyze the genetic changes in primary lesion and lymph node metastases of ESCC, to screen for and locate ESCC metastasis-related genes. METHODS: Genomic alterations in 15 pairs of primary lesions and matched metastatic lymph nodes of ESCC were analyzed by comparative genomic hybridization (CGH). RESULTS: In the 15 pairs of tissues, the most common chromosomal alterations were the gains of 3q, 8q, 6p, 20p, 5p, 18p, 2p, 2q and 1q, and the losses of 10p, 10q, 17p, 18q, 4p and 13q. Of these changes, the most significant finding was the gain of 6p with a frequency of 47% in metastatic lymph nodes and 13% in primary lesions, and the gain of 20p with a frequency of 73% in metastatic lymph nodes and 33% in primary lesions. The second interesting finding was the loss of 10p with a frequency of 53% in metastatic lymph nodes and 13% in primary lesions, and the loss of 10q with a frequency of 47% in metastatic lymph nodes and 13% in primary lesions. CONCLUSION: The gains of 6p and 20p and the losses of 10p and 10q are common genomic alterations in primary lesion and lymph node metastases of ESCC, which may code ESCC metastasis-related genes.背景與目的:食管癌早期可發生局部淋巴或血行轉移,這是導致復發和預后差的主要原因。但是,食管癌轉移發生的分子機制尚不清楚。本研究旨在分析食管癌原發灶和淋巴結轉移灶腫瘤細胞染色體變化的特征,尋找或定位與食管癌轉移相關基因,加深對其轉移機制的了解。方法:應用比較基因組雜交技術(comparativegenomichybridization,CGH)分析15例食管癌患者原發灶和其對應的淋巴結轉移灶的染色體基因組改變。結果:最常見染色體DNA拷貝數增加的部位是3q,8q,6p,20p,5p,18p,2p,2q,1q;常見的染色體DNA拷貝數丟失的部位是10p,10q,17p,18q,4p,13q。其中,最有意義的發現是6p增加(原發灶:2/15,13%,轉移灶:7/15,47%),20p增加(原發灶:5/15,33.3%,轉移灶:11/15,73.3%)。第二個發現是10p丟失(原發灶:2/15,13.3%,轉移灶:8/15,53%),10q丟失(原發灶:2/15,13.3%,轉移灶:7/15,46.6%)。結論:食管癌原發灶和淋巴結轉移灶細胞染色體基因組改變最顯著的部位是6p,20p的增加和10p,10q的丟失;這些部位可能存在與食管癌細胞淋巴結轉移相關的基因。link_to_subscribed_fulltex

    Heterogeneity of Paucigranulocytic Asthma: A Prospective Cohort Study with Hierarchical Cluster Analysis.

    BACKGROUND: Asthma, a heterogeneous disease, can be divided into 4 inflammatory phenotypes using induced sputum cell counts-eosinophilic asthma (EA), neutrophilic asthma (NA), mixed granulocytic asthma, and paucigranulocytic asthma (PGA). Although research has focused on EA and NA, there is little known about PGA. OBJECTIVE: To study the heterogeneity of PGA and identify possible PGA clusters to guide clinical treatment. METHODS: Patients with PGA were grouped by hierarchical cluster analysis and enrolled into a prospective cohort study to validate the clusters, relative to future risk of asthma exacerbations in a real-world setting. Clusters were validated by tree analysis in a separate population. Finally, we explored PGA stability. RESULTS: Cluster analysis of 145 patients with PGA identified 3 clusters: cluster 1 (n = 110, 75.9%) was "mild PGA," cluster 2 (n = 20, 13.8%) was "PGA with psychological dysfunction and rhinoconjunctivitis and other allergic diseases," and cluster 3 (n = 15, 10.3%) was "smoking-associated PGA." Cluster 3 had significantly increased risk of severe exacerbation (relative risk [RR] = 6.43, P = .01), emergency visit (RR = 8.61, P = .03), and hospitalization (RR = 12.94, P < .01). Results of the cluster analysis were successfully validated in an independent PGA population classified using decision tree analysis. Although PGA can transform into or develop from other phenotypes, 70% were stable over time. CONCLUSIONS: Among 3 identified PGA clusters, cluster 3 had a higher risk of severe exacerbation. PGA heterogeneity indicates the requirement of novel targeted interventions

    A signal regularity-based automated seizure prediction algorithm using long-term scalp EEG recordings

    The purpose of this study was to evaluate a signal regularity-based automated seizure prediction algorithm for scalp EEG. Signal regularity was quantified using the Pattern Match Regularity Statistic (PMRS), a statistical measure. The primary feature of the prediction algorithm is the degree of convergence in PMRS (“PMRS entrainment”) among the electrode groups determined in the algorithm training process. The hypothesis is that the PMRS entrainment increases during the transition between interictal and ictal states, and therefore may serve as an indicator for prediction of an impending seizure.Запропоновано алгоритм автоматизованого прогнозування епілептичного нападу на основі аналізу регулярності сигналу ЕЕГ головного мозку. Регулярність сигналу розраховується на основі введеної величини регулярної статистики збігу фрагментів (Pattern Match Regularity Statistics — PMRS). Відмінною рисою алгоритму є ступінь збіжності в значеннях PMRS, розрахованих на основі показань із різних груп електродів, визначених у процесі навчання алгоритму. В основі алгоритму лежить гіпотеза про те, що збіжність у значеннях величини PMRS збільшується під час переходу в стан нападу і в такий спосіб може слугувати індикатором для прогнозування нападу
