Penelitian menelaah salah satu bentuk karya sastra yaitu Film Snow White and Huntsman karya Rupert Sanders. Para penikmat film kerap kali mengabaikan peran tanda semiotik yang digunakan dalam sebuah film. Ketidakpedulian akan peran tanda-tanda semiotik inilah yang mencemeti penelaahan dalam Film Snow White and The Huntsman. Perian tentang tanda-tanda semiotik seperti ikon, indeks dan simbol sebagai pelantang pesan penulis diurai tuntas. Penelitian Film The Snow White and The Huntsman menggunakan Teori Semiotic oleh Sanders Pierce. Hasil analisis membuktikan bahwa (1) tidak semua adegan memiliki ketiga tanda semiotik ini, (2) meskipun tidak semua adegan memiliki kektiga tanda semiotik ini tetapi peran ketida semiotik sangat dominan sebagai pelantang pesan penulis, dan (3) pemaknaan terhadap ketiga tanda semiotik ini hendaknya dilakukan secara komprehensif dan holistik.  &nbsp

    Non-classical ProIL-1beta activation during mammary gland infection is pathogen-dependent but caspase-1 independent

    Infection of the mammary gland with live bacteria elicits a pathogen-specific host inflammatory response. To study these host-pathogen interactions wild type mice, NF-kappaB reporter mice as well as caspase-1 and IL-1beta knockout mice were intramammarily challenged with Escherichia coli (E. coli) and Staphylococcus aureus (S. aureus). The murine mastitis model allowed to compare the kinetics of the induced cytokine protein profiles and their underlying pathways. In vivo and ex vivo imaging showed that E. coli rapidly induced NF-kappaB inflammatory signaling concomitant with high mammary levels of TNF-alpha, IL-1 alpha and MCP-1 as determined by multiplex analysis. In contrast, an equal number of S. aureus bacteria induced a low NF-kappaB activity concomitant with high mammary levels of the classical IL-1beta fragment. These quantitative and qualitative differences in local inflammatory mediators resulted in an earlier neutrophil influx and in a more extensive alveolar damage post-infection with E. coli compared to S. aureus. Western blot analysis revealed that the inactive proIL-1beta precursor was processed into pathogen-specific IL-1beta fragmentation patterns as confirmed with IL-1beta knockout animals. Additionally, caspase-1 knockout animals allowed to investigate whether IL-1beta maturation depended on the conventional inflammasome pathway. The lack of caspase-1 did not prevent extensive proIL-1beta fragmentation by either of S. aureus or E. coli. These non-classical IL-1beta patterns were likely caused by different proteases and suggest a sentinel function of IL-1beta during mammary gland infection. Thus, a key signaling nodule can be defined in the differential host innate immune defense upon E. coli versus S. aureus mammary gland infection, which is independent of caspase-1

    Good reproductive health will improve reproductive health and prevent reproductive health problems. The Indonesian Family Planning Association survey and On Track Media Indonesia (OTMI) found that around 29% to 31% of adolescents in East Nusa Tenggara had premarital sex. The impact of poor reproductive health behavior is unwanted pregnancy and sexually transmitted infection. This is a quantitative descriptive study that aimed to describe reproductive health behavior in high school students in Kupang City. The population in this study were high school students from grade XI and XII (6863 students) and a sample of 378 students taken by proportionate stratified random sampling technique. The results showed a description of Genital Personal Hygiene behavior in high school students in Kupang City as good is 249 students (66%), while less is 129 students (34%). The pre-marital sexual behavior of high school students in Kupang City was at a risk is 85 students (29%) and 293 students (71%) were not at risk. The behavior of watching porn videos for high school students in Kupang City is 217 students (57.4%) have watched and 161 students (42.6%) have not watched porn videos/movies. Behavior discussing reproductive health among high school students in Kupang City is 216 students (57%) discussing reproductive health and 162 students (43%) do not discuss reproductive health.     The SECURE project – Stem canker of oilseed rape: : molecular methods and mathematical modelling to deploy durable resistance

Experiments done with near-isogenic lines of L. maculans to investigate pathogen fitness support field data that suggest a positive effect of the avirulence allele AvrLm4 on pathogen fitness, and that the loss of this allele renders isolates less competitive under field conditions on cultivars without the resistance gene Rlm4. The highlight of molecular work was the cloning of AvrLm1 and AvrLm6. L. maculans is now one of the few fungal species for which two avirulence loci have been cloned. Subsequent research focused on understanding the function of AvrLm1 and AvrLm6 and on the analysis of sequences of virulent isolates to understand molecular evolution towards virulence. Isolates of L. maculans transformed with GFP and/or DsRed were used to follow growth of the fungus in B. napus near-isogenic-lines (NIL) with or without MX (Rlm6) resistance under different temperature and wetness conditions. The results greatly enhanced our knowledge of the infection process and the rate and extent of in planta growth on different cultivars. Conclusions from work to model durability of resistance have been tested under field conditions through a series of experiments to compare durability of resistance conferred by the major resistance gene Rlm6 alone in a susceptible background (EurolMX) or in a resistant background (DarmorMX) under recurrent selection over 4 growing seasons. A major priority of the project was knowledge transfer of results and recommendations to target audiences such as plant breeding companies and extension services. CETIOM developed a “diversification scheme” that encourages French growers to make an informed choice about the cultivars that are grown within the rotation based on the resistance genes carried by the individual cultivars. Use of such schemes, in association with survey data on the population structure of L. maculans at both national and European scales will provide opportunities for breeders and the industry to manage available B. napus resistance more effectively.Non peer reviewe

    Carbohydrate scaffolds as glycosyltransferase inhibitors with in vivo antibacterial activity

    The rapid rise of multi-drug-resistant bacteria is a global healthcare crisis, and new antibiotics are urgently required, especially those with modes of action that have low-resistance potential. One promising lead is the liposaccharide antibiotic moenomycin that inhibits bacterial glycosyltransferases, which are essential for peptidoglycan polymerization, while displaying a low rate of resistance. Unfortunately, the lipophilicity of moenomycin leads to unfavourable pharmacokinetic properties that render it unsuitable for systemic administration. In this study, we show that using moenomycin and other glycosyltransferase inhibitors as templates, we were able to synthesize compound libraries based on novel pyranose scaffold chemistry, with moenomycin-like activity, but with improved drug-like properties. The novel compounds exhibit in vitro inhibition comparable to moenomycin, with low toxicity and good efficacy in several in vivo models of infection. This approach based on non-planar carbohydrate scaffolds provides a new opportunity to develop new antibiotics with low propensity for resistance induction

    Artificial intelligence for neurodegenerative experimental models

    INTRODUCTION: Experimental models are essential tools in neurodegenerative disease research. However, the translation of insights and drugs discovered in model systems has proven immensely challenging, marred by high failure rates in human clinical trials. METHODS: Here we review the application of artificial intelligence (AI) and machine learning (ML) in experimental medicine for dementia research. RESULTS: Considering the specific challenges of reproducibility and translation between other species or model systems and human biology in preclinical dementia research, we highlight best practices and resources that can be leveraged to quantify and evaluate translatability. We then evaluate how AI and ML approaches could be applied to enhance both cross-model reproducibility and translation to human biology, while sustaining biological interpretability. DISCUSSION: AI and ML approaches in experimental medicine remain in their infancy. However, they have great potential to strengthen preclinical research and translation if based upon adequate, robust, and reproducible experimental data. HIGHLIGHTS: There are increasing applications of AI in experimental medicine. We identified issues in reproducibility, cross-species translation, and data curation in the field. Our review highlights data resources and AI approaches as solutions. Multi-omics analysis with AI offers exciting future possibilities in drug discovery

    Artificial intelligence for dementia genetics and omics

    Genetics and omics studies of Alzheimer's disease and other dementia subtypes enhance our understanding of underlying mechanisms and pathways that can be targeted. We identified key remaining challenges: First, can we enhance genetic studies to address missing heritability? Can we identify reproducible omics signatures that differentiate between dementia subtypes? Can high-dimensional omics data identify improved biomarkers? How can genetics inform our understanding of causal status of dementia risk factors? And which biological processes are altered by dementia-related genetic variation? Artificial intelligence (AI) and machine learning approaches give us powerful new tools in helping us to tackle these challenges, and we review possible solutions and examples of best practice. However, their limitations also need to be considered, as well as the need for coordinated multidisciplinary research and diverse deeply phenotyped cohorts. Ultimately AI approaches improve our ability to interrogate genetics and omics data for precision dementia medicine

    Caspase deficiency alters the murine gut microbiome

    Caspases are aspartate-specific cysteine proteases that have an essential role in apoptosis and inflammation, and contribute to the maintenance of homeostasis in the intestine. These facts, together with the knowledge that caspases are implicated in host-microbe crosstalk, prompted us to investigate the effect of caspase (Casp)1, -3 and -7 deficiency on the composition of the murine gut microbiota. We observed significant changes in the abundance of the Firmicutes and Bacteroidetes phyla, in particular the Lachnospiraceae, Porphyromonodaceae and Prevotellacea families, when comparing Casp-1, -7 and -3 knockout mice with wild-type mice.
