    A Yaminahua Autobiographical Song: Caqui Caqui

    This essay proposes a theoretical framework of the relationship between autobiography and tradition. In support of this framework, I present a transcription and translation of an autobiographical song recorded among the Yaminahua (in the Western Amazon of Peru). My account of this song allows me to define an original discursive genre: the traditional autobiography. Two contradictory features characterize this genre. On the one hand, it is autobiographical because it makes reference to the personal life of the speaker. On the other hand, it is traditional in the sense that it is inherited from another speaker and faithfully reproduced. I explore the implications of this apparent paradox. En prenant appui sur la transcription et la traduction d\u27un chant yaminahua (peuple d\u27Amazonie péruvienne, de langue pano), je propose de réfléchir à la relation qu\u27un récit autobiographique peut entretenir avec la tradition orale. Une telle réflexion m\u27amène a proposer une catégorie discursive originale, celle d\u27autobiographie traditionnelle, où d\u27une part le discours est autobiographique puisqu\u27il se réfère à la vie du locuteur, mais où d\u27autre part il doit être défini comme traditionnel puisqu\u27il est hérité d\u27un autre locuteur et intégralement répété.     There are three reasons why it is well worth rereading an ancient Greek text by Epicure: firstly, becauseour civilization is marked by the crisis of European science, nihilism, post-modern culture and barbarousreasoning; secondly, because 21st century philosophical thought is primarily concerned with examiningour relationship to nature, the “natural question”, and the code of ethics best suited to our technologicalcivilization; and lastly because it is necessary to ascertain the presuppositions and implications involvedin research aimed at technoscientific development. In a letter Epicure wrote in the 4th century BC to hisfriend Ménécée, he outlines the rules to be followed for living a pleasurable life. These rules happen tobe the standard ecological ideal for living in harmony with the earth of ours.     Voltage-gated sodium channels are dynamic membrane proteins essential for signaling in nervous and muscular systems. They undergo substantial conformational changes associated with the closed, open and inactivated states. However, little information is available regarding their conformational stability. In this study circular dichroism spectroscopy was used to investigate the changes in secondary structure accompanying chemical and thermal denaturation of detergent-solubilised sodium channels isolated from Electrophorus electricus electroplax. The proteins appear to be remarkably resistant to either type of treatment, with "denatured" channels, retaining significant helical secondary structure even at 77 degrees C or in 10% SDS. Further retention of helical secondary structure at high temperature was observed in the presence of the channel-blocking tetrodotoxin. It was possible to refold the thermally-denatured (but not chemically-denatured) channels in vitro. The correctly refolded channels were capable of undergoing the toxin-induced conformational change indicative of ligand binding. In addition, flux measurements in liposomes showed that the thermally-denatured (but not chemically-denatured) proteins were able to re-adopt native, active conformations. These studies suggest that whilst sodium channels must be sufficiently flexible to undergo major conformational changes during their functional cycle, the proteins are highly resistant to unfolding, a feature that is important for maintaining structural integrity during dynamic processes.     BYKdb: the Bacterial protein tYrosine Kinase database

    Bacterial tyrosine-kinases share no resemblance with their eukaryotic counterparts and they have been unified in a new protein family named BY-kinases. These enzymes have been shown to control several biological functions in the bacterial cells. In recent years biochemical studies, sequence analyses and structure resolutions allowed the deciphering of a common signature. However, BY-kinase sequence annotations in primary databases remain incomplete. This prompted us to develop a specialized database of computer-annotated BY-kinase sequences: the Bacterial protein tyrosine-kinase database (BYKdb). BY-kinase sequences are first identified, thanks to a workflow developed in a previous work. A second workflow annotates the UniProtKB entries in order to provide the BYKdb entries. The database can be accessed through a web interface that allows static and dynamic queries and offers integrated sequence analysis tools. BYKdb can be found at http://bykdb.ibcp.fr
