Background suppression in massive TeO bolometers with Neganov-Luke amplified light detectors
Bolometric detectors are excellent devices for the investigation of
neutrinoless double-beta decay (0). The observation of such
decay would demonstrate the violation of lepton number, and at the same time it
would necessarily imply that neutrinos have a Majorana character. The
sensitivity of cryogenic detectors based on TeO is strongly limited by the
alpha background in the region of interest for the 0 of
Te. It has been demonstrated that particle discrimination in TeO
bolometers is possible measuring the Cherenkov light produced by particle
interactions. However an event-by-event discrimination with NTD-based light
detectors has to be demonstrated. We will discuss the performance of a
highly-sensitive light detector exploiting the Neganov-Luke effect for signal
amplification. The detector, being operated with NTD-thermistor and coupled to
a 750 g TeO crystal, shows the ability for an event-by-event identification
of electron/gamma and alpha particles. The extremely low detector baseline
noise, RMS 19 eV, demonstrates the possibility to enhance the sensitivity of
TeO-based 0 experiment to an unprecedented level
Comparison of short- and long-term effectiveness of ixekizumab and secukinumab in real-world practice
Background: Although secukinumab and ixekizumab both act by inhibiting IL-17A, some scientific evidence suggests that there are differences in efficacy between the two agents. Objective: The aim of this study was to compare the short- and long-term effectiveness of ixekizumab and secukinumab in clinical practice. Methods: A retrospective study was conducted on a cohort of 245 psoriatic patients receiving secukinumab or ixekizumab during the period from September 2016 to December 2019. The proportion of patients achieving PASI75, PASI90, and PASI100 at weeks 12 and 24 was calculated. We recorded the 12- and 24-month drug survival as a measure to assess long-term effectiveness. Results: A higher proportion of patients in the secukinumab group achieved PASI75, 90, and 100 at 12 weeks. The Kaplan-Meier survival curve for any reason of discontinuation showed no differences between the two groups. Instead, the multivariate analysis for ineffectiveness, adjusted for potential confounders, showed a lower drug survival rate in the secukinumab group, with an adjusted HR of 2.57 (95% CI 1.05–6.28, p 0.038). Conclusion: This real-life study demonstrated that ixekizumab and secukinumab are both highly effective in short- and long-term treatment of psoriasis, even though few differences exist concerning speed of action and long-term effectiveness
práticas de comunicação do professor
O objetivo deste projeto de investigação prende-se com a compreensão das minhas práticas de comunicação no âmbito de discussões orais coletivas em Português, considerando as ações discursivas e a comunicação multimodal do professor, bem como as principais inquietações experienciadas durante a dinamização das atividades.
O enquadramento teórico é composto por duas secções. A primeira refere-se ao ato comunicativo na sala de aula, onde se esclarece a importância da oralidade e da comunicação multimodal. A segunda apresenta o conceito de discussão oral coletiva, os pressupostos e as práticas do professor nas suas diferentes fases, bem como as práticas de comunicação do professor na dinamização de discussões orais coletivas em Português.
A metodologia adotada insere-se num paradigma interpretativo de natureza qualitativa e corresponde a uma investigação sobre a própria prática. A recolha de dados foi efetuada recorrendo à observação-participante, a entrevistas e à análise documental. Os participantes do projeto foram os alunos de uma turma de 5.º ano de escolaridade e eu própria, enquanto professora estagiária da turma. O projeto de investigação foi desenvolvido a partir da seleção de três atividades de uma sequência didática que foram dinamizadas com diferentes objetivos e metodologias de trabalho.
Os resultados do projeto evidenciam que, na preparação das atividades, as interações entre professora-alunos, alunos-professora e alunos-alunos, a ação discursiva questionar e os modos semióticos verbais e não-verbais foram antecipados e que esta prática foi fundamental para conseguir gerir as intervenções dos alunos com uma maior segurança no momento de dinamização das atividades, quando fui confrontada com inquietações relacionadas com o desinteresse dos alunos e com a gestão dos modos semióticos utilizados. Os resultados evidenciam, ainda, que, nas interações entre professora-alunos, as questões do tipo “Como?” e “Porquê?” foram as mais recorrentes, por apresentarem um maior potencial para desafiarem os alunos a discutirem os temas, e que os modos verbal, paraverbal e não-verbal se complementaram, auxiliando os alunos nas suas reflexões e na construção de significados coletivos.The goal of this investigation project is the understanding of my comunication practices in the context of colective oral discussions in Portuguese, considering the discursive actions and the multimodal communication of the teacher, as well as the main concerns experienced during the dinamization of the activities.
The theoretical framing is composed of two sections. The first one refers to the communicative act in the classroom, where the importance of the orality and multimodal communication becomes clear. The second one presents the concept of collective oral discussion, the presuppositions and the practices of the teacher in its different phases, as well as the teacher communication practices in the dinamization of colective oral discussions in Portuguese.
The adopted methodology is inserted into an interpretative paradigm of qualitative nature and corresponds to an investigation about the practice itself. The data gathering was done using participative-observation, interviews and document analysis. The project participants were the students of a 5th grade class and myself, as an intern teacher of the class. The investigation project was developed by the selection of three activities of a didatic sequence that was performed with different goals and working methodologies.
The results of the project show that, in the preparation of the activities, the interactions between teacher-students, students-teacher and students-students, the discursive action questioning and the verbal and non-verbal semiotic modes were anticipated and that this practice was fundamental to be able to manage the students interventions with a higher safety, in the moment of the dinamization of the activities, when I was confronted with concerns related with the disinterest of the students and the management of the semiotic modes used. The results still show that, in the interactions between teacher-students, the questions like "How?" and "Why?" were the most recurrent, for presenting a higher potential to challenge the students to discuss the subjects, and that the verbal, paraverbal and non-verbal modes complemented each other, helping the students in their reflections and in the construction of collective meanings
An improved non-invasive method for measuring heartbeat of intertidal animals
Since its emergence two decades ago, the use of infrared technology for noninvasively measuring the heartbeat
rates of invertebrates has provided valuable insight into the physiology and ecology of intertidal organisms.
During that time period, the hardware needed for this method has been adapted to currently available electronic
components, making the original published description obsolete. This article reviews the history of heartbeat sensing
technology, and describes the design and function of a modern and simplified infrared heartbeat rate sensing
system compatible with many intertidal and marine invertebrates. This technique overcomes drawbacks and
obstacles encountered with previous methods of heartbeat rate measurement, and due to the sensor’s small size,
versatility, and noninvasive nature, it creates new possibilities for studies across a wide range of organismal type
A New Limit on the Neutrinoless DBD of 130Te
We report the present results of CUORICINO a cryogenic experiment on
neutrinoless double beta decay (DBD) of 130Te consisting of an array of 62
crystals of TeO2 with a total active mass of 40.7 kg. The array is framed
inside of a dilution refrigerator, heavily shielded against environmental
radioactivity and high-energy neutrons, and operated at a temperature of ~8 mK
in the Gran Sasso Underground Laboratory. Temperature pulses induced by
particle interacting in the crystals are recorded and measured by means of
Neutron Transmutation Doped thermistors. The gain of each bolometer is
stabilized with voltage pulses developed by a high stability pulse generator
across heater resistors put in thermal contact with the absorber.
The calibration is performed by means of two thoriated wires routinely
inserted in the set-up. No evidence for a peak indicating neutrinoless DBD of
130Te is detected and a 90% C.L. lower limit of 1.8E24 years is set for the
lifetime of this process. Taking largely into account the uncertainties in the
theoretical values of nuclear matrix elements, this implies an upper boud on
the effective mass of the electron neutrino ranging from 0.2 to 1.1 eV. This
sensitivity is similar to those of the 76Ge experiments.Comment: 4 pages, 2 figure
Maraviroc Intensification Modulates Atherosclerotic Progression in HIV-Suppressed Patients at High Cardiovascular Risk. A Randomized, Crossover Pilot Study
Background
Experimental CCR5 antagonism with maraviroc in atherosclerosis-prone mice and preliminary data in humans suggest an anti-atherosclerotic effect of the drug. We assessed the impact of maraviroc treatment in persons living with HIV on subclinical indicators of atherosclerosis.
Methods
Persons living with HIV on effective antiretroviral therapy (ART) including only protease inhibitors were recruited if they had a Framingham risk score >20% and brachial flow-mediated dilation (bFMD) <4%, as indices of high cardiovascular risk. Maraviroc (300 mg per os for 24 weeks) was administered, in addition to ongoing ART, to all patients using a crossover design. Brachial FMD, carotid-femoral pulse wave velocity (cfPWV), and carotid intima-media thickness (cIMT) were measured as markers of atherosclerosis. Vascular competence—as expressed by the ratio of circulating endothelial microparticles (EMPs) to endothelial progenitor cells (EPCs)—and markers of systemic inflammation and monocyte and platelet activation were assessed.
Results
Maraviroc treatment significantly improved bFMD, cfPWV, and cIMT by 66%, 11%, and 13%, respectively (P = .002, P = .022, P = .038, respectively). We also found a beneficial effect of maraviroc on the EMP/EPC ratio (P < .001) and platelet/leucocyte aggregates (P = .013). No significant changes in markers of systemic inflammation, monocyte activation, and microbial translocation were observed.
Conclusions
Maraviroc led to significant improvements in several markers for cardiovascular risk, endothelial dysfunction, arterial stiffness, and early carotid atherosclerosis, which was accompanied by an increase of vascular competence, without seeming to affect systemic inflammation. Our data support the need for larger studies to test for any effects of maraviroc on preventing atherosclerosis-driven pathologies
Oxonium ion guided analysis of quantitative proteomics data reveals site-specific O-glycosylation of anterior gradient protein 2 (AGR2)
Developments in mass spectrometry (MS)-based analyses of glycoproteins have been important to study changes in glycosylation related to disease. Recently, the characteristic pattern of oxonium ions in glycopeptide fragmentation spectra had been used to assign different sets of glycopeptides. In particular, this was helpful to discriminate between O-GalNAc and O-GlcNAc. Here, we thought to investigate how such information can be used to examine quantitative proteomics data. For this purpose, we used tandem mass tag (TMT)-labeled samples from total cell lysates and secreted proteins from three different colorectal cancer cell lines. Following automated glycopeptide assignment (Byonic) and evaluation of the presence and relative intensity of oxonium ions, we observed that, in particular, the ratio of the ions at m/z 144.066 and 138.055, respectively, could be used to discriminate between O-GlcNAcylated and O-GalNAcylated peptides, with concomitant relative quantification between the different cell lines. Among the O-GalNAcylated proteins, we also observed anterior gradient protein 2 (AGR2), a protein which glycosylation site and status was hitherto not well documented. Using a combination of multiple fragmentation methods, we then not only assigned the site of modification, but also showed different glycosylation between intracellular (ER-resident) and secreted AGR2. Overall, our study shows the potential of broad application of the use of the relative intensities of oxonium ions for the confident assignment of glycopeptides, even in complex proteomics datasets.Proteomic
Proposal for taking data with the KLOE-2 detector at the DANE collider upgraded in energy
This document reviews the physics program of the KLOE-2 detector at
DANE upgraded in energy and provides a simple solution to run the
collider above the -peak (up to 2, possibly 2.5 GeV). It is shown how a
precise measurement of the multihadronic cross section in the energy region up
to 2 (possibly 2.5) GeV would have a major impact on the tests of the Standard
Model through a precise determination of the anomalous magnetic moment of the
muon and the effective fine-structure constant at the scale. With a
luminosity of about cms, DANE upgraded in energy
can perform a scan in the region from 1 to 2.5 GeV in one year by collecting an
integrated luminosity of 20 pb (corresponding to a few days of data
taking) for single point, assuming an energy step of 25 MeV. A few years of
data taking in this region would provide important tests of QCD and effective
theories by physics with open thresholds for pseudo-scalar (like
the ), scalar (, etc...) and axial-vector (, etc...)
mesons; vector-mesons spectroscopy and baryon form factors; tests of CVC and
searches for exotics. In the final part of the document a technical solution
