    Combined acute hyperglycemic and hyperinsulinemic clamp induced profibrotic and proinflammatory responses in the kidney

    Increase in matrix protein content in the kidney is a cardinal feature of diabetic kidney disease. While renal matrix protein content is increased by chronic hyperglycemia, whether it is regulated by acute elevation of glucose and insulin has not been addressed. In this study, we aimed to evaluate whether short duration of combined hyperglycemia and hyperinsulinemia, mimicking the metabolic environment of prediabetes and early type 2 diabetes, induces kidney injury. Normal rats were subjected to either saline infusion (control, n = 4) or 7 h of combined hyperglycemic- hyperinsulinemic clamp (HG+HI clamp; n = 6). During the clamp, plasma glucose and plasma insulin were maintained at about 350 mg/dl and 16 ng/ml, respectively. HG+HI clamp increased the expression of renal cortical transforming growth factor-β (TGF-β) and renal matrix proteins, laminin and fibronectin. This was associated with the activation of SMAD3, Akt, mammalian target of rapamycin (mTOR) complexes, and ERK signaling pathways and their downstream target events in the initiation and elongation phases of mRNA translation, an important step in protein synthesis. Additionally, HG+HI clamp provoked renal inflammation as shown by the activation of Toll-like receptor 4 (TLR4) and infiltration of CD68-positive monocytes. Urinary F2t isoprostane excretion, an index of renal oxidant stress, was increased in the HG+HI clamp rats. We conclude that even a short duration of hyperglycemia and hyperinsulinemia contributes to activation of pathways that regulate matrix protein synthesis, inflammation, and oxidative stress in the kidney. This finding could have implications for the control of short-term rises in blood glucose in diabetic individuals at risk of developing kidney disease. © 2014 the American Physiological Society

    Cygnus X-3 transition from the ultrasoft to the hard state

    Aims: The nature of Cygnus X-3 is still not understood well. This binary system might host a black hole or a neutron star. Recent observations by INTEGRAL have shown that Cygnus X-3 was again in an extremely ultrasoft state. Here we present our analysis of the transition from the ultrasoft state, dominated by blackbody radiation at soft X-rays plus non-thermal emission in the hard X-rays, to the low hard state. Methods: INTEGRAL observed Cyg X-3 six times during three weeks in late May and early June 2007. Data from IBIS/ISGRI and JEM-X1 were analysed to show the spectral transition. Results: During the ultrasoft state, the soft X-ray spectrum is well-described by an absorbed (NH = 1.5E22 1/cm**2) black body model, whereas the X-ray spectrum above 20 keV appears to be extremely low and hard (Gamma = 1.7). During the transition, the radio flux rises to a level of >1 Jy, and the soft X-ray emission drops by a factor of 3, while the hard X-ray emission rises by a factor of 14 and becomes steeper (up to Gamma = 4). Conclusions: The ultrasoft state apparently precedes the emission of a jet, which is apparent in the radio and hard X-ray domain.Comment: 4 pages, 3 figures, accepted for publication as A&A Research Not

    Automatic extraction of informal topics from online suicidal ideation

    Abstract Background Suicide is an alarming public health problem accounting for a considerable number of deaths each year worldwide. Many more individuals contemplate suicide. Understanding the attributes, characteristics, and exposures correlated with suicide remains an urgent and significant problem. As social networking sites have become more common, users have adopted these sites to talk about intensely personal topics, among them their thoughts about suicide. Such data has previously been evaluated by analyzing the language features of social media posts and using factors derived by domain experts to identify at-risk users. Results In this work, we automatically extract informal latent recurring topics of suicidal ideation found in social media posts. Our evaluation demonstrates that we are able to automatically reproduce many of the expertly determined risk factors for suicide. Moreover, we identify many informal latent topics related to suicide ideation such as concerns over health, work, self-image, and financial issues. Conclusions These informal topics topics can be more specific or more general. Some of our topics express meaningful ideas not contained in the risk factors and some risk factors do not have complimentary latent topics. In short, our analysis of the latent topics extracted from social media containing suicidal ideations suggests that users of these systems express ideas that are complementary to the topics defined by experts but differ in their scope, focus, and precision of language.https://deepblue.lib.umich.edu/bitstream/2027.42/144214/1/12859_2018_Article_2197.pd

    Finding information about mental health in microblogging platforms: a Case study of depression

    Searching for online health information has been well studied in web search, but social media, such as public microblogging services, are well known for different types of tacit information: personal experience and shared information. Finding useful information in public microblogging platforms is an on-going hard problem and so to begin to develop a better model of what health information can be found, Twitter posts using the word “depression” were examined as a case study of a search for a prevalent mental health issue. 13,279 public tweets were analysed using a mixed methods approach and compared to a general sample of tweets. First, a linguistic analysis suggested that tweets mentioning depression were typically anxious but not angry, and were less likely to be in the first person, indicating that most were not from individuals discussing their own depression. Second, to un-derstand what types of tweets can be found, an inductive thematic analysis revealed three major themes: 1) dissemi-nating information or link of information, 2) self-disclosing, and 3) the sharing of overall opinion; each had significantly different linguistic patterns. We conclude with a discussion of how different types of posts about mental health may be retrieved from public social media like Twitter

    We show that in theories of gravitation with torsion the helicity of fermion particles is not conserved and we calculate the probability of spin flip, which is related to the anti-symmetric part of affine connection.
