    Interactions of tectonic, igneous, and hydraulic processes in the North Tharsis Region of Mars

    Recent work on the north Tharsis of Mars has revealed a complex geologic history involving volcanism, tectonism, flooding, and mass wasting. Our detailed photogeologic analysis of this region found many previously unreported volcanic vents, volcaniclastic flows, irregular cracks, and minor pit chains; additional evidence that volcanic tectonic processes dominated this region throughout Martian geologic time; and the local involvement of these processes with surface and near surface water. Also, photoclinometric profiles were obtained within the region of troughs, simple grabens, and pit chains, as well as average spacings of pits along pit chains. These data were used together with techniques to estimate depths of crustal mechanical discontinuities that may have controlled the development of these features. In turn, such discontinuities may be controlled by stratigraphy, presence of water or ice, or chemical cementation

    Cholinergic suppression: A postsynaptic mechanism of long-term associative learning

    Food avoidance learning in the mollusc Pleurobranchaea entails reduction in the responsiveness of key brain interneurons in the feeding neural circuitry, the paracerebral feeding command interneurons (PCNs), to the neurotransmitter acetylcholine (AcCho). Food stimuli applied to the oral veil of an untrained animal depolarize the PCNs and induce the feeding motor program (FMP). Atropine (a muscarinic cholinergic antagonist) reversibly blocks the food-induced depolarization of the PCNs, implicating AcCho as the neurotransmitter mediating food detection. AcCho applied directly to PCN somata depolarizes them, indicating that the PCN soma membrane contains AcCho receptors and induces the FMP in the isolated central nervous system preparation. The AcCho response of the PCNs is mediated by muscariniclike receptors, since comparable depolarization is induced by muscarinic agonists (acetyl-ß -methylcholine, oxotremorine, pilocarpine), but not nicotine, and blocked by muscarinic antagonists (atropine, trifluoperazine). The nicotinic antagonist hexamethonium, however, blocked the AcCho response in four of six cases. When specimens are trained to suppress feeding behavior using a conventional food-avoidance learning paradigm (conditionally paired food and shock), AcCho applied to PCNs in the same concentration as in untrained animals causes little or no depolarization and does not initiate the FMP. Increasing the concentration of AcCho 10-100 times, however, induces weak PCN depolarization in trained specimens, indicating that learning diminishes but does not fully abolish AcCho responsiveness of the PCNs. This study proposes a cellular mechanism of long-term associative learning -- namely, postsynaptic modulation of neurotransmitter responsiveness in central neurons that could apply also to mammalian species

    Use of PCR to identify Leptospira in kidneys of big brown bats (Eptesicus fuscus) in Kansas and Nebraska, USA

    Bats have been implicated as potential carriers of Leptospira as a result of surveys, mostly in Australia and South America. We measured the prevalence of pathogenic leptospires in kidneys of bats from Kansas and Nebraska. From 7 August 2012 to 21 August 2012, we extracted DNA from kidneys of 98 big brown bats (Eptesicus fuscus) submitted and found negative for rabies. The DNA was processed in a two-step, seminested PCR assay with a dual-labeled Taqman probe specific for pathogenic leptospires. As a negative control, we used a saprophytic leptospire (Leptospira biflexa Patoc) and, as a pathogenic control, Leptospira interrogans Canicola. All bat kidneys were negative for pathogenic leptospires, suggesting that it is unlikely that the big brown bat, one of the most prevalent bat species in North America, is a reservoir for transmission of leptospires to dogs or humans

    Staurosporine Inhibits Frequency-Dependent Myofilament Desensitization in Intact Rabbit Cardiac Trabeculae

    Myofilament calcium sensitivity decreases with frequency in intact healthy rabbit trabeculae and associates with Troponin I and Myosin light chain-2 phosphorylation. We here tested whether serine-threonine kinase activity is primarily responsible for this frequency-dependent modulations of myofilament calcium sensitivity. Right ventricular trabeculae were isolated from New Zealand White rabbit hearts and iontophoretically loaded with bis-fura-2. Twitch force-calcium relationships and steady state force-calcium relationships were measured at frequencies of 1 and 4 Hz at 37 °C. Staurosporine (100 nM), a nonspecific serine-threonine kinase inhibitor, or vehicle (DMSO) was included in the superfusion solution before and during the contractures. Staurosporine had no frequency-dependent effect on force development, kinetics, calcium transient amplitude, or rate of calcium transient decline. The shift in the pCa50 of the force-calcium relationship was significant from 6.05 ± 0.04 at 1 Hz versus 5.88 ± 0.06 at 4 Hz under control conditions (vehicle, P < 0.001) but not in presence of staurosporine (5.89 ± 0.08 at 1 Hz versus 5.94 ± 0.07 at 4 Hz, P = NS). Phosphoprotein analysis (Pro-Q Diamond stain) confirmed that staurosporine significantly blunted the frequency-dependent phosphorylation at Troponin I and Myosin light chain-2. We conclude that frequency-dependent modulation of calcium sensitivity is mediated through a kinase-specific effect involving phosphorylation of myofilament proteins

    Effects of grain by-products as supplements for stocker cattle grazing bermudagrass

    Two experiments were conducted to compare corn, dried distillers’ grains (DDG), and pelleted soybean hulls (SH) as supplements for cattle grazing bermudagrass. In Exp. 1, 66 crossbred steers (306 ± 3.2 kg) were stratified by weight and allotted randomly to six 2.4-ha bermudagrass pastures for a 107-d study. One of three supplement treatments (corn, DDG, or SH) was assigned randomly to each pasture group and was offered at 0.5% (as fed) of body weight. Calves were weighed at 28-d intervals and supplement was adjusted after each weigh period. In Exp. 2, five ruminally cannulated steers grazed bermudagrass pasture and were individually fed supplements (corn, DDG, or SH) at 0.5% of body weight in a 3 x 3 replicated, incomplete Latin-square design with a 14-d adaptation and a 5-d sampling period. In Exp. 1, supplementation with DDG and corn increased (P \u3c 0.04) the average daily gain compared to supplementation with SH (0.89, 0.87, and 0.74 kg for DDG, corn, and SH, respectively). In Exp. 2, in situ dry-matter-disappearance kinetic measures of bermudagrass were not affected by type of supplementation. The potential extent of digestion for DDG (93%) was lower than for corn (97%, P = 0.01) and SH (96%, P = 0.06). Supplementation with corn or DDG at 0.5% of body weight improved the gain of stocker cattle grazing bermudagrass compared to supplementation with SH, but these differences were not explained by differences in bermudagrass degradation kinetic

    Mesoscale Hydrographic Variability in the Vicinity of Points Conception and Arguello During April May 1983: The OPUS 1983 Experiment

    In April and May 1983, interdisciplinary oceanographic observations were made of upwelling events in the Point Conception area off southern California. The principal objective was to observe the structure and time dependence of the upwelling system. To accomplish this, two ships, two aircraft, moorings, drogues, and satellite imagery were all included in the observational effort. During the intensive measurement period three main upwelling events and two intervening relaxation or downwelling intervals were sampled during what was a period of overall longer-term sea surface warming. Surface temperatures during upwelling were as low as 10.5°C in the upwelling center between Point Arguello and Point Conception, while during periods of nonupwelling winds, temperatures reached 14°–15°C in the same area. The upwelling center was also a source of higher-salinity water (33.7‰) relative to the offshore waters that were anomalously fresh (33.3‰). Upwelling was observed off Point Arguello, Point Conception, and the area between with a possible lag of up to a day at Point Conception relative to the other areas. The upwelling plume as observed by surface mapping and remote sensing tended to move in a variety of directions from SE to SW. Part of the movement was usually into the Santa Barbara Channel, but other parts of the plume occasionally moved southwestward. There was no obvious correlation of plume direction with the wind.     Validation of Regional CO2 Concentrations in the ECMWF Real-Time Analysis and Carbon-Tracker Reanalysis with Airborne Observations from ACT-A Field Campaign

    Through verifying against hundreds of hours of airborne in-situ measurements from the NASA-sponsored Atmospheric Carbon and Transport America (ACT-A) field campaign, this study systematically examines the regional uncertainties and biases of the carbon dioxide (CO2) concentrations from two of the state-of-the-art global analysis products, namely the real-time analysis from the European Center (EC) for Medium Range Forecasting and NOAAs near real-time Carbon Tracker (CT) reanalysis. It is found that both the EC and CT-NRT analyses agree reasonably well with the independent ACT-A flight-level CO2 measurements in the free troposphere but the uncertainties are considerably larger in the boundary layer during both the summer months of 2016 and the winter months of 2017. There are also strong variabilities in accuracy and bias between seasons, and across three different subregions in the United States (Mid-Atlantic, Midwest and South). Overall, the analysis uncertainties of the EC and CT-NRT analyses in terms of root-mean square deviations against airborne data are comparable to each other, both of which are between 1-2 ppm in the free troposphere but can be as large as 10 ppm near the surface, which are grossly consistent with the difference between the two analyses. The current study not only provides systematic uncertainty estimates for both analysis products over North America but also demonstrated that these two independent estimates can be used to approximate the overall regional CO2 analysis uncertainties. Both statistics are important in future studies in quantifying the uncertainties of regional carbon concentration and flux estimates, as well as in assessing the impact of regional transport through more refined regional modeling and analysis systems

    Liquefaction and Related Ground Failure from July 2019 Ridgecrest Earthquake Sequence

    The 2019 Ridgecrest earthquake sequence produced a 4 July M 6.5 foreshock and a 5 July M 7.1 mainshock, along with 23 events with magnitudes greater than 4.5 in the 24 hr period following the mainshock. The epicenters of the two principal events were located in the Indian Wells Valley, northwest of Searles Valley near the towns of Ridgecrest, Trona, and Argus. We describe observed liquefaction manifestations including sand boils, fissures, and lateral spreading features, as well as proximate non‐ground failure zones that resulted from the sequence. Expanding upon results initially presented in a report of the Geotechnical Extreme Events Reconnaissance Association, we synthesize results of field mapping, aerial imagery, and inferences of ground deformations from Synthetic Aperture Radar‐based damage proxy maps (DPMs). We document incidents of liquefaction, settlement, and lateral spreading in the Naval Air Weapons Station China Lake US military base and compare locations of these observations to pre‐ and postevent mapping of liquefaction hazards. We describe liquefaction and ground‐failure features in Trona and Argus, which produced lateral deformations and impacts on several single‐story masonry and wood frame buildings. Detailed maps showing zones with and without ground failure are provided for these towns, along with mapped ground deformations along transects. Finally, we describe incidents of massive liquefaction with related ground failures and proximate areas of similar geologic origin without ground failure in the Searles Lakebed. Observations in this region are consistent with surface change predicted by the DPM. In the same region, geospatial liquefaction hazard maps are effective at identifying broad percentages of land with liquefaction‐related damage. We anticipate that data presented in this article will be useful for future liquefaction susceptibility, triggering, and consequence studies being undertaken as part of the Next Generation Liquefaction project

    Clinically Relevant Behavioral Endpoints in a Recurrent Nitroglycerin Migraine Model in Rats

    BACKGROUND: This research sought to further validate the rat nitroglycerin (NTG) migraine model by comparing the effects of single versus recurrent NTG episodes on behavioral endpoints that mirror ICHD-3 diagnostic criteria for migraine, and to determine if the altered behavioral endpoints are reduced after administration of sumatriptan. METHODS: Separate cohorts of rats were administered NTG (10 mg/kg/2 ml) or saline (Experiment 1: single injection; Experiment 2: repeated injections; Experiment 3: repeated injections with sumatriptan [0.0, 0.3 and 1.0 mg/kg/ml] rescue. Behavioral endpoints were assessed 2 h after final NTG administration and included time in light/dark chambers for photophobia and activity, pain facial ratings, and cool (5 °C) and warm (46 °C) tail dip. RESULTS: The first two experiments demonstrated that repeated (n = 5) but not single NTG injections produced photophobia, decreased activity, and yielded less weight gain than saline injections. Experiment 3 showed that sumatriptan attenuated hypoactivity, reduced facial expressions of pain, and reversed weight alterations in a dose-dependent manner. CONCLUSIONS: These findings identify numerous clinical homologies of a recurrent NTG rat migraine model that may be useful for screening novel pharmacotherapies
