    Tato diplomová práce je zaměřena na politické systémy a jejich výuku v občanské výchově na základní škole. V teoretické části práce je čtenář seznámen s historickým vývojem myšlenky demokracie od antického Řecka a Říma, přes rozvoj parlamentarismu ve Velké Británii až po rozšiřování politických práv v Evropě, která tvoří základ demokratických politických institucí, jež jsou v práci podrobněji rozebrány. Takto vymezené politické instituce jsou v druhé polovině teoretické části dány do protikladu k nedemokratickým systémům. V této části práce je představen vývoj těchto režimů a jejich teoretické vymezení vůči demokracii. Jednotlivé nedemokratické systémy jsou klasifikovány a představeny na příkladech existujících nebo zhroucených států. Praktická část diplomové práce se věnuje následně teoretickému vymezení výuky politických systémů v Rámcovém vzdělávacím programu, které se uskutečňuje v předmětu Výchova k občanství a v průřezovém tématu Výchova demokratického občana. Teoreticky nabyté znalosti jsou v druhé půli praktické části převedeny do návrhu čtyř vyučovacích bloků, ve kterých se žáci postupně seznamují s demokratickými institucemi na úrovni spolu, obce a státu. In the theoretical part of the text, the historical development of the idea of democracy in ancient Greece and Rome, the development of parliamentarism in the UK and the expansion of political rights in Europe are announced to the reader. The importance of this development is considerable, as it forms the basis of democratic political institutions that are the core of this thesis. The rest of the theoretical part is dedicated to the undemocratic regimes. It deals with the development of these regimes and its theoretical definition against the democratical one. All of these undemocratic regimes are presented on examples of existing or collapsed states. The practical part of my thesis is dedicated to the tuition of political systems in Framework Educational Program which is presented in the subject Education on Citizenship and the cross-cutting theme Education of Democratic Citizen. The all theoretical knowledge is used in the draft of four tuition blocks. During these classes, students are gradually acquainted with democratic institutions on example of non-profit organisation, municipal and state level. Along with this is the quest for a good understanding of the value of water in its different uses. Using the Change in Net Income method, this paper presents an assessment of the value of water in irrigated paddy and hydroelectric power (HEP) generation in the Great Ruaha (GR) Catchment in Tanzania. The average values of water for irrigated paddy were estimated at 0.01and0.04perm3forabstractedandconsumedwaterrespectively.ForHEP,thevalueswererelativelyhigher( 0.01 and 0.04 per m3 for abstracted and consumed water respectively. For HEP, the values were relatively higher ( 0.06-0.21 per m3 for gross and consumed water respectively). Yet irrigated paddy also contributes much: it supports the livelihoods of about 30,000 agrarian families in Usangu with average annual gross income of about US $ 911.90 per annum per family and the GR paddy contributes about 14-24% to national paddy production. Understanding these benefits is key to fostering informed debate on water management and allocation, identifying the base for making ?agreeable? trade-offs, the potential for improvement, and creating linkages with water allocation options

    A Riemannian approach to randers geodesics

    In certain circumstances tools of Riemannian geometry are sufficient to address questions arising in the more general Finslerian context. We show that one such instance presents itself in the characterisation of geodesics in Randers spaces of constant flag curvature. To achieve a simple, Riemannian derivation of this special family of curves, we exploit the connection between Randers spaces and the Zermelo problem of time-optimal navigation in the presence of background fields. The characterisation of geodesics is then proven by generalising an intuitive argument developed recently for the solution of the quantum Zermelo problem

    Development of an Optimized LC-MS Method for the Detection of Specialized Pro-Resolving Mediators in Biological Samples

    The cardioprotective and anti-inflammatory effects of long chain omega-3 polyunsaturated fatty acids (n3 PUFA) are believed to be partly mediated by their oxygenated metabolites (oxylipins). In the last two decades interest in a novel group of autacoids termed specialized pro-resolving mediators (SPMs) increased. These are actively involved in the resolution of inflammation. SPMs are multiple hydroxylated fatty acids including resolvins, maresins, and protectins derived from the n3 PUFA eicosapentaenoic acid (EPA) and docosahexaenoic acid (DHA) as well as lipoxins derived from arachidonic acid (ARA). In the present paper, we developed an LC-MS/MS method for a comprehensive set of 18 SPMs derived from ARA, EPA, and DHA and integrated it into our targeted metabolomics platform. Quantification was based on external calibration utilizing five deuterated internal standards in combination with a second internal standard for quality assessment of sample preparation in each sample. The tandem mass spectrometric parameters were carefully optimized for sensitive and specific detection. The influence of source parameters of the used AB Sciex 6500 QTRAP instrument as well as electronic parameters and the selection of transitions are discussed. The method was validated/characterized based on the criteria listed in the European Medicines Agency (EMA) guideline on bioanalytical method validation and method performance is demonstrated regarding recovery of internal standards (between 78 ± 4% and 87 ± 3% from 500 μL of human serum) as well as extraction efficacy of SPMs in spiked plasma (intra-day accuracy within ±20 and ±15% at 0.1 and 0.3 nM in plasma, respectively). Based on the lower limit of quantification of 0.02–0.2 nM, corresponding to 0.18–2.7 pg on column, SPMs were generally not detectable/quantifiable in plasma and serum supporting that circulating levels of SPMs are very low, i.e., <0.1 nM in healthy subjects. Following septic shock or peritonitis, SPMs could be quantified in the samples of several patients. However, in these studies with a small number of patients no clear correlation with severity of inflammation could be observed
