    The Effect of Grass Carp Introduction on Aquatic Vegetation and Existing Fish Populations in Two Small Prairie Lakes

    Within two years after triploid grass carp introduction in 2.3 hectare Prior Lake in South Dakota, aquatic vegetation coverage and height in the water column were significantly lower (P\u3c0.005). During the same two year period, triploid grass carp did not significantly reduce aquatic vegetation coverage or height in the water column in 11.4 hectare East Lake Eureka, also in South Dakota. Stocking densities were 49 fish/hectare and 61 fish/hectare (229 mm mean total length) in Prior Lake and East Lake Eureka, respectively. The biomass of prey fish in East Lake Eureka in 1986 was quite high with 88.3, 85.3 and 17.3 kg/hectare for black bullhead (Ictalurus melas), yellow perch (Perea flavescens), and bluegill (Lepomis macrochirus), respectively. The biomass of northern pike (Esox lucius) was also high at 13.8 kg/hectare. A Proportional Stock Density (PSD) of 11 and Relative Weight (WK) of 117 indicated a relatively healthy bluegill population in East Lake Eureka. However, the PSD value for black bullheads was lower (8) while their condition was relatively good (K=l.58). The yellow perch population appeared to be over-populated in East Lake Eureka with a PSD of only 5 and a relatively low condition factor (K=l.14). The northern pike population in East Lake Eureka had a PSD value of 53, but a slightly low Wr of 94. Available prey/predator ratio indicated that there was an excess of prey fish for most size classes of predators. Because no significant vegetation reduction occurred in East Lake Eureka, no changes in existing fish populations could be attributed to grass carp introduction or aquatic vegetation reduction. Data gathered on existing fish populations in East Lake Eureka will serve as a pre-treatment data set to be compared to future conditions if aquatic vegetation is reduced. Fish sampling in Prior Lake was greatly reduced due to a winterkill which occurred in the late winter of 1986. Bluegill and largemouth bass (Micropterus salmoides) were the most abundant fish species found in Prior Lake in 1987. Bluegill appeared to be overpopulated as their PSD value was 1; a high Wr of 109 may have indicated improved conditions immediately following the winterkill. Results for the largemouth bass in Prior Lake were similar with a low PSD (7) and a high w~ (112). Winterkill (Prior Lake) and lack of aquatic vegetation control by grass carp (East Lake Eureka) prohibited making conclusions about the effect of grass carp introduction on the existing fish populations in the two lakes. Further research needs to be conducted in South Dakota to determine appropriate grass carp stocking rates for South Dakota waters, and to evaluate what effect grass carp introduction and aquatic vegetation reduction has on existing fish populations

    Conjugative transfer of ICESde3396 between three β-hemolytic streptococcal species

    Background: Integrative conjugative elements (ICEs) are mobile genetic elements (MGEs) that possess all genes necessary for excision, transfer and integration into recipient genome. They also carry accessory genes that impart new phenotypic features to recipient strains. ICEs therefore play an important role in genomic plasticity and population structure. We previously characterised ICESde 3396, the first ICE identified in the β-hemolytic Streptococcus dysgalactiae subsp equisimilis (SDSE) and demonstrated its transfer to single isolates of Streptococcus pyogenes (group A streptococcus, GAS) and Streptococcus agalactiae (group B streptococcus, GBS). While molecular studies found the ICE in multiple SDSE and GBS isolates, it was absent in all GAS isolates examined. Results: Here we demonstrate that ICESde 3396:km is transferable from SDSE to multiple SDSE, GAS and GBS isolates. However not all strains of these species were successful recipients under the same growth conditions. To address the role that host factors may have in conjugation we also undertook conjugation experiments in the presence of A549 epithelial cells and DMEM. While Horizontal Gene Transfer (HGT) occurred, conjugation efficiencies were no greater than when similar experiments were conducted in DMEM. Additionally transfer to GAS NS235 was successful in the presence of DMEM but not in Todd Hewitt Broth suggesting that nutritional factors may also influence HGT. The GAS and GBS transconjugants produced in this study are also able to act as donors of the ICE. Conclusion: We conclude that ICEs are major sources of interspecies HGT between β-hemolytic streptococci, and by introducing accessory genes imparting novel phenotypic characteristics, have the potential to alter the population structure of these species

    The Evolution of Protein Interaction Networks in Regulatory Proteins

    Interactions between proteins are essential for intracellular communication. They form complex networks which have become an important source for functional analysis of proteins. Combining phylogenies with network analysis, we investigate the evolutionary history of interaction networks from the bHLH, NR and bZIP transcription-factor families. The bHLH and NR networks show a hub-like structure with varying γ values. Mutation and gene duplication play an important role in adding and removing interactions. We conclude that in several of the protein families that we have studied, networks have primarily arisen by the development of heterodimerizing transcription factors, from an ancestral gene which interacts with any of the newly emerging proteins but also homodimerizes

    Reduction/oxidation-phosphorylation control of DNA binding in the bZIP dimerization network

    BACKGROUND: bZIPs are transcription factors that are found throughout the eukarya from fungi to flowering plants and mammals. They contain highly conserved basic region (BR) and leucine zipper (LZ) domains and often function as environmental sensors. Specifically, bZIPs frequently have a role in mediating the response to oxidative stress, a crucial environmental signal that needs to be transduced to the gene regulatory network. RESULTS: Based on sequence comparisons and experimental data on a number of important bZIP transcription factors, we predict which bZIPs are under redox control and which are regulated via protein phosphorylation. By integrating genomic, phylogenetic and functional data from the literature, we then propose a link between oxidative stress and the choice of interaction partners for the bZIP proteins. CONCLUSION: This integration permits the bZIP dimerization network to be interpreted in functional terms, especially in the context of the role of bZIP proteins in the response to environmental stress. This analysis demonstrates the importance of abiotic factors in shaping regulatory networks

    Memory-Like Responses of Brain Microglia Are Controlled by Developmental State and Pathogen Dose

    Microglia, the innate immune cells of the central nervous system, feature adaptive immune memory with implications for brain homeostasis and pathologies. However, factors involved in the emergence and regulation of these opposing responses in microglia have not been fully addressed. Recently, we showed that microglia from the newborn brain display features of trained immunity and immune tolerance after repeated contact with pathogens in a dose-dependent manner. Here, we evaluate the impact of developmental stage on adaptive immune responses of brain microglia after repeated challenge with ultra-low (1 fg/ml) and high (100 ng/ml) doses of the endotoxin LPS in vitro. We find that priming of naïve microglia derived from newborn but not mature and aged murine brain with ultra-low LPS significantly increased levels of pro-inflammatory mediators TNF-α, IL-6, IL-1β, MMP-9, and iNOS as well as neurotrophic factors indicating induction of trained immunity (p \u3c 0.05). In contrast, stimulation with high doses of LPS led to a robust downregulation of pro-inflammatory cytokines and iNOS independent of the developmental state, indicating induced immune tolerance. Furthermore, high-dose priming with LPS upregulated anti-inflammatory mediators IL-10, Arg-1, TGF- β, MSR1, and IL-4 in newborn microglia (p \u3c 0.05). Our data indicate pronounced plasticity of the immune response of neonate microglia compared with microglia derived from mature and aged mouse brain. Induced trained immunity after priming with ultra-low LPS doses may be responsible for enhanced neuro-inflammatory susceptibility of immature brain. In contrast, the immunosuppressed phenotype following high-dose LPS priming might be prone to attenuate excessive damage after recurrent systemic inflammation

    Memory-Like Inflammatory Responses of Microglia to Rising Doses of LPS: Key Role of PI3Kγ

    Trained immunity and immune tolerance have been identified as long-term response patterns of the innate immune system. The causes of these opposing reactions remain elusive. Here, we report about differential inflammatory responses of microglial cells derived from neonatal mouse brain to increasing doses of the endotoxin LPS. Prolonged priming with ultra-low LPS doses provokes trained immunity, i.e., increased production of pro-inflammatory mediators in comparison to the unprimed control. In contrast, priming with high doses of LPS induces immune tolerance, implying decreased production of inflammatory mediators and pronounced release of anti-inflammatory cytokines. Investigation of the signaling processes and cell functions involved in these memory-like immune responses reveals the essential role of phosphoinositide 3-kinase γ (PI3Kγ), one of the phosphoinositide 3-kinase species highly expressed in innate immune cells. Together, our data suggest profound influence of preceding contacts with pathogens on the immune response of microglia.     This is a transcript of statements by Attorney, Fred W. Garmone (Counsel for Sam Sheppard) regarding a short in-chambers conference pre-trial with the Honorable Judge Blythin. Mr. Garmone stated to Judge Blythin, because of the fact that his son (Ed Blythin) at the time of trial was a member of the Homicide Unit, and was actively engaged in the investigation of this death of Marilyn Sheppard, that it was his feeling that he should withdraw or disqualify himself as the Judge to hear the case. Along with Mr. Garmone\u27s statement is a brief question/answer exchange between members of the Prosecution and the Sheppard Counsel present; conversation exchange inquires of pre-trial defense attorney dissension or discord and whether there was a formal affidavit of prejudice against Judge Blythin for his personal connection to the criminal investigation. These inquiries prompted negative responses from Mr. Garmone
