    Identifying the science and technology dimensions of emerging public policy issues through horizon scanning

    Public policy requires public support, which in turn implies a need to enable the public not just to understand policy but also to be engaged in its development. Where complex science and technology issues are involved in policy making, this takes time, so it is important to identify emerging issues of this type and prepare engagement plans. In our horizon scanning exercise, we used a modified Delphi technique [1]. A wide group of people with interests in the science and policy interface (drawn from policy makers, policy adviser, practitioners, the private sector and academics) elicited a long list of emergent policy issues in which science and technology would feature strongly and which would also necessitate public engagement as policies are developed.     OBJECTIVE: To investigate whether herpes simplex virus type 1 (HSV-1) infection was associated with rates of cognitive decline or whole brain atrophy among individuals from the Dominantly Inherited Alzheimer Network (DIAN). METHODS: Among two subsets of the DIAN cohort (age range 19.6-66.6 years; median follow-up 3.0 years) we examined (i) rate of cognitive decline (N = 164) using change in mini-mental state examination (MMSE) score, (ii) rate of whole brain atrophy (N = 149), derived from serial MR imaging, calculated using the boundary shift integral (BSI) method. HSV-1 antibodies were assayed in baseline sera collected from 2009-2015. Linear mixed-effects models were used to compare outcomes by HSV-1 seropositivity and high HSV-1 IgG titres/IgM status. RESULTS: There was no association between baseline HSV-1 seropositivity and rates of cognitive decline or whole brain atrophy. Having high HSV-1 IgG titres/IgM was associated with a slightly greater decline in MMSE points per year (difference in slope - 0.365, 95% CI: -0.958 to -0.072), but not with rate of whole brain atrophy. Symptomatic mutation carriers declined fastest on both MMSE and BSI measures, however, this was not influenced by HSV-1. Among asymptomatic mutation carriers, rates of decline on MMSE and BSI were slightly greater among those who were HSV-1 seronegative. Among mutation-negative individuals, no differences were seen by HSV-1. Stratifying by APOE4 status yielded inconsistent results. INTERPRETATION: We found no evidence for a major role of HSV-1, measured by serum antibodies, in cognitive decline or whole brain atrophy among individuals at high risk of early-onset AD

    We investigated the pattern of non-starch polysaccharide (NSP) digestion along the gut of pigs fed two different wheats, which were offered with or without xylanase supplementation. The two wheats used were pre-characterised before the experiment on the basis of low and normal feed intake of young pigs. Wheat type significantly influenced feed intake and growth rate in the first 7 days, however, by day 14 the only significant effect of wheat type was on growth rate. Xylanase supplementation increased the growth performance of pigs fed the poor quality wheat to a level similar to those fed the normal wheat. It also increased the daily gain of pigs fed the normal wheat. Wheat type had no significant effect on the digestibility of dry matter (DM), energy, free sugars or the different fractions of NSP in the duodenum, ileum or in the faeces. The duodenal gross energy digestibility values for the low and high performance diets were −27.4 and −47.5%, respectively, and xylanase supplementation significantly increased the digestibility of energy back to positive levels. Dry matter digestibility values followed a similar pattern. In the duodenum, xylanase increased (P < 0.05) the digestibility values of both soluble and insoluble NSP, whereas in the ileum, xylanase had a significant effect only on the digestibility of the soluble NSP fraction. Xylanase did not affect free sugar digestibility. The reduction in soluble NSP level coincided with a marked reduction in the amount of fucose, a prominent component of mucosal polysaccharides. This suggests that soluble NSP substantially increase endogenous losses. The absence of differences in the digestibility of the measured NSP between the two wheat samples suggests that the structures of the NSP, rather than just their amount and solubility, are important for the anti-nutritional properties of NSP in pig diets.     Phytase Supplementation of Wheat-Based Broiler Diets Reduces Dependence on Meat-and-Bone Meal

    It is permissible to include meat-and-bone meal (MBM) in broiler diets in many countries outside the European Union, including Australia, where MBM is usually advantaged by being a relatively inexpensive source of phosphorus and protein. However, some producers may prefer to reduce their dependence on MBM as this feed ingredient has several disadvantages and may be a barrier to exports of chicken-meat. Therefore, the purpose of this experiment was to investigate the extent to which phytase supplementation can reduce MBM inclusion levels in wheat-based broiler diets. Standard and modified starter, grower and finisher diets were formulated so that modified diets contained lower MBM inclusions and reduced nutrient specifications. The standard and modified diets, the latter supplemented with 0, 500 or 750FTU/kg phytase (Phyzyme&reg; XP), were offered to a total of 840 broilers in 24 floor pens from 1-42 days of age and the effects of dietary treatments on growth performance were determined. For the grower phase (15-28 days), five broilers from each floor pen were transferred to cages to permit total excreta collection to determine apparent metabolisable energy (AME) and nitrogen (N) retention. Acid insoluble ash was included in the caged birds&rsquo; diets and ileal digesta collected at 28 days to determine the effect of dietary treatments on apparent ileal digestibility (AID) of amino acids. Lowering nutrient specifications reduced growth performance and increased mortality rates over 42 days but this decline in performance was overcome by the addition of 750FTU/kg phytase to modified diets. Feed ingredient costs per unit of live weight gain were also reduced by phytase addition. Reducing dietary levels of MBM increased amino acid digestibilities in the modified grower diet. Moreover, phytase supplementation of modified diets linearly increased AID coefficients of alanine, aspartic acid, glycine, isoleucine, leucine, serine, threonine and tyrosine to a significant extent. Also, phytase (750FTU/kg) supplementation of the modified diet significantly increased AME by 0.27MJ/kg on a dry matter (DM) basis. Overall consumption of MBM was reduced from 233g to 14g per bird from 1-42 days of age following dietary modifications. It is noteworthy that reduced inclusions of MBM, coupled with phytase supplementation, enhanced ileal digestibility of total amino acids by 5.3% (0.834 versus 0.792). This study demonstrates that it is feasible to reduce MBM contents and nutrient specifications in wheat-based broiler diets in association with phytase supplementation without the addition of inorganic phosphorus sources

    Effect of in ovo creatine monohydrate on hatchability, post-hatch performance, breast muscle yield and fiber size in chicks from young breeder flocks

    ABSTRACT: Younger broiler breeder flocks produce smaller eggs containing smaller yolks, with potentially lower energy reserves for the developing chick. Creatine is a naturally occurring energy source and is abundant in metabolically active tissues; providing this to chicks in ovo should provide additional energy to improve hatchability and post-hatch growth. Thus, post-hatch performance of male and female chicks hatched from younger breeder flocks supplemented with creatine monohydrate (CrM) in ovo was investigated. Four hundred eggs from Ross 308 breeder hens aged 27 to 29 wk were collected and at d 14 assigned to a treatment group and received 1) no injection, 2) 0.75% saline injection, or 3) 8.16 mg creatine monohydrate in 0.75% saline. At hatch 72 birds (24/treatment) were euthanized and BW, breast muscle, heart and liver weight were obtained, and breast muscle tissue was placed in 10% buffered formalin. Birds were then placed in raised metal pens (24 pens; 10–11 birds/pen; 8 replicates/treatment) and grown to d 42 with BW and pen feed intake measured once a week. At d 42, ninty-six birds were euthanized (2 male and 2 female/pen) and the process occurred as at hatch. Body composition was obtained for 48 birds (2/pen; 1 male,1 female) with a dual energy X-ray absorptiometry (DXA) scanner. Breast muscle tissue was processed for histological analysis and breast muscle fiber parameters were analyzed by ImageJ. While not statistically significant, the CrM treatment group saw an improved hatch rate (CrM: 93.5%, Saline: 88.6%, Control: 88.8%) and reduced early post hatch mortality. Chicks given in ovo CrM had significantly increased creatine concentrations in both liver and heart tissue at hatch compared to those in the saline and control groups. BW, BW gain, and final body composition parameters were not statistically different between treatments and in ovo CrM did not affect breast muscle fiber number or area. The creatine injection likely improved the energy status of the growing embryo resulting in the improved hatch rate but leaving little reserves for post-hatch growth

    Guanidinoacetic acid as a partial replacement to arginine with or without betaine in broilers offered moderately low crude protein diets

    Guanidinoacetic acid (GAA) is the direct precursor of creatine and can spare arginine (Arg) for creatine synthesis in low crude protein (CP) broiler diets. This study aimed to determine the extent GAA could spare Arg in broilers offered low CP diets and if supplemental betaine provides additional benefits. Seven hundred twenty-day-old Ross 308 male broilers were assigned into 9 dietary treatments with 8 replicates of 10 birds each. The treatments were; normal CP diet, a low CP (−15 g/kg) diet deficient in Arg, a low CP diet sufficient in Arg, and low CP diets with GAA, where 0.1% added L-Arg was spared by GAA at 50, 100, and 150% with and without 0.1% betaine. The treatments were offered during grower (d 10–24) and finisher (d 25–42) phases. The birds offered a low CP Arg deficient diet had 7.8% lower weight gain, 10 points higher FCR, 8.5% lower breast meat yield, 27.2% lower breast meat creatine concentration and 30.4% more abdominal fat pad compared to those offered a normal CP diet. When Arg was added back to the Arg deficient diet, growth performance, breast meat yield and creatine concentration loss were restored. When GAA spared Arg at 150%, feed intake, weight gain, FCR, breast and abdominal fat yields, breast meat moisture, drip loss, and breast meat creatine concentration became comparable to Arg sufficient low CP and normal CP treatments. When GAA spared Arg at 100 and 50%, FCR was 3 and 5 points lower than the normal CP treatment. Breast meat creatine concentration was positively correlated to feed efficiency (r = 0.70, P P P
