    Deciphering Strawberry Ripening by Tissue Specific Gene Regulatory Networks

    During ripening, fruits undergo a number of metabolic and physiological changes leading to softening and improvement of characters such as flavor and palatability. Insights into transcriptome changes during strawberry fruit ripening have been reported, but always using either complete fruits in the analysis or separating achenes and the fleshy part (receptacle). However, the receptacle is composed of heterogeneous cell types, each of them with different characteristics and functions. Hence, transcriptomic studies performed so far may have lost important regulatory elements which expression is low but important in a specific cell-type specific. In our study, we use Laser Capture Microdissection (LCM) technique for the isolation of cells from specific tissue types such as the epidermis, vascular bundles, cortex, and pith. Transcriptome profiling of these tissue types was performed by RNAseq. A gene co-expression analysis was performed by Weighted Correlation Network Analysis (WGCNA). Ontology analysis of each module showed wax biosynthesis as the main biological pathway enriched at the red epidermis specific module. In order to elucidate the putative regulatory elements that control the synthesis of waxes in this tissue, a Gene Regulatory Network (GRN) was generated using GENIST (de Luis Balaguer, 2017). As a result, we have identified a set of transcription factors that might regulate the expression of eceriferum genes and a fatty acid elongase necessary for wax biosynthesis in ripe epidermis. Ultimately, our results open the possibility of implementing novel targeted breeding approaches. Moreover, this work shows that LCM followed by RNAseq is a powerful tool that can be used to clarify the regulatory scenario of tissue-specific biological processes during strawberry ripening.Universidad de Málaga.     Controllable production of Janus ligaments by AC fields in a flow-focusing junction

    We report the production of bicomponent Janus filaments of miscible aqueous fluids in a micro- fluidic electro-flow focusing device under the action of an AC electric field. The production of liquid filaments can lead to the generation of microfibers by adding a subsequent process of polymerization. Janus microfi- bers are of paramount importance in biomedical appli- cations such as tissue production on crimped scaffolds. We show that the filament length is a function of the frequency signal, voltage amplitude and of the visco- sity and conductivity of the dispersed phase. In par- ticular, Janus filaments with diameters ∼ 10μm and longer than 1 mm are produced by AC voltages with frequencies below 150 kHz and a viscosity of the dis- persed phase ∼ 10 cP

    Droplet group production in an AC electro-flow-focusing microdevice

    We report the production of droplet groups with a controlled number of drops in a microfluidic electro-flow focusing device under the action of an AC electric field. This regime appears for moderate voltages (500-700 V peak-to-peak) and signal frequencies between 25 and 100 Hz, much smaller than the droplet production rate ( ≈500 Hz). For this experimental conditions the production frequency of a droplet package is twice the signal frequency.     An Antimethanogenic Nutritional Intervention in Early Life of Ruminants Modifies Ruminal Colonization by Archaea

    The aim of this work was to study whether feeding a methanogen inhibitor from birth of goat kids and their does has an impact on the archaeal population colonizing the rumen and to what extent the impact persists later in life. Sixteen goats giving birth to two kids were used. Eight does were treated (D+) with bromochloromethane after giving birth and over 2 months. The other 8 goats were not treated (D−). One kid per doe in both groups was treated with bromochloromethane (k+) for 3 months while the other was untreated (k−), resulting in four experimental groups: D+/k+, D+/k−, D−/k+, and D−/k−. Rumen samples were collected from kids at weaning and 1 and 4 months after (3 and 6 months after birth) and from does at the end of the treating period (2 months). Pyrosequencing analyses showed a modified archaeal community composition colonizing the rumen of kids, although such effect did not persist entirely 4 months after; however, some less abundant groups remained different in treated and control animals. The different response on the archaeal community composition observed between offspring and adult goats suggests that the competition occurring in the developing rumen to occupy different niches offer potential for intervention

    AOLI: Near-diffraction limited imaging in the visible on large ground -based telescopes

    The combination of Lucky Imaging with a low order adaptive optics system was demonstrated very successfully on the Palomar 5m telescope nearly 10 years ago. It is still the only system to give such high-resolution images in the visible or near infrared on ground-based telescope of faint astronomical targets. The development of AOLI for deployment initially on the WHT 4.2 m telescope in La Palma, Canary Islands, will be described in this paper. In particular, we will look at the design and status of our low order curvature wavefront sensor which has been somewhat simplified to make it more efficient, ensuring coverage over much of the sky with natural guide stars as reference object.     Mechanisms involved in the adaptation of Escherichia coli O157:H7 to the host intestinal microenvironment

    Host adaptation of pathogens may increase intra- and interspecies transmission. We showed previously that the passage of a clinically isolated enterohemorrhagic Escherichia coli (EHEC) O157 strain (125/99) through the gastrointestinal tract of mice increases its pathogenicity in the same host. In this work, we aimed to elucidate the underlying mechanism(s) involved in the patho-adaptation of the stool-recovered (125RR) strain. We assessed the global transcription profile by microarray and found almost 100 differentially expressed genes in 125RR strain compared with 125/99 strain. We detected an overexpression of Type Three Secretion System (TTSS) proteins at the mRNA and protein levels and demonstrated increased adhesion to epithelial cell lines for the 125RR strain. Additional key attributes of the 125RR strain were: increased motility on semisolid agar, which correlated with an increased fliC mRNA level; reduced Stx2 production at the mRNA and protein levels; increased survival at pH 2.5, as determined by acid resistance assays. We tested whether the overexpression of the LEE-encoded regulator (ler) in trans in the 125/99 strain could recreate the increased pathogenicity observed in the 125RR strain. As anticipated ler overexpression led to increased expression of TTSS proteins and bacterial adhesion to epithelial cells in vitro but also increased mortality and intestinal colonization in vivo. We conclude that this host-adaptation process required changes in several mechanisms that improved EHEC O157 fitness in the new host. The research highlights some of the bacterial mechanisms required for horizontal transmission of these zoonotic pathogens between their animal and human populations.Fil: Fernández Brando, Romina Jimena.     Characterisation, prevalence and severity of skin lesions caused by ophidiomycosis in a population of wild snakes

    Ophidiomycosis is an emerging infectious disease affecting wild snakes in the Northern Hemisphere. Recently confirmed in Great Britain, the prevalence, severity and significance of ophidiomycosis has yet to be characterised in free-living snakes at a population level in Europe. Therefore, a population of barred grass snakes (Natrix helvetica) in eastern England was monitored for three seasons (May 2019 to October 2021), to investigate the prevalence (25.5%; 191/750 snakes) and severity of skin lesions and their aetiology. The most frequently observed skin lesion characteristics were changes in scale colour, crusting, and scale margin erosion. The majority of such lesions (96.9%; 185/191 snakes) was observed on the ventral surface along the length of the body. The severity of skin lesions was considered mild in more than half of the cases (53.1%; 98/191 snakes). Predominantly, skin lesions were observed in adult snakes (72.8%; 139/191 snakes). Combined histological examinations and qPCR tests of skin lesions from N. helvetica sloughs and/or carcasses confirmed a diagnosis of ophidiomycosis. Further targeted surveillance, supported by molecular and histological examinations to confirm skin lesion aetiology, is required to determine the extent to which our findings reflect the occurrence of ophidiomycosis in populations within wider landscapes

    Sexual selection on male vocal fundamental frequency in humans and other anthropoids

J.R.W. was supported by a National Science Foundation predoctoral fellowship.In many primates, including humans, the vocalizations of males and females differ dramatically, with male vocalizations and vocal anatomy often seeming to exaggerate apparent body size. These traits may be favoured by sexual selection because low-frequency male vocalizations intimidate rivals and/or attract females, but this hypothesis has not been systematically tested across primates, nor is it clear why competitors and potential mates should attend to vocalization frequencies. Here we show across anthropoids that sexual dimorphism in fundamental frequency (F0) increased during evolutionary transitions towards polygyny, and decreased during transitions towards monogamy. Surprisingly, humans exhibit greater F0 sexual dimorphism than any other ape. We also show that low-F0 vocalizations predict perceptions of men’s dominance and attractiveness, and predict hormone profiles (low cortisol and high testosterone) related to immune function. These results suggest that low male F0 signals condition to competitors and mates, and evolved in male anthropoids in response to the intensity of mating competition.PostprintPeer reviewe
