Factors That Inhibit and Support the Work of School-Based Mental Health Providers
This research brief comes from the MERC Supporting Mental Health in Schools study. Launched in 2021 in the wake of the COVID-19 pandemic, this study explores research-based strategies for supporting mental health in PK-12 school settings, as well as the emotional and professional capacity of PK-12 faculty and staff in the MERC region to support the social and emotional wellness of their students. This brief shares takeaways from a systematic literature review that highlight the factors that inhibit and support the work of school-based mental health providers, including school counselors, school social workers, and school psychologists. It outlines the roles and responsibilities of these professionals, the challenges they face, and the factors that help facilitate their work. It concludes with a series of recommendations for practice, policy, and research
Metabolic Regulation of Neuronal Plasticity by the Energy Sensor AMPK
Long Term Potentiation (LTP) is a leading candidate mechanism for learning and memory and is also thought to play a role in the progression of seizures to intractable epilepsy. Maintenance of LTP requires RNA transcription, protein translation and signaling through the mammalian Target of Rapamycin (mTOR) pathway. In peripheral tissue, the energy sensor AMP-activated Protein Kinase (AMPK) negatively regulates the mTOR cascade upon glycolytic inhibition and cellular energy stress. We recently demonstrated that the glycolytic inhibitor 2-deoxy-D-glucose (2DG) alters plasticity to retard epileptogenesis in the kindling model of epilepsy. Reduced kindling progression was associated with increased recruitment of the nuclear metabolic sensor CtBP to NRSF at the BDNF promoter. Given that energy metabolism controls mTOR through AMPK in peripheral tissue and the role of mTOR in LTP in neurons, we asked whether energy metabolism and AMPK control LTP. Using a combination of biochemical approaches and field-recordings in mouse hippocampal slices, we show that the master regulator of energy homeostasis, AMPK couples energy metabolism to LTP expression. Administration of the glycolytic inhibitor 2-deoxy-D-glucose (2DG) or the mitochondrial toxin and anti-Type II Diabetes drug, metformin, or AMP mimetic AICAR results in activation of AMPK, repression of the mTOR pathway and prevents maintenance of Late-Phase LTP (L-LTP). Inhibition of AMPK by either compound-C or the ATP mimetic ara-A rescues the suppression of L-LTP by energy stress. We also show that enhanced LTP via AMPK inhibition requires mTOR signaling. These results directly link energy metabolism to plasticity in the mammalian brain and demonstrate that AMPK is a modulator of LTP. Our work opens up the possibility of using modulators of energy metabolism to control neuronal plasticity in diseases and conditions of aberrant plasticity such as epilepsy
The Carnegie Supernova Project: Analysis of the First Sample of Low-Redshift Type-Ia Supernovae
We present the analysis of the first set of low-redshift Type Ia supernovae
(SNe Ia) by the Carnegie Supernova Project. Well-sampled, high-precision
optical (ugriBV) and near-infrared (NIR; YJHKs) light curves obtained in a
well-understood photometric system are used to provide light-curve parameters,
and ugriBVYJH template light curves. The intrinsic colors at maximum light are
calibrated to compute optical--NIR color excesses for the full sample, thus
allowing the properties of the reddening law in the host galaxies to be
studied. A low value of Rv~1.7, is derived when using the entire sample of SNe.
However, when the two highly reddened SNe in the sample are excluded, a value
Galactic standard of Rv~3.2 is obtained. The colors of these two events are
well matched by a reddening model due to circumstellar dust. The peak
luminosities are calibrated using a two-parameter linear fit to the decline
rates and the colors, or alternatively, the color excesses. In both cases,
dispersions in absolute magnitude of 0.12--0.16 mag are obtained, depending on
the filter-color combination. In contrast to the results obtained from color
excesses, these fits give Rv~1--2, even when the two highly reddened SNe are
excluded. This discrepancy suggests that, beyond the "normal" interstellar
reddening produced in the host galaxies, there is an intrinsic dispersion in
the colors of SNe Ia which is correlated with luminosity but independent of the
decline rate. Finally, a Hubble diagram is produced by combining the results of
the fits for each filter. The resulting scatter of 0.12 mag appears to be
limited by peculiar velocities as evidenced by the strong correlation between
the distance-modulus residuals among the different filters. The implication is

Fibrinogen regulates the cytotoxicity of mycobacterial trehalose dimycolate, but is not required for cell recruitment, cytokine response, or control of mycobacterial infection
During inflammatory responses and wound healing, the conversion of soluble fibrinogen to fibrin, an insoluble extracellular matrix, long has been assumed to create a scaffold for the migration of leukocytes and fibroblasts. Previous studies concluded that fibrinogen is a necessary cofactor for mycobacterial trehalose 6,6-dimycolate-induced responses, because trehalose dimycolate-coated beads, to which fibrinogen was ad-sorbed, were more inflammatory than those to which other plasma proteins were adsorbed. Herein, we investigate roles for fibrin(ogen) in an in vivo model of mycobacterial granuloma formation and in infection with Mycobacterium tuberculosis, the causative agent of tuberculosis. In wild-type mice, the subcutaneous injection of trehalose dimycolate-coated polystyrene microspheres, suspended within Matrigel, elicited a pyogranulomatous response during the course of 12 days. In fibrinogen-deficient mice, neutrophils were recruited but a more suppurative lesion developed, with the marked degradation and disintegration of the matrix. Compared to that in wild-type mice, the early formation of granulation tissue in fibrinogen-deficient mice was edematous, hypocellular, and disorganized. Normative Alethic Pluralism
Some philosophers have argued that truth is a norm of judgement and have provided a variety of formulations of this general thesis. In this paper, I shall side with these philosophers and assume that truth is a norm of judgement. What I am primarily interested in here are two core questions concerning the judgement-truth norm: (i) what are the normative relationships between truth and judgement? And (ii) do these relationships vary or are they constant? I argue for a pluralist picture—what I call Normative Alethic Pluralism (NAP)—according to which (i) there is more than one correct judgement-truth norm and (ii) the normative relationships between truth and judgement vary in relation to the subject matter of the judgement. By means of a comparative analysis of disagreement in three areas of the evaluative domain—refined aesthetics, basic taste and morality—I show that there is an important variability in the normative significance of disagreement—I call this the variability conjecture. By presenting a variation of Lynch’s scope problem for alethic monism, I argue that a monistic approach to the normative function of truth is unable to vindicate the conjecture. I then argue that normative alethic pluralism provides us with a promising model to account for it
Omega-3 fatty acids attenuate cardiovascular effects of short-term exposure to ambient air pollution
Exposure to air pollution is associated with elevated cardiovascular risk. Evidence shows that omega-3 polyunsaturated fatty acids (omega-3 PUFA) may attenuate the adverse cardiovascular effects of exposure to fine particulate matter (PM2.5). However, it is unclear whether habitual dietary intake of omega-3 PUFA protects against the cardiovascular effects of short-term exposure to low-level ambient air pollution in healthy participants. In the present study, sixty-two adults with low or high dietary omega-3 PUFA intake were enrolled. Blood lipids, markers of vascular inflammation, coagulation and fibrinolysis, and heart rate variability (HRV) and repolarization were repeatedly assessed in 5 sessions separated by at least 7 days. This study was carried out in the Research Triangle area of North Carolina, USA between October 2016 and September 2019. Daily PM2.5 and maximum 8-h ozone (O3) concentrations were obtained from nearby air quality monitoring stations. Linear mixed-effects models were used to assess the associations between air pollutant concentrations and cardiovascular responses stratified by the omega-3 intake levels
Fusion of the molecular adjuvant C3d to cleavage-independent native-like HIV-1 Env trimers improves the elicited antibody response
An effective HIV vaccine likely requires the elicitation of neutralizing antibodies (NAbs) against multiple HIV-1 clades. The recently developed cleavage-independent native flexibly linked (NFL) envelope (Env) trimers exhibit well-ordered conformation and elicit autologous tier 2 NAbs in multiple animal models. Here, we investigated whether the fusion of molecular adjuvant C3d to the Env trimers can improve B- cell germinal center (GC) formation and antibody responses. To generate Env-C3d trimers, we performed a glycine-serine- based (G4S) flexible peptide linker screening and identified a linker range that allowed native folding. A 30–60- amino- acid- long linker facilitates Env-to-C3d association and achieves the secretion of well-ordered trimers and the structural integrity and functional integrity of Env and C3d. The fusion of C3d did not dramatically affect the antigenicity of the Env trimers and enhanced the ability of the Env trimers to engage and activate B cells in vitro. In mice, the fusion of C3d enhanced germinal center formation, the magnitude of Env-specific binding antibodies, and the avidity of the antibodies in the presence of an adjuvant. The Sigma Adjuvant System (SAS) did not affect the trimer integrity in vitro but contributed to altered immunogenicity in vivo, resulting in increased tier 1 neutralization, likely by increased exposure of variable region 3 (V3). Taken together, the results indicate that the fusion of the molecular adjuvant, C3d, to the Env trimers improves antibody responses and could be useful for Env-based vaccines against HIV
The Carnegie Supernova Project: First Photometry Data Release of Low-Redshift Type Ia Supernovae
The Carnegie Supernova Project (CSP) is a five-year survey being carried out
at the Las Campanas Observatory to obtain high-quality light curves of ~100
low-redshift Type Ia supernovae in a well-defined photometric system. Here we
present the first release of photometric data that contains the optical light
curves of 35 Type Ia supernovae, and near-infrared light curves for a subset of
25 events. The data comprise 5559 optical (ugriBV) and 1043 near-infrared
(YJHKs) data points in the natural system of the Swope telescope. Twenty-eight
supernovae have pre-maximum data, and for 15 of these, the observations begin
at least 5 days before B maximum. This is one of the most accurate datasets of
low-redshift Type Ia supernovae published to date. When completed, the CSP
dataset will constitute a fundamental reference for precise determinations of
cosmological parameters, and serve as a rich resource for comparison with
