    Transformation of the Cyanobacterium Anacystis Nidulans 6301 with the Escherichia Coli Plasmid pBR322

    Anacystis nidulans 6301 has been transformed in the light to ampicillin resistance with the plasmid pBR322. Permeaplasts prepared by 2-hr treatment of cells with lysozyme and EDTA are transformed with a 50-fold higher efficiency than that observed for cells. beta-Lactamase is present in A. nidulans transformed either with pBR322 or the plasmid pCH1 as evidenced by hydrolysis of the beta-lactam ring of Nitrocefin in extracts of transformants. beta-Lactamase also can be immunoprecipitated from extracts of [35S]methionine-labeled pBR322 transformants and coprecipitates with ribulose-bisphosphate carboxylase. Expression of the carboxylase is apparently amplified in pBR322 transformants as is that for several soluble proteins in pCH1 transformants. Chromosomal DNA per cell is increased about 6-fold after transformation of A. nidulans 6301 with either pBR322 or pCH1. A 4.3-kilobase-pair plasmid can be isolated from pBR322 transformants in addition to the endogenous plasmids pUH24 and pUH25

    High fidelity imaging and high performance computing in nonlinear EIT

    We show that nonlinear EIT provides images with well defined characteristics when smoothness of the image is used as a constraint in the reconstruction process. We use the gradient of the logarithm of resistivity as an effective measure of image smoothness, which has the advantage that resistivity and conductivity are treated with equal weight. We suggest that a measure of the fidelity of the image to the object requires the explicit definition and application of such a constraint. The algorithm is applied to the simulation of intra-ventricular haemorrhaging (IVH) in a simple head model. The results indicate that a 5% increase in the blood content of the ventricles would be easily detectable with the noise performance of contemporary instrumentation. The possible implementation of the algorithm in real time via high performance computing is discussed

    Policy analysis and policy analytics

    Working from a description of what policy analysis entails, we review the emergence of the recent field of analytics and how it may impact public policy making. In particular, we seek to expose current applications of, and future possibilities for, new analytic methods that can be used to support public policy problem-solving and decision processes, which we term policy analytics. We then review key contributions to this special volume, which seek to support policy making or delivery in the areas of energy planning, urban transportation planning, medical emergency planning, healthcare, social services, national security, defence, government finance allocation, understanding public opinion, and fire and police services. An identified challenge, which is specific to policy analytics, is to recognize that public sector applications must balance the need for robust and convincing analysis with the need for satisfying legitimate public expectations about transparency and opportunities for participation. This opens up a range of forms of analysis relevant to public policy distinct from those most common in business, including those that can support democratization and mediation of value conflicts within policy processes. We conclude by identifying some potential research and development issues for the emerging field of policy analytics

    Evaluating Participatory Modeling: Developing a Framework for Cross-case Analysis

    Participatory modeling is increasingly recognised as an effective way to assist collective decision-making processes in the domain of natural resource management. This paper introduces a framework for evaluating projects that have adopted a participatory modeling approach. This framework – known as the ‘Protocol of Canberra’ – was developed through a collaboration between French and Australian researchers engaged in participatory modeling and evaluation research. The framework seeks to assess the extent to which different participatory modeling practices reinforce or divert from the theoretical assumptions they are built upon. The paper discusses the application of the framework in three case-studies, two from Australia and one from the Pacific island of the Republic of Kiribati. The paper concludes with some comments for future use of the framework in a range of participatory modeling contexts, including fostering consideration of why and how different methodological approaches are used to achieve project aims and to build a collective vision amongst diverse stakeholders.participation, modeling, evaluation, complex systems science

    Molecular Strategies for Gene Containment in Transgenic Crops

    The potential of genetically modified (GM) crops to transfer foreign genes through pollen to related plant species has been cited as an environmental concern. Until more is known concerning the environmental impact of novel genes on indigenous crops and weeds, practical and regulatory considerations will likely require the adoption of gene-containment approaches for future generations of GM crops. Most molecular approaches with potential for controlling gene flow among crops and weeds have thus far focused on maternal inheritance, male sterility, and seed sterility. Several other containment strategies may also prove useful in restricting gene flow, including apomixis (vegetative propagation and asexual seed formation), cleistogamy (self-fertilization without opening of the flower), genome incompatibility, chemical induction/deletion of transgenes, fruit-specific excision of transgenes, and transgenic mitigation (transgenes that compromise fitness in the hybrid). As yet, however, no strategy has proved broadly applicable to all crop species, and a combination of approaches may prove most effective for engineering the next generation of GM crops

    Pioneer 10 Doppler data analysis: disentangling periodic and secular anomalies

    This paper reports the results of an analysis of the Doppler tracking data of Pioneer probes which did show an anomalous behaviour. A software has been developed for the sake of performing a data analysis as independent as possible from that of J. Anderson et al. \citep{anderson}, using the same data set. A first output of this new analysis is a confirmation of the existence of a secular anomaly with an amplitude about 0.8 nms−2^{-2} compatible with that reported by Anderson et al. A second output is the study of periodic variations of the anomaly, which we characterize as functions of the azimuthal angle φ\varphi defined by the directions Sun-Earth Antenna and Sun-Pioneer. An improved fit is obtained with periodic variations written as the sum of a secular acceleration and two sinusoids of the angles φ\varphi and 2φ2\varphi. The tests which have been performed for assessing the robustness of these results are presented.Comment: 13 pages, 6 figures, minor amendment

    An Anglo-Saxon execution cemetery at Walkington Wold, Yorkshire

    This paper presents a re-evaluation of a cemetery excavated over 30 years ago at Walkington Wold in east Yorkshire. The cemetery is characterized by careless burial on diverse alignments, and by the fact that most of the skeletons did not have associated crania. The cemetery has been variously described as being the result of an early post-Roman massacre, as providing evidence for a ‘Celtic’ head cult or as an Anglo-Saxon execution cemetery. In order to resolve the matter, radiocarbon dates were acquired and a re-examination of the skeletal remains was undertaken. It was confirmed that the cemetery was an Anglo-Saxon execution cemetery, the only known example from northern England, and the site is set into its wider context in the paper

    Towards an art and science of decision aiding for water management and planning: a participatory modelling process

    Planning and management of water resources are faced with increasingly high levels of complexity, uncertainty and conflict. Traditional technical and top-down management strategies have proved inadequate, forcing a move to more integrated forms of management, planning and decision making that can include stakeholders and communities, as well as technical experts and policy makers. These integrated forms of management require not only good technical or scientific ability, but a range of art-like skills including communication, creativity and the capacity to acknowledge and integrate diverse points of view. However, processes designed to aid such inter-organisational or multi-stakeholder decisionmaking are rare and in need of investigation. This paper proposes a process of participatory modelling using a series of semi-structured collective decision cycles which can aid decisions involving multiple stakeholders in water management and planning. The participatory modelling process outlined in this paper is designed to capture and integrate both tacit and explicit knowledge from stakeholders, right from the problem identification phase through to the final decision making, implementation and ongoing monitoring and evaluation. A brief idealised example of the participatory modelling process testing in Montpellier, France, is highlighted, as well as further questions and identified priority research areas
