    Genetic diversity of Pogonatherum paniceum (Lam.) Hack in Southwest China revealed by ISSR

    Inter-simple sequence repeats markers were used to estimate the genetic diversity of Pogonatherum paniceum (Lam.) Hack. from Sichuan Province, Yunnan Province, Chongqing City and Guangxi Zhuang Autonomous Region in China. 100 primers were carried out on 22 wild populations, 14 could produce highly reproducible inter-simple sequence repeats markersbands. Out of the 239 discernable DNA fragments, 227 were polymorphic. The percentage of polymorphic bands was 94.98% at the species level. Nei’s gene diversity and Shannon information index were 0.312 and 0.471, respectively. This indicated that the genetic diversity of P. paniceum (Lam.) Hack. was low. The values of genetic identity ranged from 0.548 to 0.820 with a mean of 0.673. Nei’s genetic distance between 22 populations ranged from 0.198 to 0.601. Unweighted pair group method with arithmetic mean cluster analysis based on Nei’s genetic distance showed that most populations were positioned into the relevant areas. Significant correlation between genetic and geographic altitude distances among populations was found by Mantel test. The high score of percentage of polymorphic bands might be caused by low frequent polymorphism distributed in different populations

    This study aims to determine the prevalence of lower urinary tract symptoms (LUTS) and level of awareness among male outpatients in Universiti Kebangsaan Malaysia Medical Centre (UKMMC). A questionnaire consisting of demographic data, questions related to knowledge, attitude and practice on BPH and the International Prostate Symptom Score (IPSS) was used for this study. Uroflowmetry and bladder scan were used to evaluate the function of the urinary tract and severity of BPH. Urine dipstick was done for glycosuria, proteinuria and haematuria. A total of 220 respondents were surveyed. The prevalence of moderately and severely symptomatic LUTS was 42.7%. The most commonly reported LUTS were nocturia (78.2%), frequency (58.2%) and incomplete emptying (44.6%). The prevalence of glycosuria, proteinuria and haematuria were 23.6%, 11.4% and 1.8% respectively. There was a significant association between increasing age with the severity of LUTS (p=0.005). Out of 102 respondents with voided urine volume greater than 150 mL, there was a significant decrease in maximum (Qmax) (p=0.039) and average (Qave) urine flow rates with every 10 years increase of age (p=0.001). The majority of respondents (59.5%) have heard of BPH before. Over 78.2% of the respondents would seek medical attention if they have LUTS with 15.9% saying they would seek traditional treatment. In conclusion, the prevalence of LUTS was high and the level of awareness was satisfactory

    Quick and sensitive determination of gene expression of fatty acid synthase in vitro by using real-time polymerase chain reaction amplification (PCR)

    Obesity results from an imbalance between energy intake and energy expenditure, which leads to a pathological accumulation of adipose tissue, but the underlying mechanism at gene level, is far from being elucidated. The objective of this study was to investigate the correlation between mRNA express from fatty acid synthase (FAS) with a different glucose level in primary adipocytes by real-time polymerase chain reaction amplification (PCR), which can aid in the understanding of the mechanism of obesity in vitro. By using the following formula, this study was able to quantify the mRNA expression of FAS of unknown samples: Y = -3.156X + 41.21 (Y = threshold cycle, X = log starting quantity). The high concentrations of glucose group significantly improved the mRNA expression of FAS (P < 0.01) rather than 0.25 and 0% concentrations of glucose. These results provide significant data that confirm an association between different glucose level and FAS expression in preadipocytes. The glucose concentration of the high group substantially augmented the mRNA expression of FAS.Key words: Expression, fatty acid synthase, lipid deposition, real-time polymerase chain reaction amplification (PCR)

    Catalysts with a single atom site allow highly tuning of the activity, stability, and reactivity of heterogeneous catalysts. Therefore, atomistic understanding of the pertinent mechanism is essential to simultaneously boost the intrinsic activity, site density, electron transport, and stability. Here, we report that atomically dispersed nickel (Ni) in zincblende cadmium-zinc sulfide quantum dots (ZCS QDs) delivers an efficient and durable photocatalytic performance for water splitting under sunlight. The finely tuned Ni atoms dispersed in ZCS QDs exhibit an ultrahigh photocatalytic H2 production activity of 18.87 mmol hour-1 g-1. It could be ascribed to the favorable surface engineering to achieve highly active sites of monovalent Ni(I) and the surface heterojunctions to reinforce the carrier separation owing to the suitable energy band structures, built-in electric field, and optimized surface H2 adsorption thermodynamics. This work demonstrates a synergistic regulation of the physicochemical properties of QDs for high-efficiency photocatalytic H2 production

    Electrospinning of poly(methyl methacrylate) nanofibers in a pump-free process

    The effects of processing parameters, including solution concentration, viscosity, nozzle diameter, voltage bias and the nozzle to collector distance, on the morpho logy and diameters of poly(methyl methacrylate) (PMMA) fibers have been systematically investigated, using a unique pump-free electrospinning method. For PMMA solution concentrations less than the critical entanglement concentration, c e, prolate spheroidshaped droplets or beads with fibers were formed, whereas at concentrations above c e, good quality beadfree fibers were formed. Quantitative analysis revealed a linear dependence between the solution viscosity and fiber diameter. Larger fiber diameters were achieved by increasing the nozzle diameter and voltage bias. Increasing the bias voltage has the additional effect of broadening the diameter distribution, as a result of splaying and splitting. By contrast, when the strength of the electrical field was reduced by increasing the distance between the nozzle and collector, the overall fiber dia meter was reduced

    Crassostrea Ariakensis In Chesapeake Bay: Growth, Disease And Mortality In Shallow Subtidal Environments

    In April 2004, triploid native (Crassostrea virginica) and nonnative (Crassostrea ariakensis) oysters were deployed in cages at four sites along a salinity gradient in Chesapeake Bay. In Maryland, the lowest salinity site was located in the Severn River and two low to mid-salinity sites were located in the Choptank and Patuxent Rivers. The highest salinity site was located in the York River in Virginia. Growth, disease acquisition, and mortality were measured in the deployed oysters through August 2006. Although ANOVA revealed that the nonnative oysters were significantly larger at the end of the experiment than the native oysters at all sites, the differences were much greater at the Virginia site (59 mm) than in Maryland waters (9-23 mm). With the exception of C. ariakensis in the Severn River, Perkinsus marinus infected both species at all sites. Prevalences and weighted prevalences in both species remained relatively low throughout the experiment, but native oysters consistently acquired higher prevalences and weighted prevalences than C. ariakensis by August 2006. With the exception of several mortality-inducing events including winter freezing and hypoxic exposure, mortality was generally low in both species. No disease-related mortality was suspected in either species given the low weighted prevalences observed. In the York River, where a substantial natural spatfall occurred in 2004, more native spat were found on C. ariakensis than on C. virginica. To our knowledge, this is the first comparison of triploid C. ariakensis to triploid C. virginica conducted in the field. Because we did not observe substantial disease-related mortality, it is too soon to draw conclusions regarding the disease tolerance of C. ariakensis in the field or its viability as a replacement for the native species

    Immunomodulatory Potential of Patchouli Alcohol Isolated from Pogostemon cablin (Blanco) Benth (Lamiaceae) in Mice

    Purpose: To isolate and purify patchouli alcohol (PA), a tricyclic sesquiterpene constituent of Pogostemon cablin, and investigate its immunomodulatory potential in Kunming mice.Methods: PA was prepared from an ethanol aqueous extract of P. cablin by silica gel column chromatography, and further purified by crystallization using n-hexane. Purity was assessed by analytical gas chromatography (GC) and confirmation of chemical structure performed by Fourier transform infrared (FTIR), nuclear magnetic resonance (NMR) spectroscopy and mass spectrometry (MS). The effect of PA from Pogostemon cablin on immunological function was studied by macrophage phagocytosis, immune organ index, serum immunoglobulin level and delayed type  hypersensitivity (DTH) in mice that were administered orally doses of 20, 40 and 80 mg/kg.Results: The purity of PA was 99.3%. The oral administration of PA (40, or 80 mg/kg body weight) significantly increased the phagocytic index (p < 0.05), compared with prednisone acetate (PR) group. Administration of PA (80 mg/kg) boosted the production of circulating serum IgM (0.081 ± 0.010) and IgG (1.296 ± 0.120), while IgM and IgG in PR group was 0.069 ± 0.011 (p < 0.01) and 1.180 ± 0.070 (p < 0.01) respectively. However, PA (20 mg/kg) treatment elicited significant decrease in DTH induced by 2, 4-dinitro-chlorobenzene (DNCB) in mice (1.03 ± 0.40, p < 0.05), in comparison to DNCB-induced group (1.67 ± 0.84 mg).Conclusion: These results suggest that PA has significant immunomodulatory properties which probably act by activating mononuclear phagocytic system, augmenting humoral immune response while suppressing cellular immune response.Keywords: Patchouli alcohol, Pogostemon cablin, Immunomodulatory, Phagocytic index, Macrophag
