    Water transport through (7,7) carbon nanotubes of different lengths using molecular dynamics

    Non-equilibrium molecular dynamics simulations are used to investigate water transport through (7,7) CNTs, examining how changing the CNT length affects the internal flow dynamics. Pressure-driven water flow through CNT lengths ranging from 2.5 to 50 nm is simulated. We show that under the same applied pressure difference an increase in CNT length has a negligible effect on the resulting mass flow rate and fluid flow velocity. Flow enhancements over hydrodynamic expectations are directly proportional to the CNT length. Axial profiles of fluid properties demonstrate that entrance and exit effects are significant in the transport of water along CNTs. Large viscous losses in these entrance/exit regions lead into central “developed” regions in longer CNTs where the flow is effectively frictionless

    Tropomyosin (Tm) is an evolutionarily conserved ?-helical coiled-coil protein, dimers of which form end-to-end polymers capable of associating with and stabilising actin-filaments and regulate myosin function. The fission yeast, Schizosaccharomyces pombe, possesses a single essential Tm, Cdc8, which can be acetylated on its amino terminal methionine to increase its affinity for actin and enhance its ability to regulate myosin function. We have designed and generated a number of novel Cdc8 mutant proteins with amino terminal substitutions to explore how stability of the Cdc8-polymer overlap region affects the regulatory function of this Tm. By correlating the stability of each protein, its propensity to form stable polymers, its ability to associate with actin and to regulate myosin, we have shown the stability of the amino terminal of the Cdc8 ?-helix is crucial for Tm function. In addition we have identified a novel Cdc8 mutant with increased amino-terminal stability, dimers of which are capable of forming Tm-polymers significantly longer than the wild-type protein. This protein had a reduced affinity for actin with respect to wild type, and was unable to regulate actomyosin interactions. The data presented here are consistent with acetylation providing a mechanism for modulating the formation and stability of Cdc8 polymers within the fission yeast cell. The data also provide evidence for a mechanism in which Tm dimers form end-to-end polymers on the actin-filament, consistent with a cooperative model for Tm binding to actin

DiRAC is part of the National E-Infrastructure.We investigate how star formation efficiency can be significantly decreased by the removal of a molecular cloud’s envelope by feedback from an external source. Feedback from star formation has difficulties halting the process in dense gas but can easily remove the less dense and warmer envelopes where star formation does not occur. However, the envelopes can play an important role keeping their host clouds bound by deepening the gravitational potential and providing a constraining pressure boundary. We use numerical simulations to show that removal of the cloud envelopes results in all cases in a fall in the star formation efficiency (SFE). At 1.38 free-fall times our 4 pc cloud simulation experienced a drop in the SFE from 16 to six percent, while our 5 pc cloud fell from 27 to 16 per cent. At the same time, our 3 pc cloud (the least bound) fell from an SFE of 5.67 per cent to zero when the envelope was lost. The star formation efficiency per free-fall time varied from zero to ≈0.25 according to α, defined to be the ratio of the kinetic plus thermal to gravitational energy, and irrespective of the absolute star forming mass available. Furthermore the fall in SFE associated with the loss of the envelope is found to even occur at later times. We conclude that the SFE will always fall should a star forming cloud lose its envelope due to stellar feedback, with less bound clouds suffering the greatest decrease.PostprintPeer reviewe

    3D terrain mapping and filtering from coarse resolution data cubes extracted from real-aperture 94 GHz radar

In this study, we develop a new technique, called the PCFilt-94 algorithm, to extract 3-D point clouds from coarse-resolution millimeter-wave radar data cubes and quantify their associated uncertainties. A technique to noncoherently average neighboring waveforms surrounding each AVTIS2 range profile was developed to reduce speckles and was found to reduce point cloud uncertainty by 13% at long range and 20% at short range. Furthermore, a Voronoi-based point cloud outlier removal algorithm was implemented, which iteratively removes outliers in a point cloud until the process converges to the removal of 0 points. Taken together, the new processing methodology produces a stable point cloud, which means that: 1) it is repeatable even when using different point cloud extraction and filtering parameter values during preprocessing and 2) is less sensitive to overfiltering through the point cloud processing workflow. Using an optimal number of ground control points (GCPs) for georeferencing, which was determined to be 3 at close ranges (3 km), point cloud uncertainty was estimated to be approximately 1.5 m at 1.5 km to 3 m at 3 km and followed a Lorentzian distribution. These uncertainties are smaller than those reported for other close-range radar systems used for terrain mapping. The results of this study should be used as a benchmark for future application of millimeter-wave radar systems for 3-D terrain mapping.Peer reviewe

    PurposeThe RPE cell line ARPE-19 provides a dependable and widely used alternative to native RPE. However, replication of the native RPE phenotype becomes more difficult because these cells lose their specialized phenotype after multiple passages. Compounding this problem is the widespread use of ARPE-19 cells in an undifferentiated state to attempt to model RPE functions. We wished to determine whether suitable culture conditions and differentiation could restore the RPE-appropriate expression of genes and proteins to ARPE-19, along with a functional and morphological phenotype resembling native RPE. We compared the transcriptome of ARPE-19 cells kept in long-term culture with those of primary and other human RPE cells to assess the former's inherent plasticity relative to the latter.MethodsARPE-19 cells at passages 9 to 12 grown in DMEM containing high glucose and pyruvate with 1% fetal bovine serum were differentiated for up to 4 months. Immunocytochemistry was performed on ARPE-19 cells grown on filters. Total RNA extracted from ARPE-19 cells cultured for either 4 days or 4 months was used for RNA sequencing (RNA-Seq) analysis using a 2 × 50 bp paired end protocol. The RNA-Seq data were analyzed to identify the affected pathways and recognize shared ontological classification among differentially expressed genes. RPE-specific mRNAs and miRNAs were assessed with quantitative real-time (RT)-PCR, and proteins with western blotting.ResultsARPE-19 cells grown for 4 months developed the classic native RPE phenotype with heavy pigmentation. RPE-expressed genes, including RPE65, RDH5, and RDH10, as well as miR-204/211, were greatly increased in the ARPE-19 cells maintained at confluence for 4 months. The RNA-Seq analysis provided a comprehensive view of the relative abundance and differential expression of the genes in the differentiated ARPE-19 cells. Of the 16,757 genes with detectable signals, nearly 1,681 genes were upregulated, and 1,629 genes were downregulated with a fold change of 2.5 or more differences between 4 months and 4 days of culture. Gene Ontology analysis showed that the upregulated genes were associated with visual cycle, phagocytosis, pigment synthesis, cell differentiation, and RPE-related transcription factors. The majority of the downregulated genes play a role in cell cycle and proliferation.ConclusionsThe ARPE-19 cells cultured for 4 months developed a phenotype characteristic of native RPE and expressed proteins, mRNAs, and miRNAs characteristic of the RPE. Comparison of the ARPE-19 RNA-Seq data set with that of primary human fetal RPE, embryonic stem cell-derived RPE, and native RPE revealed an important overall similar expression ratio among all the models and native tissue. However, none of the cultured models reached the absolute values in the native tissue. The results of this study demonstrate that low-passage ARPE-19 cells can express genes specific to native human RPE cells when appropriately cultured and differentiated

    Simultaneous Modelling of the Stellar Halo and Globular Cluster System of NGC 5128

    An important test for models of galaxy formation lies in the metallicity distribution functions (MDFs) of spheroid stars and their globular clusters (GCs).We have compared the MDFs obtained from spectroscopy of the GCs and the star-by-star photometry of red giants in the nearby elliptical NGC 5128, with the predictions of a semi-analytic galaxy formation model. We have selected model ellipticals comparable in luminosity and environment to NGC 5128, and derived their MDFs. A direct comparison between models and data shows that the MDFs are qualitatively similar, both have stellar components which are predominantly metal-rich (0.8Z), with a small fraction of metal-poor stars extending down to 0.002Z. The model MDFs show only small variations, whether they are brightest cluster galaxies or low luminosity group ellipticals. Our comparison also reveals that these model MDFs harbour a greater fraction of stars above solar metallicity than the observations, producing generally more metal-rich (by 0.1 dex) MDFs. One possibility is that the outer-bulge observations are missing some of the highest metallicity stars. We find good agreement between the model and observed GC MDFs, provided that the metal-poor GC formation is halted early in the model. Under this proviso, both the models and data are bimodal with peaks at 0.1Z and Z, and cover similar metallicity ranges. This broad agreement for the stars and GCs suggests that the bulk of the stellar population in NGC 5128 may have been built up in a hierarchical fashion, involving both quiescent and merger-induced star formation. The existence of age structure amongst the metal-rich GCs needs to be tested against high-quality data for this galaxy.Comment: 13 pages, 9 figures and a table, accepted by MNRA

    The Kinematics and Dynamics of the Globular Clusters and the Planetary Nebulae of NGC 5128

    A new kinematic and dynamic study of the halo of the giant elliptical galaxy, NGC 5128, is presented. From a spectroscopically confirmed sample of 340 globular clusters and 780 planetary nebulae, the rotation amplitude, rotation axis, velocity dispersion, and the total dynamical mass are determined for the halo of NGC 5128. The globular cluster kinematics were searched for both radial dependence and metallicity dependence by subdividing the globular cluster sample into 158 metal-rich ([Fe/H] > -1.0) and 178 metal-poor ([Fe/H] < -1.0) globular clusters. Our results show the kinematics of the metal-rich and metal-poor subpopulations are quite similar. The kinematics are compared to the planetary nebula population where differences are apparent in the outer regions of the halo. The total mass of NGC 5128 is found using the Tracer Mass estimator (Evans et al. 2003), to determine the mass supported by internal random motions, and the spherical component of the Jeans equation to determine the mass supported by rotation. We find a total mass of (1.0+/-0.2) x 10^(12) Msun from the planetary nebulae data out to a projected radius of 90 kpc and (1.3+/-0.5) x 10^(12) Msun from the globular clusters out to a projected radius of 50 kpc. Lastly, we present a new and homogeneous catalog of known globular clusters in NGC 5128. This catalog combines all previous definitive cluster identifications from radial velocity studies and HST imaging studies, as well as 80 new globular clusters from a study of M.A. Beasley et al.
