    Reduction in squamous cell carcinomas in mouse skin by dietary zinc supplementation.

    Inadequate dietary Zn consumption increases susceptibility to esophageal and other cancers in humans and model organisms. Since Zn supplementation can prevent cancers in rodent squamous cell carcinoma (SCC) models, we were interested in determining if it could have a preventive effect in a rodent skin cancer model, as a preclinical basis for considering a role for Zn in prevention of human nonmelanoma skin cancers, the most frequent cancers in humans. We used the 7,12-dimethyl benzanthracene carcinogen/phorbol myristate acetate tumor promoter treatment method to induce skin tumors in Zn-sufficient wild-type and Fhit (human or mouse protein) knockout mice. Fhit protein expression is lost in \u3e50% of human cancers, including skin SCCs, and Fhit-deficient mice show increased sensitivity to carcinogen induction of tumors. We hypothesized that: (1) the skin cancer burdens would be reduced by Zn supplementation; (2) Fhit(-/-) (Fhit, murine fragile histidine triad gene) mice would show increased susceptibility to skin tumor induction versus wild-type mice. 30 weeks after initiating treatment, the tumor burden was increased ~2-fold in Fhit(-/-) versus wild-type mice (16.2 versus 7.6 tumors, P \u3c 0.001); Zn supplementation significantly reduced tumor burdens in Fhit(-/-) mice (males and females combined, 16.2 unsupplemented versus 10.3 supplemented, P = 0.001). Most importantly, the SCC burden was reduced after Zn supplementation in both strains and genders of mice, most significantly in the wild-type males (P = 0.035). Although the mechanism(s) of action of Zn supplementation in skin tumor prevention is not known in detail, the Zn-supplemented tumors showed evidence of reduced DNA damage and some cohorts showed reduced inflammation scores. The results suggest that mild Zn supplementation should be tested for prevention of skin cancer in high-risk human cohorts

    Hamiltonian-Driven Adaptive Dynamic Programming with Efficient Experience Replay

    This article presents a novel efficient experience-replay-based adaptive dynamic programming (ADP) for the optimal control problem of a class of nonlinear dynamical systems within the Hamiltonian-driven framework. The quasi-Hamiltonian is presented for the policy evaluation problem with an admissible policy. With the quasi-Hamiltonian, a novel composite critic learning mechanism is developed to combine the instantaneous data with the historical data. In addition, the pseudo-Hamiltonian is defined to deal with the performance optimization problem. Based on the pseudo-Hamiltonian, the conventional Hamilton–Jacobi–Bellman (HJB) equation can be represented in a filtered form, which can be implemented online. Theoretical analysis is investigated in terms of the convergence of the adaptive critic design and the stability of the closed-loop systems, where parameter convergence can be achieved under a weakened excitation condition. Simulation studies are investigated to verify the efficacy of the presented design scheme

    Metallicity Gradients in the Milky Way Disk as Observed by the SEGUE Survey

    The observed radial and vertical metallicity distribution of old stars in the Milky Way disk provides a powerful constraint on the chemical enrichment and dynamical history of the disk. We present the radial metallicity gradient, \Delta[Fe/H]/\Delta R, as a function of height above the plane, |Z|, using 7010 main sequence turnoff stars observed by the Sloan Extension for Galactic Understanding and Exploration (SEGUE) survey. The sample consists of mostly old thin and thick disk stars, with a minimal contribution from the stellar halo, in the region 6 < R < 16 kpc, 0.15 < |Z| < 1.5 kpc. The data reveal that the radial metallicity gradient becomes flat at heights |Z| > 1 kpc. The median metallicity at large |Z| is consistent with the metallicities seen in outer disk open clusters, which exhibit a flat radial gradient at [Fe/H] ~ -0.5. We note that the outer disk clusters are also located at large |Z|; because the flat gradient extends to small R for our sample, there is some ambiguity in whether the observed trends for clusters are due to a change in R or |Z|. We therefore stress the importance of considering both the radial and vertical directions when measuring spatial abundance trends in the disk. The flattening of the gradient at high |Z| also has implications on thick disk formation scenarios, which predict different metallicity patterns in the thick disk. A flat gradient, such as we observe, is predicted by a turbulent disk at high redshift, but may also be consistent with radial migration, as long as mixing is strong. We test our analysis methods using a mock catalog based on the model of Sch\"onrich & Binney, and we estimate our distance errors to be ~25%.     ENZYMATIC DETERMINATION OF TEPP RESIDUES ON RED-WINGED BLACKBIRDS

    In 1966 the U.S. Bureau of Sport Fisheries and Wildlife, since renamed the U.S. Fish and Wildlife Service, was considering the use of commercial tetraethylpyrophosphate (TEPP) for spraying problem bird roosts. Although TEPP was known to be a fast degrading organophosphate, the Bureau decided that a determination of TEPP residues on birds sprayed under simulated field conditions would be useful in reaching a decision on possible use of this material. The determination was done with an enzymatic Warburg manometric method for measuring the inhibition of acetylcholinesterase (AChE), a major cause of neurotoxication. It was found that at simulated roost temperatures of 7·8°C with relative humidity of 57·60%, the AChE inhibition activity of TEPP in red·winged blackbirds diminished rapidly in the first 2 days. However, after 19 days, an indicated 2.2% (2.8% statistically possible) remained as cholinergic inhibition residues that could be hazardous to humans or nontarget species, considerably more than 99% loss in 45.2 hr at 26°C that had been previously reported. This information, among others, was used by the Bureau in deciding not to pursue the use of TEPP in spraying problem bird roosts
