Cell Recovery in Bronchoalveolar Lavage Fluid in Smokers Is Dependent on Cumulative Smoking History
Background: Smoking is a risk factor for various lung diseases in which BAL may be used as a part of a clinical investigation. Interpretation of BAL fluid cellularity is however difficult due to high variability, in particular among smokers. In this study we aimed to evaluate the effect of smoking on BAL cellular components in asymptomatic smokers. The effects of smoking cessation, age and gender were also investigated in groups of smokers and exsmokers. Methods: We performed a retrospective review of BAL findings, to our knowledge the largest single center investigation, in our department from 1999 to 2009. One hundred thirty two current smokers (48 males and 84 females) and 44 ex-smokers (16 males and 28 females) were included. A group of 295 (132 males and 163 females) never-smokers served as reference. Result: The median [5β95 pctl] total number of cells and cell concentration in current smokers were 63.4 [28.6β132.1]610 6 and 382.1 [189.7β864.3]610 6 /L respectively and correlated positively to the cumulative smoking history. Macrophages were the predominant cell type (96.7 % [90.4β99.0]) followed by lymphocytes (2 % [0.8β7.7]) and neutrophils (0.6 % [0β2.9]). The concentration of all inflammatory cells was increased in smokers compared to never smokers and ex-smokers. BAL fluid recovery was negatively correlated with age (p,0.001). Smoking men had a lower BAL fluid recovery than smoking women. Conclusion: Smoking has a profound effect on BAL fluid cellularity, which is dependent on smoking history. Isolated and dynamical horizons and their applications
Over the past three decades, black holes have played an important role in
quantum gravity, mathematical physics, numerical relativity and gravitational
wave phenomenology. However, conceptual settings and mathematical models used
to discuss them have varied considerably from one area to another. Over the
last five years a new, quasi-local framework was introduced to analyze diverse
facets of black holes in a unified manner. In this framework, evolving black
holes are modeled by dynamical horizons and black holes in equilibrium by
isolated horizons. We review basic properties of these horizons and summarize
applications to mathematical physics, numerical relativity and quantum gravity.
This paradigm has led to significant generalizations of several results in
black hole physics. Specifically, it has introduced a more physical setting for
black hole thermodynamics and for black hole entropy calculations in quantum
gravity; suggested a phenomenological model for hairy black holes; provided
novel techniques to extract physics from numerical simulations; and led to new

High systemic IL-6 is associated with worse prognosis in patients with non-small cell lung cancer
Characteristic cytokine patterns have been described in different cancer patients and they are related to their diagnosis, prognosis, prediction of treatment responses and survival. A panel of cytokines was evaluated in the plasma of non-small cell lung cancer (NSCLC) patients and healthy controls to investigate their profile and relationship with clinical characteristics and overall survival. The case-controlled cross-sectional study design recruited 77 patients with confirmed diagnosis of NSCLC (cases) and 91 healthy subjects (controls) aimed to examine peripheral pro-inflammatory and anti-inflammatory cytokines (IL-2, IL-4, IL-6, IL-10, IL-17A, TNF and IFN-gamma) by Cytometry Beads Arrays (CBA Flex) in. The cytokine IL-6 showed a statistically significant difference among groups with increased expression in the case group (p < 0.001). The correlation between the cytokines expression with patient's clinical characteristics variables revealed the cytokine IL-6 was found to be associated with gender, showing higher levels in male (p = 0.036), whereas IL-17A levels were associated with TNM stage, being higher in III-IV stages (p = 0.044). We observed worse overall survival for individuals with high levels of IL-6 when compared to those with low levels of this cytokine in 6, 12 and 24 months. Genome-Wide Screen of Three Herpesviruses for Protein Subcellular Localization and Alteration of PML Nuclear Bodies
Herpesviruses are large, ubiquitous DNA viruses with complex host interactions, yet many of the proteins encoded by these viruses have not been functionally characterized. As a first step in functional characterization, we determined the subcellular localization of 234 epitope-tagged proteins from herpes simplex virus, cytomegalovirus, and EpsteinβBarr virus. Twenty-four of the 93 proteins with nuclear localization formed subnuclear structures. Twelve of these localized to the nucleolus, and five at least partially localized with promyelocytic leukemia (PML) bodies, which are known to suppress viral lytic infection. In addition, two proteins disrupted Cajal bodies, and 19 of the nuclear proteins significantly decreased the number of PML bodies per cell, including six that were shown to be SUMO-modified. These results have provided the first functional insights into over 120 previously unstudied proteins and suggest that herpesviruses employ multiple strategies for manipulating nuclear bodies that control key cellular processes
1-methylnicotinamide and its structural analog 1,4-dimethylpyridine for the prevention of cancer metastasis
Background: 1-methylnicotinamide (1-MNA), an endogenous metabolite of nicotinamide, has recently gained interest due to its anti-inflammatory and anti-thrombotic activities linked to the COX-2/PGI2 pathway. Given the previously reported anti-metastatic activity of prostacyclin (PGI2), we aimed to assess the effects of 1-MNA and its structurally related analog, 1,4-dimethylpyridine (1,4-DMP), in the prevention of cancer metastasis. Methods: All the studies on the anti-tumor and anti-metastatic activity of 1-MNA and 1,4-DMP were conducted using the model of murine mammary gland cancer (4T1) transplanted either orthotopically or intravenously into female BALB/c mouse. Additionally, the effect of the investigated molecules on cancer cell-induced angiogenesis was estimated using the matrigel plug assay utilizing 4T1 cells as a source of pro-angiogenic factors. Results: Neither 1-MNA nor 1,4-DMP, when given in a monotherapy of metastatic cancer, influenced the growth of 4T1 primary tumors transplanted orthotopically; however, both compounds tended to inhibit 4T1 metastases formation in lungs of mice that were orthotopically or intravenously inoculated with 4T1 or 4T1-luc2-tdTomato cells, respectively. Additionally, while 1-MNA enhanced tumor vasculature formation and markedly increased PGI2 generation, 1,4-DMP did not have such an effect. The anti-metastatic activity of 1-MNA and 1,4-DMP was further confirmed when both agents were applied with a cytostatic drug in a combined treatment of 4T1 murine mammary gland cancer what resulted in up to 80 % diminution of lung metastases formation. Conclusions: The results of the studies presented below indicate that 1-MNA and its structural analog 1,4-DMP prevent metastasis and might be beneficially implemented into the treatment of metastatic breast cancer to ensure a comprehensive strategy of metastasis control

Macrophages in Breast Cancer: Do Involution Macrophages Account for the Poor Prognosis of Pregnancy-Associated Breast Cancer?
Macrophage influx is associated with negative outcomes for women with breast cancer and has been demonstrated to be required for metastasis of mammary tumors in mouse models. Pregnancy-associated breast cancer is characterized by particularly poor outcomes, however the reasons remain obscure. Recently, post-pregnancy mammary involution has been characterized as having a wound healing signature. We have proposed the involution-hypothesis, which states that the wound healing microenvironment of the involuting gland is tumor promotional. Macrophage influx is one of the prominent features of the involuting gland, identifying the macrophage a potential instigator of tumor progression and a novel target for breast cancer treatment and prevention
Loop Quantum Cosmology
Quantum gravity is expected to be necessary in order to understand situations
where classical general relativity breaks down. In particular in cosmology one
has to deal with initial singularities, i.e. the fact that the backward
evolution of a classical space-time inevitably comes to an end after a finite
amount of proper time. This presents a breakdown of the classical picture and
requires an extended theory for a meaningful description. Since small length
scales and high curvatures are involved, quantum effects must play a role. Not
only the singularity itself but also the surrounding space-time is then
modified. One particular realization is loop quantum cosmology, an application
of loop quantum gravity to homogeneous systems, which removes classical
singularities. Its implications can be studied at different levels. Main
effects are introduced into effective classical equations which allow to avoid
interpretational problems of quantum theory. They give rise to new kinds of
early universe phenomenology with applications to inflation and cyclic models.
To resolve classical singularities and to understand the structure of geometry
around them, the quantum description is necessary. Classical evolution is then
replaced by a difference equation for a wave function which allows to extend
space-time beyond classical singularities. One main question is how these
homogeneous scenarios are related to full loop quantum gravity, which can be
dealt with at the level of distributional symmetric states. Finally, the new
structure of space-time arising in loop quantum gravity and its application to
