    Evaluation of the supramolecular interaction of Congo red with cucurbiturils using mass spectrometry and spectroscopic methods

    The ability of cucurbit[n]urils (CB[n]) to decolourise aqueous solutions of the azo dye Congo red (CR) was described more than a century ago alongside the first synthesis of CB[n]. No subsequent studies of the nature of the physical interactions have been reported despite the interest in using CB[n] as adsorbents for the removal of CR and related organic dyes from wastewaters. In the present work the supramolecular interaction between CB[n] (n = 7, 8) and CR was studied by electrospray ionisation mass spectrometry (ESI-MS), 1H NMR, and solid-state characterisation of isolated complexes. Under positive ESI, the formation of host–guest complexes in the gas phase was not observed, suggesting that CR anions do not interact with the portals and the nonpolar inner cavity of the CB[n] molecules. Conversely, under negative ESI, 1 : 1 and higher order (1 : 2, 2 : 1, 3 : 1 and 2 : 2) CR: CB[7] and CR: CB[8] adducts were detected, which is attributed to interaction between CR and the outer surface hydrogens of CB[n]. Solid-state supramolecular adducts between CB[n] and CR were isolated from aqueous media under either ambient conditions, giving structures denoted as CR@CB[n](RT), or hydrothermal (100 ºC) conditions, giving structures denoted as CR@CB[n](100). The adducts were characterised by elemental and thermogravimetric analyses (TGA), powder X-ray diffraction (PXRD), and spectroscopic methods (FT-IR, FT-Raman, 13C{1H} CP MAS NMR, UV/vis and near-IR absorption, fluorescence excitation and emission). The interaction conditions and acid content of the starting CB[n] influenced the protonation state of CR molecules. CR@CB[7](RT) contained only unprotonated CR, while CR@CB[8](100) contained exclusively protonated CR (ammonium and azonium/quinoid structures).     Foi desenvolvido um protótipo de semeadora, apropriado para semeadura de sementes pequenas (de 1 a 5 mm diâmetro) e que utiliza um fluido não-newtoniano (pastoso) como veículo condutor de sementes e como fonte inicial de umidade. Sementes de alface, cebola e mostarda foram misturadas ao fluido em diferentes níveis de concentrações para posterior avaliação do espaçamento entre sementes. O equipamento permitiu diferentes distribuições de sementes, podendo atender às necessidades de plantio de cada cultura. A drill prototype, for sowing small seeds (1 to 5 mm in diameter) using a non-Newtonian fluid as a seed carrying medium and also as an initial source of moisture has been developed. In evaluating the performance of the machine, seeds of lettuce, onion and mustard were mixed with the fluid at different concentrations. The equipment has allowed different seed distributions, achieving crop sowing requirements

    Big data and data repurposing – using existing data to answer new questions in vascular dementia research

    Introduction: Traditional approaches to clinical research have, as yet, failed to provide effective treatments for vascular dementia (VaD). Novel approaches to collation and synthesis of data may allow for time and cost efficient hypothesis generating and testing. These approaches may have particular utility in helping us understand and treat a complex condition such as VaD. Methods: We present an overview of new uses for existing data to progress VaD research. The overview is the result of consultation with various stakeholders, focused literature review and learning from the group’s experience of successful approaches to data repurposing. In particular, we benefitted from the expert discussion and input of delegates at the 9th International Congress on Vascular Dementia (Ljubljana, 16-18th October 2015). Results: We agreed on key areas that could be of relevance to VaD research: systematic review of existing studies; individual patient level analyses of existing trials and cohorts and linking electronic health record data to other datasets. We illustrated each theme with a case-study of an existing project that has utilised this approach. Conclusions: There are many opportunities for the VaD research community to make better use of existing data. The volume of potentially available data is increasing and the opportunities for using these resources to progress the VaD research agenda are exciting. Of course, these approaches come with inherent limitations and biases, as bigger datasets are not necessarily better datasets and maintaining rigour and critical analysis will be key to optimising data use

    Co-circulation of Multidrug-resistant Shigella Among Men Who Have Sex With Men in Australia.

    BACKGROUND: In urban Australia, the burden of shigellosis is either in returning travelers from shigellosis-endemic regions or in men who have sex with men (MSM). Here, we combine genomic data with comprehensive epidemiological data on sexual exposure and travel to describe the spread of multidrug-resistant Shigella lineages. METHODS: A population-level study of all cultured Shigella isolates in the state of Victoria, Australia, was undertaken from 1 January 2016 through 31 March 2018. Antimicrobial susceptibility testing, whole-genome sequencing, and bioinformatic analyses of 545 Shigella isolates were performed at the Microbiological Diagnostic Unit Public Health Laboratory. Risk factor data on travel and sexual exposure were collected through enhanced surveillance forms or by interviews. RESULTS: Rates of antimicrobial resistance were high, with 17.6% (95/541) and 50.6% (274/541) resistance to ciprofloxacin and azithromycin, respectively. There were strong associations between antimicrobial resistance, phylogeny, and epidemiology. Specifically, 2 major MSM-associated lineages were identified: a Shigellasonnei lineage (n = 159) and a Shigella flexneri 2a lineage (n = 105). Of concern, 147/159 (92.4%) of isolates within the S. sonnei MSM-associated lineage harbored mutations associated with reduced susceptibility to recommended oral antimicrobials: namely, azithromycin, trimethoprim-sulfamethoxazole, and ciprofloxacin. Long-read sequencing demonstrated global dissemination of multidrug-resistant plasmids across Shigella species and lineages, but predominantly associated with MSM isolates. CONCLUSIONS: Our contemporary data highlight the ongoing public health threat posed by resistant Shigella, both in Australia and globally. Urgent multidisciplinary public health measures are required to interrupt transmission and prevent infection

    Early-Life Determinants of Total and HDL Cholesterol Concentrations in 8-Year-Old Children; The PIAMA Birth Cohort Study

    BACKGROUND: Adult cholesterol concentrations might be influenced by early-life factors, such as breastfeeding and birth weight, referred to as "early programming". How such early factors exert their influence over the life course is still poorly understood. Evidence from studies in children and adolescents is scarce and conflicting. We investigated the influence of 6 different perinatal risk factors on childhood total and HDL cholesterol concentrations and total-to-HDL cholesterol ratio measured at 8 years of age, and additionally we studied the role of the child's current Body Mass Index (BMI). METHODS: Anthropometric measures and blood plasma samples were collected during a medical examination in 751 8-year-old children participating in the prospective Prevention and Incidence of Asthma and Mite Allergy (PIAMA) birth cohort study. Linear and logistic regression were performed to estimate associations of total and HDL cholesterol concentrations with breastfeeding, birth weight, infant weight gain, maternal overweight before pregnancy, gestational diabetes and maternal smoking during pregnancy, taking into account the child's current BMI. RESULTS: Linear regressions showed an association between total-to-HDL cholesterol ratio and maternal pre-pregnancy overweight (β = 0.15, Confidence Interval 95% (CI): 0.02, 0.28), rapid infant weight gain (β = 0.13, 95%CI: 0.01, 0.26), and maternal smoking during pregnancy (β = 0.14, 95%CI: 0.00, 0.29). These associations were partly mediated by the child's BMI. CONCLUSION: Total-to-HDL cholesterol ratio in 8-year-old children was positively associated with maternal pre-pregnancy overweight, maternal smoking during pregnancy and rapid infant weight gain
