    Teaching research methods: Introducing a psychogeographical approach

    This paper explores teaching business students research methods using a psychogeographical approach, specifically the technique of dérive. It responds to calls for new ways of teaching in higher education and addresses the dearth of literature on teaching undergraduate business students qualitative research methods. Psychogeography challenges the dominance of questionnaires and interviews, introduces students to data variety, problematizes notions of success and illuminates the importance of observation and location. Using two studies with undergraduate students, the authors emphasize place and setting, the perception of purpose, the choice of data, criteria of success and the value of guided reflection and self-reflection in students’ learning. Additionally the data reflect on the way students perceive research about management and the nature of management itself. The paper concludes that the deployment of psychogeography to teach business research methods although complex and fraught with difficulty is nevertheless viable, educationally productive and worthy of further research

    Crisis as a plague on organisation: Defoe and A Journal of the Plague Year

    Purpose – The purpose of this paper is to enrich the understanding of current models of organisational response to crises and offer additional perspectives on some of these models. It is also intended to confirm the value of fiction as a truth-seeking and hermeneutic device for enriching the imagination. Design/methodology/approach – The study uses Daniel Defoe’s 1722 novel A Journal of the Plague Year to draw parallels between his portrayal of the London Great Plague of 1665 and the management of modern-day crises. Defoe uses London’s ordeal of the Great Plague to advise those subjected to future crises. Through his representation of plague-ridden streets, Defoe shows stakeholders acting in ways described in current crisis management literature. Findings – The authors note how the management of the Plague crisis was unsuccessful and they challenge the very idea of managing a true crisis. The authors are able to illustrate and offer refinements to the Pearson and Clair (1998) and Janes (2010) models of crisis management as well as confirming the value of their constructs across a lapse of centuries. Research limitations/implications – Although it is an examination of a single novel, the findings suggest value in conceptualising organisational crises in innovative and more imaginative ways. Originality/value – It confirms the heuristic value of using fiction to understand organisational change and adds value to current model

    Supporting students with learning disabilities to explore linear relationships using online learning objects

However, students’ abilities connect different representations of linear relationships have proven to be challenging. In response, a computer-based instructional sequence was designed to support students’ understanding of the connections among representations. In this paper we report on the affordances of this dynamic mode of representation specifically for students with learning disabilities.     Energetics of Shaker K channels block by inactivation peptides

    A synthetic peptide of the NH2-terminal inactivation domain of the ShB channel blocks Shaker channels which have an NH2-terminal deletion and mimics many of the characteristics of the intramolecular inactivation reaction. To investigate the role of electrostatic interactions in both peptide block and the inactivation process we measured the kinetics of block of macroscopic currents recorded from the intact ShB channel, and from ShB delta 6-46 channels in the presence of peptides, at different ionic strengths. The rate of inactivation and the association rate constants (k(on)) for the ShB peptides decreased with increasing ionic strength. k(on) for a more positively charged peptide was more steeply dependent on ionic strength consistent with a simple electrostatic mechanism of enhanced diffusion. This suggests that a rate limiting step in the inactivation process is the diffusion of the NH2-terminal domain towards the pore. The dissociation rates (k(off)) were insensitive to ionic strength. The temperature dependence of k(on) for the ShB peptide was very high, (Q10 = 5.0 +/- 0.58), whereas k(off) was relatively temperature insensitive (Q10 approximately 1.1). The results suggest that at higher temperatures the proportion of time either the peptide or channel spends in the correct conformation for binding is increased. There were two components to the time course of recovery from block by the ShB peptide, indicating two distinct blocked states, one of which has similar kinetics and dependence on external K+ concentration as the inactivated state of ShB. The other is voltage-dependent and at -120 mV is very unstable. Increasing the net charge on the peptide did not increase sensitivity to knock-off by external K+. We propose that the free peptide, having fewer constraints than the tethered NH2-terminal domain binds to a similar site on the channel in at least two different conformations

    The trafficking and targeting of P2X receptors

    The functional expression of P2X receptors at the plasma membrane is dependent on their trafficking along secretory and endocytic pathways. There are seven P2X receptor subunits, and these differ in their subcellular distributions because they have very different trafficking properties. Some are retained within the endoplasmic reticulum (ER), while others are predominantly at the cell surface or within endosomes and lysosomes. Changes in recruitment of receptors to and from the plasma membrane provides a way of rapidly up- or down-regulating the cellular response to adenosine triphosphate (ATP). An additional layer of regulation is the targeting of these receptors within the membranes of each compartment, which affects their stability, function and the nature of the effector proteins with which they form signaling complexes. The trafficking and targeting of P2X receptors is regulated by their interactions with other proteins and with lipids and we can expect this to vary in a cell-type specific manner and in response to changes in the environment giving rise to differences in receptor activity and function

    Interactions of amino terminal domains of Shaker K channels with a pore blocking site studied with synthetic peptides

    Synthetic peptides of the five alternative NH2-terminal sequences of Shaker when applied to the cytoplasmic side of ShB channels that have an NH2-terminal deletion (ShB delta 6-46) block the channel with potencies correlated with the rate of inactivation in the corresponding variant. These peptides share no sequence similarity and yet three out of the five have apparent dissociation constants between 2 and 15 microM, suggesting that the specificity requirements for binding are low. To identify the primary structural determinants required for effective block of ShB delta 6-46, we examined the effects of substitutions made to the 20 residue ShB peptide on association and dissociation rates. Nonpolar residues within the peptide appear to be important in stabilizing the binding through hydrophobic interactions. Substitutions to leucine-7 showed there was a clear correlation between hydrophobicity and the dissociation rate constant (koff) with little effect on the association rate constant (kon). Substituting charged residues for hydrophobic residues within the region 4-8 disrupted binding. Within the COOH-terminal half of the peptide, substitutions that increased the net positive charge increased kon with relatively small changes in koff, suggesting the involvement of long-range electrostatic interactions in increasing the effective concentration of the peptide. Neutralizing charged residues produced small changes in koff. Charges within the region 12-20 act equivalently; alterations which conserved net charge produced little effect on either kon or koff. The results are consistent with this region of the peptide having an extended conformation and suggest that when bound this region makes few contacts with the channel protein and remains relatively unconstrained. Analogous mutations within the NH2-terminal domain of the intact ShB channel produced qualitatively similar effects on blocking and unblocking rates

    Plasma Wakefield Acceleration with a Modulated Proton Bunch

    The plasma wakefield amplitudes which could be achieved via the modulation of a long proton bunch are investigated. We find that in the limit of long bunches compared to the plasma wavelength, the strength of the accelerating fields is directly proportional to the number of particles in the drive bunch and inversely proportional to the square of the transverse bunch size. The scaling laws were tested and verified in detailed simulations using parameters of existing proton accelerators, and large electric fields were achieved, reaching 1 GV/m for LHC bunches. Energy gains for test electrons beyond 6 TeV were found in this case.Comment: 9 pages, 7 figure

    Intersectionality, Resistance, and History-Making: A Conversation Between Carolyn D'Cruz, Ruth DeSouza, Samia Khatun, and Crystal McKinnon, Facilitated by Jordana Silverstein

    A good, solid, history-writing practice is one which, I think, shakes people's ideas of the world and their place in it, compelling them to imagine new social, cultural and political formations which can provide an account of life. Kimberle Crenshaw's development of the term 'intersectionality', and the ways it has been taken up by people of colour within the academy internationally, as well as by activists, provides one example of such imaginative work. Because when you spend some time in the Australian History academic scene, at conferences, in departments, talking to other academics, it's quickly noticeable that one of its key features is its hegemonic whiteness. Even in those spaces that aspire to avoid whiteness, it's inescapable, visible daily, as well as in the themes at conferences, the keynote speakers chosen, the food served, the knowledge shared. When it came time for the Australian Women's History Network conference in 2016, which carried the theme of 'Intersections in History', it felt like this could provide a way of modelling a different kind of Australian academic History space. What would a conversation look like that skipped over the presence of white Anglo Australians, I wondered? What if we just left them to the side? What if we gathered together some of the smartest, sharpest thinkers in Melbourne academia, and spoke amongst ourselves, coming up with new formations of knowledge? And so we did: Crystal, Samia, Ruth and Carolyn gathered together, I asked them some questions, and we had a conversation that, in numerous ways, challenged white hegemonies. We've recreated some of that conversation below, as a way of continuing to think together, and to find new ways of making this thinking public

    Validation and refinement of allometric equations for roots of northern hardwoods

    The allometric equations developed by Whittaker et al. (1974. Ecol. Monogr. 44: 233–252), at the Hubbard Brook Experimental Forest have been used to estimate biomass and productivity in northern hardwood forest systems for over three decades. Few other species-specific allometric estimates of belowground biomass are available because of the difficulty in collecting the data, and such equations are rarely validated. Using previously unpublished data from Whittaker’s sampling effort, we extended the equations to predict the root crown and lateral root components for the three dominant species of the northern hardwood forest: American beech (Fagus grandifolia Ehrh.), yellow birch (Betula alleghaniensis Britt), and sugar maple (Acer saccharum Marsh.). We also refined the allometric models by eliminating the use of very small trees for which the original data were unreliable. We validated these new models of the relationship of tree diameter to the mass of root crowns and lateral roots using root mass data collected from 12 northern hardwood stands of varying age in central New Hampshire. These models provide accurate estimates of lateral roots (diameter) in northern hardwood stands \u3e20 years old (mean error 24%–32%). For the younger stands that we studied, allometric equations substantially underestimated observed root biomass (mean error \u3e60%), presumably due to remnant mature root systems from harvested trees supporting young root-sprouted trees
