    Pseudospin symmetry and its approximation in real nuclei

    The origin of pseudospin symmetry and its broken in real nuclei are discussed in the relativistic mean field theory. In the exact pseudospin symmetry, even the usual intruder orbits have degenerate partners. In real nuclei, pseudospin symmetry is approximate, and the partners of the usual intruder orbits will disappear. The difference is mainly due to the pseudo spin-orbit potential and the transition between them is discussed in details. The contribution of pseudospin-orbit potential for intruder orbits is quite large, compared with that for pseudospin doublets. The disappearance of the pseudospin partner for the intruder orbit can be understood from the properties of its wave function.Comment: 10 pages, 3 figure

    Genotype-free demultiplexing of pooled single-cell RNA-seq.

    A variety of methods have been developed to demultiplex pooled samples in a single cell RNA sequencing (scRNA-seq) experiment which either require hashtag barcodes or sample genotypes prior to pooling. We introduce scSplit which utilizes genetic differences inferred from scRNA-seq data alone to demultiplex pooled samples. scSplit also enables mapping clusters to original samples. Using simulated, merged, and pooled multi-individual datasets, we show that scSplit prediction is highly concordant with demuxlet predictions and is highly consistent with the known truth in cell-hashing dataset. scSplit is ideally suited to samples without external genotype information and is available at: https://github.com/jon-xu/scSplit

    The Reproducibility of Lists of Differentially Expressed Genes in Microarray Studies

    Reproducibility is a fundamental requirement in scientific experiments and clinical contexts. Recent publications raise concerns about the reliability of microarray technology because of the apparent lack of agreement between lists of differentially expressed genes (DEGs). In this study we demonstrate that (1) such discordance may stem from ranking and selecting DEGs solely by statistical significance (P) derived from widely used simple t-tests; (2) when fold change (FC) is used as the ranking criterion, the lists become much more reproducible, especially when fewer genes are selected; and (3) the instability of short DEG lists based on P cutoffs is an expected mathematical consequence of the high variability of the t-values. We recommend the use of FC ranking plus a non-stringent P cutoff as a baseline practice in order to generate more reproducible DEG lists. The FC criterion enhances reproducibility while the P criterion balances sensitivity and specificity

    Development and Evaluation of Peanut Germplasm with Resistance to Aspergillus flavus from Core Collection

    Peanut (Arachis hypogaea L.), one of the main oil and cash crops in the world, is easily susceptible to Aspergillus flavus, resulting huge loss in its quality, so Aspergillus flavus infection greatly limits peanut production and industry in China. Therefore, it is imperative to develop new peanut germplasm with resistance to Aspergillus flavus in breeding program. The core collection is well accepted as a useful way to improve the efficiency of crop germplasm evaluation and utilization, which contains a subset of accessions from the entire collection that covers the most of available genetic information. In the present study, a total of 561 accessions of Chinese peanut core collection and 155 accessions of ICRISAT mini core collection were identified. Eight varieties with resistance to Aspergillus flavus invasion and aflatoxin production each were developed, including one (51002-6) with elite agronomic traits. The peanut germplasm with resistance to Aspergillus flavus invasion and aflatoxin production in ICRISAT mini core were more than those in Chinese peanut core collection. In addition, the percentages of accessions with resistance to Aspergillus flavus invasion in var. hypogaea, and accessions resistant to aflatoxin production in var. hirsuta were relatively high in comparison with others. Genetic diversity in the resistant peanut selections was evaluated based on morphological traits and SSR approach. ICG12625 with resistance to aflatoxin production and ICG4750 with resistance to aflatoxin invasion were evaluated by SSR, the genetic distance of them with high-yielding cultivars such as Zhonghua 5, Zhonghua 6 and Zhonghua 12 and Yuanza 9102 was larger.     A new multivariate empirical mode decomposition method for improving the performance of SSVEP-based brain–computer interface

    Objective: Accurate and efficient detection of steady-state visual evoked potentials (SSVEP) in electroencephalogram (EEG) is essential for the related brain–computer interface (BCI) applications. Approach: Although the canonical correlation analysis (CCA) has been applied extensively and successfully to SSVEP recognition, the spontaneous EEG activities and artifacts that often occur during data recording can deteriorate the recognition performance. Therefore, it is meaningful to extract a few frequency sub-bands of interest to avoid or reduce the influence of unrelated brain activity and artifacts. This paper presents an improved method to detect the frequency component associated with SSVEP using multivariate empirical mode decomposition (MEMD) and CCA (MEMD-CCA). EEG signals from nine healthy volunteers were recorded to evaluate the performance of the proposed method for SSVEP recognition. Main results: We compared our method with CCA and temporally local multivariate synchronization index (TMSI). The results suggest that the MEMD-CCA achieved significantly higher accuracy in contrast to standard CCA and TMSI. It gave the improvements of 1.34%, 3.11%, 3.33%, 10.45%, 15.78%, 18.45%, 15.00% and 14.22% on average over CCA at time windows from 0.5 s to 5 s and 0.55%, 1.56%, 7.78%, 14.67%, 13.67%, 7.33% and 7.78% over TMSI from 0.75 s to 5 s. The method outperformed the filter-based decomposition (FB), empirical mode decomposition (EMD) and wavelet decomposition (WT) based CCA for SSVEP recognition. Significance: The results demonstrate the ability of our proposed MEMD-CCA to improve the performance of SSVEP-based BCI

    Status of Muon Collider Research and Development and Future Plans

    The status of the research on muon colliders is discussed and plans are outlined for future theoretical and experimental studies. Besides continued work on the parameters of a 3-4 and 0.5 TeV center-of-mass (CoM) energy collider, many studies are now concentrating on a machine near 0.1 TeV (CoM) that could be a factory for the s-channel production of Higgs particles. We discuss the research on the various components in such muon colliders, starting from the proton accelerator needed to generate pions from a heavy-Z target and proceeding through the phase rotation and decay (πμνμ\pi \to \mu \nu_{\mu}) channel, muon cooling, acceleration, storage in a collider ring and the collider detector. We also present theoretical and experimental R & D plans for the next several years that should lead to a better understanding of the design and feasibility issues for all of the components.     Lithium, an anti-psychotic drug, greatly enhances the generation of induced pluripotent stem cells

    Somatic cells can be reprogrammed into induced pluripotent stem cells (iPSCs) by defined factors. The low efficiency of reprogramming and genomic integration of oncogenes and viral vectors limited the potential application of iPSCs. Here we report that Lithium (Li), a drug used to treat mood disorders, greatly enhances iPSC generation from both mouse embryonic fibroblast and human umbilical vein endothelial cells. Li facilitates iPSC generation with one (Oct4) or two factors (OS or OK). The effect of Li on promoting reprogramming only partially depends on its major target GSK3β. Unlike other GSK3β inhibitors, Li not only increases the expression of Nanog, but also enhances the transcriptional activity of Nanog. We also found that Li exerts its effect by promoting epigenetic modifications via downregulation of LSD1, a H3K4-specific histone demethylase. Knocking down LSD1 partially mimics Li's effect in enhancing reprogramming. Our results not only provide a straightforward method to improve the iPSC generation efficiency, but also identified a histone demethylase as a critical modulator for somatic cell reprogramming

    Type IV Pili of Acidithiobacillus ferrooxidans Are Necessary for Sliding, Twitching Motility, and Adherence

    We used conventional methods to investigate the mechanism by which Acidithiobacillus ferrooxidans colonizes a solid surface by assessing pili-mediated sliding, twitching motility, and adherence. A. ferrooxidans slided to form circular oxidized zones around each colony. This suggested that slide motility occurs through pili or flagella, though A. ferrooxidans strains ATCC 19859 and ATCC 23270 lack flagella. The results of reverse transcription-PCR demonstrated that the putative major pili gene of A. ferrooxidans strains ATCC 19859, ATCC 23270, and BY3 genes were transcribed. Culture of A. ferrooxidans between silicone gel and glass led to the production of type IV pili and the formation of rough twitching motility zones. When the bacteria were grown on lean ore cubes, pyrite was colonized readily by A. ferrooxidans and there is a correlation between pilus expression and strong attachment. However, non-pili bacteria attached minimally to the mineral surface. The results show a correlation between these functions and pilus expression

    Effect of haemoglobin concentration on the clinical outcomes in patients with acute myocardial infarction and the factors related to haemoglobin

    <p>Abstract</p> <p>Background</p> <p>The impact of haemoglobin concentrations on clinical outcomes is still a controversial issue. To determine the association between haemoglobin concentrations on admission and clinical outcomes and the related factors, this study was performed in a Chinese hospital.</p> <p>Findings</p> <p>We conducted a retrospective study on 1394 Chinese patients with acute myocardial infarction. Patients were categorized according to the haemoglobin concentration on admission, and data were evaluated to determine whether there was an association between the haemoglobin concentrations on admission and 30-day in-hospital MACEs (major cardiovascular events). Patients with hemoglobin values between 141 and 150 g/L were used as the reference, the MACEs increased as hemoglobin concentrations fell below 140 g/L or rose > 150 g/L, with an adjusted OR (odds ratio) of 5.96[95% CI (confidence interval) 2.00 to 17.68, p = 0.0013], 4.39(1.37 to 14.08, p = 0.0128), 3.99(1.46 to 10.92, p = 0.0071), 3.19(1.27 to 8.05, p = 0.0139), 2.37(0.94 to 6.01, p = 0.0687), 2.11(0.66 to 6.74, p = 0.2065), 2.01(0.60 to 6.68, p = 0.2559) in patients with haemoglobin concentrations <100 g/L, 101-110 g/L, 111-120 g/L, 121-130 g/L, 131-140 g/L, 151-160 g/L, and >160 g/L respectively. Partial correlation analysis showed that age, albumin and creatinine were significantly associated with hemoglobin concentration.</p> <p>Conclusions</p> <p>Our results demonstrated that haemoglobin concentration affected MACEs in patients with acute myocardial infarction, and that haemoglobin concentration was associated with age, albumin and creatinine.</p
