    Microrheology with optical tweezers: data analysis

    We present a data analysis procedure that provides the solution to a long-standing issue in microrheology studies, i.e. the evaluation of the fluids' linear viscoelastic properties from the analysis of a finite set of experimental data, describing (for instance) the time-dependent mean-square displacement of suspended probe particles experiencing Brownian fluctuations. We report, for the first time in the literature, the linear viscoelastic response of an optically trapped bead suspended in a Newtonian fluid, over the entire range of experimentally accessible frequencies. The general validity of the proposed method makes it transferable to the majority of microrheology and rheology techniques

    Assessing the Impact of the Concrete Canoe and Steel Bridge Competitions on Civil Engineering Technology Students

    The new accreditation criteria (TC2K) of the Technology Accreditation Commission of ABET require an assessment of Program Intended Learning Outcomes. Some of the learning outcomes required by the “a” through “k” and the Civil criteria of TC2K include leadership skills, teamwork skills, project management skills, communication skills, and design and construction skills. The American Society of Civil Engineers (ASCE) and the American Institute for Steel Construction (AISC) sponsors regional and national concrete canoe and steel bridge competitions on an annual basis. The Civil Engineering Technology (CET) students at the Rochester Institute of Technology (RIT) have participated in these competitions for the past nine years. These competitions give students a chance to apply what they have learned in the classroom. To assess the impact of the concrete canoe and steel bridge competitions on student learning and development, the authors carried out a survey of current students and graduates of the CET program at RIT who have been members of the concrete canoe and steel bridge teams. The issues addressed in our survey include the following: impact of these activities on leadership skills, communication skills, teamwork skills, design and construction skills, project management skills, confidence levels, students’ enthusiasm for the profession, and a positive image and exposure for students and CET programs. In this paper, we discuss the results of this survey and some of the challenges to student participation in these non-credit activities at RIT, and present some suggestions for enhancing the level of student participation

    Use of zinc deposited in deciduous teeth as a retrospective measurement of dietary zinc exposure during early development

    Purpose: We proposed that zinc (Zn) deposition in deciduous teeth would be a timed record of exposure to this essential micronutrient over very early life. We tested this hypothesis by gathering information on the maternal and child's diet during pregnancy and early infancy and measuring mineral deposition in the dentine at points during deciduous tooth development.Methods: We developed a short food frequency questionnaire (S-FFQ) to record consumption of food containing Zn during pregnancy and over the first year of life of the child in an Indonesian population. Zn, Sr and Ca were measured by laser ablation ICP-MS in a series of points across the developmental timeline in deciduous teeth extracted from 18 children undergoing the process as part of dental treatment whose mothers completed the SFFQ. Mothers and children were classified into either high Zn or low Zn groups according to calculated daily Zn intake.Results: The Zn/Sr ratio in dentine deposited over late pregnancy and 0–3 months post-partum was higher (p < 0.001, 2-way ANOVA; p < 0.05 by Holm-Sidak post hoc test) in the teeth of children of mothers classified as high Zn consumers (n = 10) than in children of mothers classified as low Zn consumers (n = 8).Conclusion: The S-FFQ was validated internally as adequately accurate to measure zinc intake retrospectively during pregnancy and post-partum (∼7 years prior) by virtue of the correlation with measurements of zinc in deciduous teeth. The ratio of Zn/Sr in deciduous teeth appears to be a biomarker of exposure to zinc nutrition during early development and offers promise for use as a record of prior exposure along a timeline for research studies and, potentially, to identify individuals at heightened risk of detrimental impacts of poor early life zinc nutrition on health in later life and to implement preventative interventions

    Projecting the long-term benefits of single pill combination therapy for patients with hypertension in five countries.

    Objective: To project the 10-year clinical outcomes associated with single pill combination (SPC) therapies compared with multi-pill regimens for the management of hypertension in five countries (Italy, Russia, China, South Korea and Mexico). Methods: A microsimulation model was designed to project health outcomes between 2020 and 2030 for populations with hypertension managed according to four different treatment pathways: current treatment practices (CTP), single drug with dosage titration then sequential addition of other agents (start low and go slow, SLGS), free choice combination with multiple pills (FCC) and combination therapy in the form of a single pill (SPC). Model inputs were derived from the Global Burden of Disease 2017 dataset. Simulated outcomes of mortality, chronic kidney disease (CKD), stroke, ischemic heart disease (IHD), and disability-adjusted life years (DALYs) were estimated for 1,000,000 patients on each treatment pathway. Results: SPC therapy was projected to improve clinical outcomes over SLGS, FCC and CTP in all countries. SPC reduced mortality by 5.4% in Italy, 4.9% in Russia, 4.5% in China, 2.3% in South Korea and 3.6% in Mexico versus CTP and showed greater reductions in mortality than SLGS and FCC. The projected incidence of clinical events was reduced by 11.5% in Italy, 9.2% in Russia, 8.4% in China, 4.9% in South Korea and 6.7% in Mexico for SPC versus CTP. Conclusions: Ten-year projections indicated that combination therapies (FCC and SPC) are likely to reduce the burden of hypertension compared with conventional management approaches, with SPC showing the greatest overall benefits due to improved adherence

    Maar-diatreme geometry and deposits: Subsurface blast experiments with variable explosion depth

    Basaltic maar-diatreme volcanoes, which have craters cut into preeruption landscapes (maars) underlain by downward-tapering bodies of fragmental material commonly cut by hypabyssal intrusions (diatremes), are produced by multiple subsurface phreatomagmatic explosions. Although many maar-diatremes have been studied, the link between explosion dynamics and the resulting deposit architecture is still poorly understood. Scaled experiments employed multiple buried explosions of known energies and depths within layered aggregates in order to assess the effects of explosion depth, and the morphology and compaction of the host on the distribution of host materials in resulting ejecta, the development of subcrater structures and deposits, and the relationships between them. Experimental craters were 1–2 m wide. Analysis of high-speed video shows that explosion jets had heights and shapes that were strongly influenced by scaled depth (physical depth scaled against explosion energy) and by the presence or absence of a crater. Jet properties in turn controlled the distribution of ejecta deposits outside the craters, and we infer that this is also reflected in the diverse range of deposit types at natural maars. Ejecta were dominated by material that originated above the explosion site, and the shallowest material was dispersed the farthest. Subcrater deposits illustrate progressive vertical mixing of host materials through successive explosions. We conclude that the progressive appearance of deeper-seated material stratigraphically upward in deposits of natural maars probably records the length and time scale for upward mixing through multiple explosions with ejection by shallow blasts, rather than progressive deepening of explosion sites in response to draw down of aquifers

    Chemical data quantify Deepwater Horizon hydrocarbon flow rate and environmental distribution

The definitive version was published in Proceedings of the National Academy of Sciences of the United States of America (2012), doi:10.1073/pnas.1110564109.Detailed airborne, surface, and subsurface chemical measurements, primarily obtained in May and June 2010, are used to quantify initial hydrocarbon compositions along different transport pathways – in deep subsurface plumes, in the initial surface slick, and in the atmosphere – during the Deepwater Horizon (DWH) oil spill. Atmospheric measurements are consistent with a limited area of surfacing oil, with implications for leaked hydrocarbon mass transport and oil drop size distributions. The chemical data further suggest relatively little variation in leaking hydrocarbon composition over time. While readily soluble hydrocarbons made up ~25% of the leaking mixture by mass, subsurface chemical data show these compounds made up ~69% of the deep plume mass; only ~31% of deep plume mass was initially transported in the form of trapped oil droplets. Mass flows along individual transport pathways are also derived from atmospheric and subsurface chemical data. Subsurface hydrocarbon composition, dissolved oxygen, and dispersant data are used to provide a new assessment of release of hydrocarbons from the leaking well. We use the chemical measurements to estimate that (7.8±1.9) x106 kg of hydrocarbons leaked on June 10, 2010, directly accounting for roughly three-quarters of the total leaked mass on that day. The average environmental release rate of (10.1 ± 2.0) x106 kg/day derived using atmospheric and subsurface chemical data agrees within uncertainties with the official average leak rate of (10.2 ± 1.0) x106 kg/day derived using physical and optical methods.This research was supported by the National Science Foundation through grants to D.     Transformation Optics for Plasmonics

    A new strategy to control the flow of surface plasmon polaritons at metallic surfaces is presented. It is based on the application of the concept of Transformation Optics to devise the optical parameters of the dielectric medium placed on top of the metal surface. We describe the general methodology for the design of Transformation-Optical devices for surface plasmons and analyze, for proof-of-principle purposes, three representative examples with different functionalities: a beam shifter, a cylindrical cloak and a ground-plane cloak.Comment: 15 pages, 3 figure

    RNA-Seq identifies genes whose proteins are upregulated during syncytia development in murine C2C12 myoblasts and human BeWo trophoblasts

    The fusion of villous cytotrophoblasts into the multinucleated syncytiotrophoblast is critical for the essential functions of the mammalian placenta. Using RNA-Seq gene expression, quantitative protein expression, and siRNA knockdown we identified genes and their cognate proteins which are similarly upregulated in two cellular models of mammalian syncytia development (human BeWo cytotrophoblast to syncytiotrophoblast and murine C2C12 myoblast to myotube). These include DYSF, PDE4DIP, SPIRE2, NDRG1, PLEC, GPR146, HSPB8, DHCR7, and HDAC5. These findings provide avenues for further understanding of the mechanisms underlying mammalian placental syncytiotrophoblast development
