    Identification of atropine-and P2X1 receptor antagonist-reistant, neurogenic contractions of the urinary bladder

    Acetylcholine and ATP are excitatory cotransmitters in parasympathetic nerves. We used P2X1 receptor antagonists to further characterize the purinergic component of neurotransmission in isolated detrusor muscle of guinea pig urinary bladder. In the presence of atropine (1 μm) and prazosin (100 nm), pyridoxalphosphate-6-azophenyl-2′,4′-disulfonic acid (PPADS) (0.1–100 μm) and suramin (1–300 μm) inhibited contractions evoked by 4 Hz nerve stimulation in a concentration-dependent manner (IC50 of 6.9 and 13.4 μm, respectively). Maximum inhibition was 50–60%, which was unaffected by coadministration of the ectonucleotidase inhibitor ARL67156 (6-N,N-diethyl-d-β,γ-dibromomethyleneATP) (100 μm). The remaining responses were abolished by tetrodotoxin (1 μm). PPADS and suramin also reduced contractions to exogenous ATP (300 μm) by 40–50%, but abolished those to the P2X1 agonist α,β-methyleneATP (α,β-meATP) (1 μm). The P2X1 antagonists reactive blue 2, NF279 (8,8′-[carbonylbis(imino-4,1-phenylenecarbonylimino-4,1-phenylenecarbonylimino)] bis-1,3,5-naphthalenetrisulfonic acid), MRS2159 (pyridoxal-α5-phosphate-6-phenylazo-4′-carboxylic acid) (100 μm), and NF449 [4,4′,4,4-(carbonylbis(imino-5,1,3-benzenetriylbis(carbonylimino)))tetrakis-benzene-1,3-disulfonic acid] (3 μm) abolished contractions to α,β-meATP (1 μm; n = 4–5), but only reduced contractions evoked by 4 Hz nerve stimulation by ∼40–60% (n = 4–6) and ATP by 30–60% (n = 4–7). However, prolonged exposure to α,β-meATP (50 μm) abolished contractions evoked by all three stimuli (n = 5–12). PPADS (100 μm) and suramin (300 μm) reduced the peak neurogenic contraction of the mouse urinary bladder to 30–40% of control. At the same concentrations, the P2X1 antagonists abolished the nonadrenergic, purinergic component of neurogenic contractions in the guinea pig vas deferens (n = 4–5). Thus, P2X1 receptor antagonists inhibit, but do not abolish, the noncholinergic component of neurogenic contractions of guinea pig and mouse urinary bladder, indicating a second mode of action of neuronally released ATP. This has important implications for treatment of dysfunctional urinary bladder, for which this atropine- and P2X1 antagonist-resistant site represents a novel therapeutic target

    Exploring the Link Between Self-Construal and Distress Among African American and Asian American College Students

    The authors investigated ethnicity, self-construal, and distress among African American and Asian American college students. African American students expressed more salient independent self-construals, whereas Asian American students expressed more salient interdependent self-construals. As hypothesized, among African American participants, distress was positively related to interdependent self-construal and negatively associated with independent self-construal. Contrary to prediction, the same pattern was found for Asian American participants. Multicultural clinical practice implications are presented

The global economic crisis, the economic crisis in our country need to radically change approaches to the sources and factors of economic growth of the domestic economy and to pay special attention to social factors of economic growth. Education should not be considered as area that is a burden for the budget of the company and is not related to economic development. Vocational on-the-job staff education is an area of social and economic investments that are essential for constant development of enterprises. In order to the effectiveness of vocational on-the-job staff training is high, vocational training should be a integrated continuous process consisting of several stages: setting of vocational training aims; determination of training needs; training activities; training process; test of knowledge; evaluation of the training effectiveness. In order to training effectiveness is the highest one, employers need to pay enough attention to each stage of the training process. Inattention to any stages likely would nullify all the results of this and future training processes in the organization.Рассмотрены важные этапы процесса обучения на производстве.     Investigation into the role of endogenous abscisic acid during ripening of imported avocado cv. Hass

    BACKGROUND The importance of ethylene in avocado ripening has been extensively studied. In contrast, little is known about the possible role of abscisic acid (ABA). The present work studied the effect of 1-methylcyclopropene (1-MCP) (0.3 μL L−1), e+® Ethylene Remover and the combination thereof on the quality of imported avocado cv. Hass fruit stored for 7 days at 12 °C. Ethylene production, respiration, firmness, colour, heptose (C7) sugars and ABA concentrations in mesocarp tissue were measured throughout storage. RESULTS Treatment with e+® Ethylene Remover reduced ethylene production, respiration rate and physiological ripening compared with controls. Fruit treated with 1-MCP + e+® Ethylene Remover and, to a lesser extent 1-MCP alone, had the lowest ethylene production and respiration rate and hence the best quality. Major sugars measured in mesocarp tissue were mannoheptulose and perseitol, and their content was not correlated with ripening parameters. Mesocarp ABA concentration, as determined by mass spectrometry, increased as fruit ripened and was negatively correlated with fruit firmness. CONCLUSIONS Results suggest a relationship between ABA and ethylene metabolism since blocking ethylene, and to a larger extent blocking and removing ethylene, resulted in lower ABA concentrations. Whether ABA influences avocado fruit ripening needs to be determined in future research. © 2017 Society of Chemical Industr

    The Association between Four Citation Metrics and Peer Rankings of Research Influence of Australian Researchers in Six Fields of Public Health

    Doubt about the relevance, appropriateness and transparency of peer review has promoted the use of citation metrics as a viable adjunct or alternative in the assessment of research impact. It is also commonly acknowledged that research metrics will not replace peer review unless they are shown to correspond with the assessment of peers. This paper evaluates the relationship between researchers' influence as evaluated by their peers and various citation metrics representing different aspects of research output in 6 fields of public health in Australia. For four fields, the results showed a modest positive correlation between different research metrics and peer assessments of research influence. However, for two fields, tobacco and injury, negative or no correlations were found. This suggests a peer understanding of research influence within these fields differed from visibility in the mainstream, peer-reviewed scientific literature. This research therefore recommends the use of both peer review and metrics in a combined approach in assessing research influence. Future research evaluation frameworks intent on incorporating metrics should first analyse each field closely to determine what measures of research influence are valued highly by members of that research community. This will aid the development of comprehensive and relevant frameworks with which to fairly and transparently distribute research funds or approve promotion applications

    Determinants and sources of iron intakes of australian toddlers: Findings from the SMILE cohort study

During this time a lack of key nutrients, including iron, can have long-lasting effects on motor and cognitive performance. The purpose of this cross-sectional study was to determine intake and sources of iron in a cohort of 828 toddlers (mean age; 13.1 mo) participating in the Adelaide-based Study of Mothers’ and Infants’ Life Events affecting oral health (SMILE), and to identify determinants of iron intake. At approximately 12 months of age, 3 non-consecutive days of dietary intake data were collected using a 24-h recall and 2-days food record. The Multiple Source Method was used to combine data from the 24-h recall and each day of the food record to estimate usual iron intake and descriptive statistics were used to report sources of iron. Linear regression was used to identify associations between iron intake and non-dietary determinants (maternal age, education, country of birth, BMI, socioeconomic position, parity, toddler sex) and primary milk feeding method at 12 months. The mean intake of iron was 7.0 (95% CI 6.7–7.2) mg/day and 18.2% of children had usual intakes below the estimated average requirement of 4 mg/day. The main sources of iron included infant and toddler cereals and formulas. Milk feeding method and parity were significantly associated with iron intake. Toddlers with siblings and those who received breast milk as their primary milk feed had significantly lower iron intakes than only children and those who received formula, respectively. The Australian Infant Feeding Guidelines promote the importance of iron-iron-rich complementary foods such as meat and meat alternatives. However, low intakes of this food group suggest that parents do not recognize the importance of these foods or understand the specific foods that toddlers should be eating

    Re-introduction of structurally complex wood jams promotes channel and habitat recovery from overwidening: Implications for river conservation

Large wood is a powerful geomorphic agent in rivers, providing important habitat functions for a range of aquatic organisms, but has been subject to a long history of removal. Internationally, approaches to river restoration are increasingly incorporating large wood features, but generally favour simple flow deflectors (e.g. single logs, stripped of branches and anchored in place) over more complex structures that more accurately mimic natural wood jams. This paper explores channel response to wood-based restoration of an overwidened lowland chalk stream that incorporated whole felled trees. Hydraulics, sediment, topography and vegetation data were assessed for a 3year period for two restored reaches: an upstream reach where pre-restoration baseline data were obtained, and a downstream reach restored before data collection. Where pre-restoration data were available, the introduction of wood jams generated sediment deposition within jams leading to the development of vegetated marginal ‘benches’ and bed scour in adjacent areas of flow convergence. Patterns were less clear in the downstream reach, where restoration design was less ambitious and outcomes may have been affected by subsequent restoration work upstream. The results indicate that reintroduction of large wood (whole trees), can promote channel and habitat recovery from overwidening in lowland rivers, creating important ecological benefits through the provision of structurally complex marginal habitat and associated food resources. Longer-term assessments are required to establish whether the trajectories of change are persistent. The work emphasizes the effectiveness of restoration approaches that aim to ‘work with nature’. The ambitious design, incorporating structurally complex wood jams, was also low-cost, using materials available from the river corridor (existing riparian trees). Furthermore, ecosystem engineering effects were amplified by the colonization of wood jams by aquatic vegetation. The approach should, therefore, be transferable to other lowland rivers, subject to wider catchment constraints
