    Impacts of in vivo and in vitro exposures to tamoxifen: comparative effects on human cells and marine organisms

    Tamoxifen (TAM) is a first generation-SERM administered for hormone receptor-positive (HER+) breast cancer in both pre- and post-menopausal patients and may undergo metabolic activation in organisms that share similar receptors and thus face comparable mechanisms of response. The present study aimed to assess whether environmental trace concentrations of TAM are bioavailable to the filter feeder M. galloprovincialis (100 ng L-1) and to the deposit feeder N. diversicolor (0.5, 10, 25 and 100 ng L-1) after 14 days of exposure. Behavioural impairment (burrowing kinetic), neurotoxicity (AChE activity), endocrine disruption by alkali-labile phosphate (ALP) content, oxidative stress (SOD, CAT, GPXs activities), biotransformation (GST activity), oxidative damage (LPO) and genotoxicity (DNA damage) were assessed. Moreover, this study also pertained to compare TAM cytotoxicity effects to mussels and targeted human (i.e. immortalized retinal pigment epithelium - RPE; and human transformed endothelial cells - HeLa) cell lines, in a range of concentrations from 0.5 ng L-1 to 50 μg L-1. In polychaetes N. diversicolor, TAM exerted remarkable oxidative stress and damage at the lowest concentration (0.5 ng L-1), whereas significant genotoxicity was reported at the highest exposure level (100 ng L-1). In mussels M. galloprovincialis, 100 ng L-1 TAM caused endocrine disruption in males, neurotoxicity, and an induction in GST activity and LPO byproducts in gills, corroborating in genotoxicity over the exposure days. Although cytotoxicity assays conducted with mussel haemocytes following in vivo exposure was not effective, in vitro exposure showed to be a feasible alternative, with comparable sensitivity to human cell line (HeLa).info:eu-repo/semantics/publishedVersio

    Na piscicultura, diversos produtos fitoterápicos vêm sendo usados no tratamento das parasitoses, principalmente causadas por helmintos. Mentha piperita, planta usada na medicina popular para tratar nauseas, vômitos, indigestão, desordens estomacais, cólicas mestruais e verminoses não tem sido testada em peixes, incluindo pirarucu Arapaima gigas. Porém, é importante conhecer a toxicidade de qualquer produto antes de recomendar a sua concentração terapêutica. Este ensaio realizado no Laboratório de Sanidade de Organismos Aquáticos da Embrapa Amapá, Macapá (AP), teve como objetivo determinar a concentração letal média (CL50-4h) do óleo essencial (OE) de M. piperita para pirarucus. Neste ensaio de toxicidade utilizou-se 168 alevinos de pirarucu (34,7 ± 12,4 g e 16,8 ± 2,0 cm) que após aclimatação foram distribuídos aleatoriamente em 14 tanques com volume para 80 L de água e na densidade de 12 peixes/tanque. O delineamento experimental foi inteiramente casualizado, constituído por dois grupos controles (um somente água do cultivo e outro água do cultivo contendo 80 ?L de álcool etílico absoluto) e seis concentrações (20, 40, 80, 100, 130 e 160 mg/L) de OE de M. piperita em duplicatas. As concentrações do OE foram diluídas em álcool etílico (1g:10 mL). Durante o período de 4 horas de exposição ao OE, os peixes foram mantidos sem alimentação, em sistema estático de água e a CL50-4h, foi calculada usando o método de Trimmed Spearman-Karber. Os resultados mostraram que a CL50-4h de M. piperita foi 38 mg/L, com intervalo de confiança (95%) variando de 30-40 mg/L. Portanto, o teste de toxicidade aguda indica que as concentrações clínicas para banhos terapêuticos de pirarucus com OE de M. piperita devem ser abaixo desses valores obtidos, para evitar mortalidade dos peixes, que é indesejada em qualquer piscicultura durante o tratamento antiparasitário.US100

    Advanced Control Facility for the CERN-UNICOS Framework

    CERN, during last decade, has extensively applied the CERN/UNICOS framework to large scale cryoplant control system. An increase of interested to advanced control techniques and innovative simulation environment applied to cryogenic processes has also occur. Since new control algorithm development into UNICOS framework requires significant time, a control testing platform which can be externally connected can improve and simplify the procedure of testing advanced controllers implementation. In this context, the present paper describes the development of a control testing tool at CERN, which allows rapid control strategies implementation through the Matlab/Simulink® environment, coupled with the large scale cryogenics UNICOS control system or with the CERN PROCOS simulation environment. The time delays which are inherently introduced by network links and communication protocols are analyzed and experimentally identified. Security and reliability issues are also discussed

    Palliative treatment of metastatic bone pain with radiopharmaceuticals: a perspective beyond Strontium-89 and Samarium-153

    PurposeThe present review article aims to provide an overview of the available radionuclides for palliative treatment of bone metastases beyond 89Sr and 153Sm. In addition, it aims to review and summarize the clinical outcomes associated with the palliative treatment of bone metastases using different radiopharmaceuticals.Materials and methodsA literature search was conducted on Science Direct and PubMed databases (1990 - 2015). The following search terms were combined in order to obtain relevant results: bone, metastases, palliative, care, therapy, treatment, radiotherapy, review, radiopharmaceutical, phosphorus-32, strontium-89, yttrium-90, tin-117m, samarium-153, holmium-166, thulium-170, lutetium-177, rhenium-186, rhenium-188 and radium-223. Studies were included if they provided information regarding the clinical outcomes.Results and conclusionsA comparative analysis of the measured therapeutic response of different radiopharmaceuticals, based on previously published data, suggests that there is a lack of substantial differences in palliative efficacy among radiopharmaceuticals. However, when the comparative analysis adds factors such as patients life expectancy, radionuclides physical characteristics (e.g. tissue penetration range and half-life) and health economics to guide the rational selection of a radiopharmaceutical for palliative treatment of bone metastases, 177Lu and 188Re-labeled radiopharmaceuticals appear to be the most suitable radiopharmaceuticals for treatment of small and medium/large size bone lesions, respectively

    Long-term variability of the optical spectra of NGC 4151: II. Evolution of the broad Ha and Hb emission-line profiles

    Results of the long-term (11 years, from 1996 to 2006) Hα\alpha and Hβ\beta line variations of the active galactic nucleus of NGC 4151 are presented. High quality spectra (S/N>50 and R~8A) of Hα\alpha and Hβ\beta were investigated. We analyzed line profile variations during monitoring period. Comparing the line profiles of Hα\alpha and Hβ\beta, we studied different details (bumps, absorption features) in the line profiles. The variations of the different Hα\alpha and Hβ\beta line profile segments have been investigated. Also, we analyzed the Balmer decrement for whole line and for line segments. We found that the line profiles were strongly changing during the monitoring period, showing blue and red asymmetries. This indicates a complex BLR geometry of NGC 4151 with, at least, three kinematically distinct regions: one that contributes to the blue line wing, one to the line core and one to the red line wing. Such variation can be caused by an accelerating outflow starting very close to the black hole, where the red part may come from the region {closer to the black hole than the blue part, which is coming} from the region having the highest outflow velocities. Taking into account the fact that the BLR of NGC 4151 has a complex geometry (probably affected by an outflow) and that a portion of the broad line emission seems to have not a pure photoionization origin, one can ask the question whether the study of the BLR by reverberation mapping may be valid in the case of this galaxy.Comment: 24 pages, 18 figures, accepted for publications in A&
