    Ecohydraulics exemplifies the emerging “paradigm of the interdisciplines”

    The basic premise underlying ecohydraulics is deceptively simple: create a new discipline focused on the effects of water movement in aquatic ecosystems by melding principles of aquatic ecology (including aspects of fluvial geomorphology) and engineering hydraulics. However, advancing ecohydraulics as a synthetic, organized field of study is challenging because hydraulic engineers and ecologists (1) study processes that differ substantially in spatial and/or temporal scale; (2) have very different approaches to modelling; (3) utilize different sets of mathematical formulations, concepts, and assumptions; and (4) address problems with vastly different patterns of complexity and uncertainty. The differences between engineering and ecology must be reconciled within a set of concepts and practices applicable to ecohydraulics. This reconciliation is essential if ecohydraulics is to achieve the scientific esteem of its parent disciplines. First, we review how the competing paradigms of determinism and empiricism structure engineering and ecology, respectively. We then derive two guiding principles that facilitate the integration of ecology and hydraulics, the single reference framework and the multiple reference framework guiding principles. Third, we provide illustrative examples of these principles using a simple hydraulic fish habitat analysis based on physical habitat simulation (PHABSIM) system of the instream flow incremental methodology (IFIM) and a detailed fish movement model using Eulerian–Lagrangian–Agent methods (ELAMs). Based on these examples, we develop insights and conclusions to guide further advances in ecohydraulics and, perhaps even serve as a template to aid development of other interdisciplinary fields

    A large geometric distortion in the first photointermediate of rhodopsin, determined by double-quantum solid-state NMR

    Double-quantum magic-angle-spinning NMR experiments were performed on 11,12-C-13(2)-retinylidene-rhodopsin under illumination at low temperature, in order to characterize torsional angle changes at the C11-C12 photoisomerization site. The sample was illuminated in the NMR rotor at low temperature (similar to 120 K) in order to trap the primary photointermediate, bathorhodopsin. The NMR data are consistent with a strong torsional twist of the HCCH moiety at the isomerization site. Although the HCCH torsional twist was determined to be at least 40A degrees, it was not possible to quantify it more closely. The presence of a strong twist is in agreement with previous Raman observations. The energetic implications of this geometric distortion are discussed

    Microhabitat use by the white-clawed crayfish in a Tuscan stream.

    Habitat modification, pollution, overfishing, poaching, competition from nonindigenous species, and diseases have led to the extinction in Europe of many populations of indigenous crayfish. Under the rationale that any programme of reintroduction should be preceded by a thorough understanding of habitat requirements of the species of concern, we studied the microhabitat use of an Austropotamobius pallipes population in Tuscany, central Italy. Microhabitat use was assessed for water depth, current velocity, substrate, percentages of boulders, underwater tree roots, and in-stream vegetation cover. Results show that A. pallipes’ habitat use is size-partitioned. Smaller individuals mostly occupy stream edges in shallow waters with submerged roots, whereas larger individuals use deeper waters, often associated with boulders. Crayfish spatial distribution is restricted mostly to the microhabitats characterised by extensive cover and slow current velocity. The study highlights the importance of habitat heterogeneity and cover elements for the protection of this indigenous species

    This advert makes me cry: Disclosure of emotional response to advertisement on Facebook

    As social media is transforming how consumers interact with brands and how brand-related content is consumed, this paper aims to investigate if and how Facebook users express their emotions towards advertisements of brand share on the site. Seven hundred and three comments about the Lloyds 250th Anniversary advertisement on Facebook were analysed as positive, negative or neutral attitude towards the advert. Facebook users found the advertisement emotionally appealing and voluntarily report their emotion of love, pride and in some cases anger. The presence of an iconic image like the black horse and the cover music was found to be emotionally appealing. The background music as well aroused positive emotions and engaging. This study introduces the possibility of analysing Facebook comments on brand content to understand consumers’ emotional responses and attitudes to the brand. Managers can explore these opportunities to identify what consumers find interesting in advertisements and how best to develop their creative strategies. It also offers the opportunity to allocate resources better to engage consumers with creative advertisement. Unlike interviews or surveys, this is a pioneering study on measuring emotional responses to advertisement through users’ self-report on social media

    Three new chondrosarcoma cell lines: one grade III conventional central chondrosarcoma and two dedifferentiated chondrosarcomas of bone

    BackgroundChondrosarcoma is the second most common primary sarcoma of bone. High-grade conventional chondrosarcoma and dedifferentiated chondrosarcoma have a poor outcome. In pre-clinical research aiming at the identification of novel treatment targets, the need for representative cell lines and model systems is high, but availability is scarce.MethodsWe developed and characterized three cell lines, derived from conventional grade III chondrosarcoma (L835), and dedifferentiated chondrosarcoma (L2975 and L3252) of bone. Proliferation and migration were studied and we used COBRA-FISH and array-CGH for karyotyping and genotyping. Immunohistochemistry for p16 and p53 was performed as well as TP53 and IDH mutation analysis. Cells were injected into nude mice to establish their tumorigenic potential.ResultsWe show that the three cell lines have distinct migrative properties, L2975 had the highest migration rate and showed tumorigenic potential in mice. All cell lines showed chromosomal rearrangements with complex karyotypes and genotypic aberrations were conserved throughout late passaging of the cell lines. All cell lines showed loss of CDKN2A, while TP53 was wild type for exons 5–8. L835 has an IDH1 R132C mutation, L2975 an IDH2 R172W mutation and L3252 is IDH wild type.ConclusionsBased on the stable culturing properties of these cell lines and their genotypic profile resembling the original tumors, these cell lines should provide useful functional models to further characterize chondrosarcoma and to evaluate new treatment strategies

    Bovine liver slices combined with an androgen transcriptional activation assay: an in-vitro model to study the metabolism and bioactivity of steroids

    Previously we described the properties of a rapid and robust yeast androgen bioassay for detection of androgenic anabolic compounds, validated it, and showed its added value for several practical applications. However, biotransformation of potent steroids into inactive metabolites, or vice versa, is not included in this screening assay. Within this context, animal-friendly in-vitro cellular systems resembling species-specific metabolism can be of value. We therefore investigated the metabolic capacity of precision-cut slices of bovine liver using 17β-testosterone (T) as a model compound, because this is an established standard compound for assessing the metabolic capacity of such cellular systems. However, this is the first time that slice metabolism has been combined with bioactivity measurements. Moreover, this study also involves bioactivation of inactive prohormones, for example dehydroepiandrosterone (DHEA) and esters of T, and although medium extracts are normally analyzed by HPLC, here the metabolites formed were identified with more certainty by ultra-performance liquid chromatography time-of-flight mass spectrometry (UPLC–TOFMS) with accurate mass measurement. Metabolism of T resulted mainly in the formation of the less potent phase I metabolites 4-androstene-3,17-dione (4-AD), the hydroxy-T metabolites 6α, 6β, 15β, and 16α-OH-T, and the phase II metabolite T-glucuronide. As a consequence the overall androgenic activity, as determined by the yeast androgen bioassay, decreased. In order to address the usefulness of bovine liver slices for activation of inactive steroids, liver slices were exposed to DHEA and two esters of T. This resulted in an increase of androgenic activity, because of the formation of 4-AD and T

    The diploid genome sequence of an Asian individual

    Here we present the first diploid genome sequence of an Asian individual. The genome was sequenced to 36-fold average coverage using massively parallel sequencing technology. We aligned the short reads onto the NCBI human reference genome to 99.97% coverage, and guided by the reference genome, we used uniquely mapped reads to assemble a high-quality consensus sequence for 92% of the Asian individual's genome. We identified approximately 3 million single-nucleotide polymorphisms (SNPs) inside this region, of which 13.6% were not in the dbSNP database. Genotyping analysis showed that SNP identification had high accuracy and consistency, indicating the high sequence quality of this assembly. We also carried out heterozygote phasing and haplotype prediction against HapMap CHB and JPT haplotypes (Chinese and Japanese, respectively), sequence comparison with the two available individual genomes (J. D. Watson and J. C. Venter), and structural variation identification. These variations were considered for their potential biological impact. Our sequence data and analyses demonstrate the potential usefulness of next-generation sequencing technologies for personal genomics

We combine water resources management, water quality and habitat analysis tools that were developed with the Decision Support System AQUATOOL at a basin scale. The water management model solves the allocation problem through network flow optimisation and considers the environmental flows in some river stretches. Once volumes and flows are estimated, the water quality model is applied. Furthermore, the flows are evaluated from an ecological perspective by using time series of aquatic species habitat indicators. This approach was applied in the Tormes River Water System, where agricultural demands jeopardise the environmental needs of the river ecosystem. Additionally, water quality problems in the lower part of the river result from wastewater loading and agricultural pollution. Our methodological framework can be used to define water management rules that maintain water supply, aquatic ecosystem and water quality legal standards. The integration of ecological and water management criteria in a software platform with objective criteria and heuristic optimisation procedures allows for the realistic assessment and application of environmental flows.
