    Norepinephrine Remains Increased in the Six-Minute Walking Test after Heart Transplantation

    OBJECTIVE: We sought to evaluate the neurohormonal activity in heart transplant recipients and compare it with that in heart failure patients and healthy subjects during rest and just after a 6-minute walking test. INTRODUCTION: Despite the improvements in quality of life and survival provided by heart transplantation, the neurohormonal profile is poorly described. METHODS: Twenty heart transplantation (18 men, 49±11 years and 8.5±3.3 years after transplantation), 11 heart failure (8 men, 43±10 years), and 7 healthy subjects (5 men 39±8 years) were included in this study. Blood samples were collected immediately before and during the last minute of the exercise. RESULTS: During rest, patients' norepinephrine plasma level (659±225 pg/mL) was higher in heart transplant recipients (463±167 pg/mL) and heathy subjects (512±132), p<0.05. Heart transplant recipient's norepinephrine plasma level was not different than that of healthy subjects. Just after the 6-minute walking test, the heart transplant recipient's norepinephrine plasma level (1248±692 pg/mL) was not different from that of heart failure patients (1174±653 pg/mL). Both these groups had a higher level than healthy subjects had (545±95 pg/mL), p<0.05. CONCLUSION: Neurohormonal activity remains increased after the 6-minute walking test after heart transplantation

Publisher Copyright: © 2023 The AuthorsMaraging is a high alloy steel sensitive to the precipitation of intermetallics from a supersaturated solid solution during aging heat treatments. The precipitation phenomenon is well described in the literature, which gave rise to the alloy name regarding a martensite matrix with precipitates formed by aging treatment. Many studies have been performed to characterize this material, usually subjected to many sample preparations for transmission microscopy, which is very laborious, and few EBSD analyses. Therefore, in this work, a large study was conducted using electron backscatter diffraction (EBSD) to analyze maraging microstructures through band-contrast, kernel average misorientation (KAM), grain orientation spread (GOS), and misorientation distribution. Our results show that solubilization of the additive manufactured maraging steel resulted in a more homogeneous microstructure with new angle grain boundaries. Furthermore, tempering heat treatments relieve martensite strain, increasing band contrast values. Posterior aging presented a low impact on the results of EBSD analysis.publishersversionpublishe

    The fatty acid site is coupled to functional motifs in the SARS-CoV-2 spike protein and modulates spike allosteric behaviour

    The SARS-CoV-2 spike protein is the first contact point between the SARS-CoV-2 virus and host cells and mediates membrane fusion. Recently, a fatty acid binding site was identified in the spike (Toelzer et al. Science 2020). The presence of linoleic acid at this site modulates binding of the spike to the human ACE2 receptor, stabilizing a locked conformation of the protein. Here, dynamical-nonequilibrium molecular dynamics simulations reveal that this fatty acid site is coupled to functionally relevant regions of the spike, some of them far from the fatty acid binding pocket. Removal of a ligand from the fatty acid binding site significantly affects the dynamics of distant, functionally important regions of the spike, including the receptor-binding motif, furin cleavage site and fusion-peptide-adjacent regions. Simulations of the D614G mutant show differences in behaviour between these clinical variants of the spike: the D614G mutant shows a significantly different conformational response for some structural motifs relevant for binding and fusion. The simulations identify structural networks through which changes at the fatty acid binding site are transmitted within the protein. These communication networks significantly involve positions that are prone to mutation, indicating that observed genetic variation in the spike may alter its response to linoleate binding and associated allosteric communication

    Introdução: Um estudo epidemiológico sobre causas externas, utilizando taxas de mortalidade, foi realizado no Município de São José do Rio Preto, Estado de São Paulo, Brasil. O período considerado foi o triênio 1996-1998. Objetivos: Descrever as taxas de mortalidade por causas externas no Município de São José do Rio Preto, no período de 1996-1998. Metodologia: O estudo foi baseado em dados oficiais do Sistema de Informação de Mortalidade do Ministério da Saúde, e de investigação baseada nas informações dos médicos legistas (IML).A análise dos dados foi feita através dos softwares Epinfo 6.0 e Tabwin. Resultados: Os resultados foram comparados com os valores, obtidos na literatura, de algumas cidades e estados do Brasil. No caso de São José do Rio Preto, observou-se, no peírodo, diminuição no número dos óbitos por causas externas, particularmente nas mortes devidas a acidentes de trânsito, que são mais altas em relação aos valores observados no Brasil e no Estado de São Paulo. O estudo também detectou aumento no número absoluto de homicídios, principalmente nas faixas etárias de 15 a 39 anos de idade. Conclusão: Os resultados demonstram a necessidade de iniciar programas de prevenção à mortalidade pelas citadas causas externas e monitorizar, em nível local, suas ocorrências .Introduction: An epidemiological study of the mortality rates by injuries in São José do Rio Preto, State of São Paulo, Brazil, has been carried out. The period considered is 1996-1998. Objectives: To show injuries mortality rates in São José do Rio Preto-SP in 1996-98. Methods: The study was based on official data from the Mortality Information Service of the Brazilian Ministry of Health, and in a survey based on data from medical forensic examiners (IML).Software Epinfo6.0 and Tabwin was utilized for data analises. Results:The results were compared to values obtained in the literature for the other cities of São Paulo and Brazil. In the case of São José do Rio Preto, was observed a decrease in the number of deaths associated with external causes in the period, but particularly in deaths due to traffic accidents with motor vehicles an increase in cases of pedestrians killed by hit-and-run drivers, that is much larger than the corresponding values for Brazil and for the State of São Paulo. The study showed increase in rates of homicides, mainly in the 15 to 39 years age group . Conclusion: This result suggests that is important to start programs to reduce the number of deaths by injuries, as well as the necessity of monitoring locally all these data

    O presente trabalho é um estudo preliminar que discute a possibilidade do Centro de Educação Infantil do município de Vitória/ES ser um campo de atuação da engenharia de produção, uma vez que apresenta aproximações deste espaço com os espaços culturais, por meio de um questionário semiestruturado aplicado aos professores e aos pedagogos. Os resultados indicam que a comunidade escolar não tem hábito de ir há museus, livrarias, shows ou teatros, logo é no Centro de Educação Infantil que a comunidade escolar tem acesso às atividades culturais

    Generation and characterization of a recombinant chimeric protein (rCpLi) consisting of B-cell epitopes of a dermonecrotic protein from Loxosceles intermedia spider venom

    AbstractA chimeric protein was constructed expressing three epitopes of LiD1, a dermonecrotic toxin from the venom of Loxosceles intermedia spider. This species is responsible for a large number of accidents involving spiders in Brazil. We demonstrated that the chimeric protein (rCpLi) generated is atoxic and that antibodies previously developed in rabbits against synthetic epitopes reactive with rCpLi in ELISA and immunoblot assays. The antibody response in rabbits against the rCpLi was evaluated by ELISA and we have detected an antibody response in all immunized animals. Overlapping peptides covering the amino acid sequence of the rCpLi were synthesized on a cellulose membrane, and their recognition by rabbit anti-rCpLi serum assessed. Three different antigenic regions were identified. The percentage of inhibition of the dermonecrotic, hemorrhagic and edematogenic activities caused by the recombinant protein LiD1r in naïve rabbits was assessed by pre-incubation with anti-rCpLi antibodies. Anti-rCpLi induced good dermonecrotic and hemorrhagic protection. The levels of protection were similar to the antiboides anti-LiD1r. In summary, we have developed a polyepitope recombinant chimeric protein capable of inducing multiple responses of neutralizing antibodies in a rabbit model. This engineered protein may be a promising candidate for therapeutic serum development or vaccination

    Crescimento inicial de espécies vegetais na presença dos herbicidas imazapyr + imazapic em água

    The objective of this research was to evaluate the initial growth of different plant species in order toselect those that can be used as bioindicators of herbicides imazapyr + imazapic in water. The species evaluated were: radish, tomato, cucumber, sorghum, common beans, and rice (Cypress CL, IRGA 422 CL and IRGA 417). The experiment was accomplished using the floating system for seedling production. The following herbicide concentrations were tested: control, 1, 10, 100, and 1,000 g/L of BAS 714 GD (imazapyr + imazapic). The parametersevaluated included dry weight biomass, root length and plant height at seven and 14 days. All the species and rice cultivars tested showed injured symptoms due to the herbicide residue in water. The best species to detect imazapyr + imazapic residue in water were rice, cvs. IRGA 417 and IRGA 422 CL, radish, cucumber, tomato and sorghum. These species are sensitive to the herbicides even at the minimum tested concentrantions specially at14 DAS.O objetivo deste trabalho foi avaliar o crescimento inicial de espécies vegetais visando selecionar aquelas mais adequadas para uso como indicadoras da presença dos herbicidas imazapyr + imazapic, em água. As espécies avaliadas foram rabanete, tomate, pepino, sorgo, feijão e as cultivares de arroz Cypress CL, IRGA 422 CL e IRGA 417. As concentrações de herbicidas foram zero, 1, 10, 100 e 1.000μg/L da formulação WG do herbicida codificado como BAS 714 (imazapyr + imazapic), utilizando o sistema de produção de mudas “floating”. As avaliações foram realizadas aos sete e 14 dias após a semeadura (DAS), sendo determinados a fitomassa seca total e o comprimento do sistema aéreo e da raiz principal das plântulas. Todas as espécies testadas têm o desenvolvimento inicial de plântulas afetado pelo herbicida BAS 714. As melhores bioindicadoras da presença do herbicida em solução são as cultivares de arroz IRGA 417 e IRGA 422 CL, rabanete, tomate, pepino e sorgo, por apresentarem suscetibilidade a partir das concentrações mínimas avaliadas. As variáveis avaliadas (produção de fitomassa seca,comprimento do sistema aéreo e comprimento da raiz principal) são adequadas para a avaliação da presença do herbicida BAS 714 em água. Aos 14 DAS, as plantas expressam de forma mais evidente os sintomas do herbicida

    Atividade do peptídeo antimicrobiano P34 contra o alfaherpesvírus bovino tipo 1

    Previous studies have demonstrated the antimicrobial activity of the peptide P34. In this study, the antiviral potential of P34 and the in vitro mechanism of action were investigated against bovine alphaherpesvirus type 1 (BoHV1). P34 exhibited low toxicity, a high selectivity index (22.9) and a percentage of inhibition of up to 100% in MDBK cells. Results from antiviral assays indicated that P34 did not interact with cell receptors, but it was able to inhibit the viral penetration immediately after pre-adsorption. In addition, BoHV1 growth curve in MDBK cells in the presence of P34 revealed a significant reduction in virus titer only 8h post-infection, also suggesting an important role at late stages of the replicative cycle. Virucidal effect was observed only in cytotoxic concentrations of the peptide. These findings showed that the antimicrobial peptide P34 may be considered as a potential novel inhibitor of in vitro herpesviruses and must encourage further investigation of its antiherpetic activity in animal models as well as against a wide spectrum of viruses.A atividade antimicrobiana do peptídeo P34 já foi previamente demonstrada. Neste estudo, o potencial antiviral do P34 e o mecanismo de ação in vitro contra o alfaherpesvírus bovino tipo 1 (BoHV1) foram investigados. O P34 exibiu baixa toxicidade, alto índice de seletividade (22.9) e percentagem de inibição viral de até 100% em células MDBK. Os resultados dos ensaios antivirais indicaram que não interage com receptores celulares, mas é capaz de inibir a penetração viral, imediatamente após a pré-adsorção. Além disso, a curva de crescimento do BoHV1 em células MDBK na presença do P34 revelou uma significativa redução no título somente após 8h de infecção, sugerindo também uma importante atividade do peptídeo nas fases finais do ciclo replicativo. Efeito virucida frente / BoHV1 foi observado apenas em concentrações citotóxicas do peptídeo. Os dados obtidos indicam que o peptídeo antimicrobiano P34 pode ser considerado um potencial composto inibidor de herpesvírus, in vitro, e estimulam posteriores investigações sobre sua atividade anti-herpética em modelos animais, bem como contra outros vírus
