    MULTI-SCALE ASSESSMENT OF GROUNDWATER VULNERABILITY TO POLLUTION: STUDY CASES FROM CAMPANIA REGION (SOUTHERN ITALY)

    In the Campania region (southern Italy), assessment of groundwater vulnerability is an important factor to be tackled for a proper management of risk to aquifer pollution, which is fostered by both high dependence of aqueduct systems on groundwater resources and the widespread agricultural and urbanized land uses of the territory. The different physiographic, geological and hydrogeological conditions of the region, coupled with high anthropic pressure, make such assessment complex to be dealt with, but at the same time mandatory to be accomplished. The proposed study has been developed into the framework of the “Campania Trasparente” project, which has been funded by the regional government to assess environmental factors controlling the quality of agricultural and livestock food productions. Specifically, it belongs to the research topic regarding the assessment of groundwater quality, with a special focus on the assessment of groundwater vulnerability. In this research a multi-scale approach for the assessment of groundwater vulnerability is carried out in order to propose suitable methods depending on extension of territory to be studied and related types and spatial density of available data. Scales considered were a) regional, including the whole Campania region; b) intermediate, identified with that of a single representative aquifer; c) site-specific, or local, related to a portion of aquifer for which a high spatial density of data is available.The applied methods were chosen among the many known in literature and adapted to the specific study cases. At the regional scale, the parametric SINTACS method (CIVITA & DE MAIO, 2000) has been applied to the whole region and adapted to types and spatial density of available data. At the intermediate scale, or aquifer scale, the Mt. Terminio karst aquifer was chosen as representative for the application of different parametric methods, also specifically designed for karst aquifers. At the site-specific scale, a representative sector of a shallow alluvial aquifer, located in the adjoining Casalnuovo di Napoli-Volla municipalities, in the Campania plain, at the eastern border of the city of Naples, has been studied by numerical modeling for the estimation of travel time of nitrate pollutant through the vadose zone. The obtained results can be conceived as useful for supporting a proper territorial planning aimed at the management of risk to pollution of groundwater resources

    hTERT Transduction Extends the Lifespan of Primary Pediatric Low-Grade Glioma Cells While Preserving the Biological Response to NGF

    The neurotrophin nerve growth factor (NGF) modulates the growth of human gliomas and is able to induce cell differentiation through the engagement of tropomyosin receptor kinase A (TrkA) receptor, although the role played in controlling glioma survival has proved controversial. Unfortunately, the slow growth rate of low-grade gliomas (LGG) has made it difficult to investigate NGF effects on these tumors in preclinical models. In fact, patient-derived low-grade human astrocytoma cells duplicate only a limited number of times in culture before undergoing senescence. Nevertheless, replicative senescence can be counteracted by overexpression of hTERT, the catalytic subunit of telomerase, which potentially increases the proliferative potential of human cells without inducing cancer-associated changes. We have extended, by hTERT transduction, the proliferative in vitro potential of a human LGG cell line derived from a pediatric pilocytic astrocytoma (PA) surgical sample. Remarkably, the hTERT-transduced LGG cells showed a behavior similar to that of the parental line in terms of biological responses to NGF treatment, including molecular events associated with induction of NGF-related differentiation. Therefore, transduction of LGG cells with hTERT can provide a valid approach to increase the in vitro life-span of patient-derived astrocytoma primary cultures, characterized by a finite proliferative potential

    Experimental study for evaluation of a suitable ground displacement monitoring system: Pilot hole Campi Flegrei Deep Drilling Project case

    The paper presents an experimental study carried out in 2012 during the drilling activity for a pilot hole performed in the framework of the Campi Flegrei Deep Drilling Project. A monitoring network has been installed to test and choose a suitable ground deformation system for the subsequent deep drilling of about 3.5 km in the Campi Flegrei Caldera (Italy). We describe the seismic network installed to characterize the structure of the pilot hole area and the ground deformation monitoring system chosen for the small drilling area.     Effectiveness and safety of secukinumab in Italian patients with psoriasis: an 84 week, multicenter, retrospective real-world study

    Background: Long term data on the real-life use of secukinumab are scant. The aim of this study was to investigate the real-life effectiveness, safety and treatment persistence of secukinumab in patients with moderate-to-severe psoriasis. Research design and methods: This 84-week, multicenter (n = 7) retrospective study analyzed data from patients who initiated and received at least 6 months of secukinumab treatment between June 2016 and June 2018 in the Campania region of Italy. Patient demographic and treatment characteristics, duration of treatment and reasons for discontinuation as well as Psoriasis Area and Severity Index (PASI), Body Surface Area (BSA), and Dermatology Life Quality Index (DLQI) scores were assessed. Results: 324 patients (63% male, mean age 50.2 years) were enrolled and received a mean 11.7 months of secukinumab treatment. Overall, 9.5% discontinued secukinumab, including 5.2% who discontinued due to secondary inefficacy and 1.8% due to adverse events. PASI, BSA and DLQI scores were significantly improved from baseline at every follow-up visit (p < 0.001) and mean PASI decreased from 15.3 ± 6.3 at baseline to 0.5 ± 1.0 at week 84. Secukinumab had comparable effectiveness in biologic naïve and non-naïve patients. Conclusions: This study confirmed the effectiveness and safety of secukinumab in real-world patients with psoriasis

    LIFE Monza: comparison between ante and post-operam noise and air quality monitoring activities in a Noise Low Emission Zone

    AbstractLIFE MONZA project (Methodologies fOr Noise low emission Zones introduction And management) aims at defining an easy-replicable method for the identification and management of theNoise Low Emission Zones(Noise LEZ), urban areas subject to traffic restrictions, usually introduced in order to ensure compliance with the air pollutants limit values, prescribed by the European Directive on ambient air quality 2008/50/EC, whose impacts and potential benefits regarding noise issues have been taken into account, tested and analysed in a pilot area of the city of Monza, located in North Italy. Noise LEZ has been established in Libertà district, introducing infrastructural interventions carried out by the municipality (top-down actions) and encouraging an active involvement of the citizens, in the definition of a more sustainable lifestyle (bottom-up actions). The analysis of potential effects on noise reduction due to the Noise LEZ can contribute to the implementation of the EU Directive 2002/49/EC, related to the assessment and management of environmental noise (Environmental Noise Directive – END), which introduces noise action plans, designed to manage noise issues and their effects, suggesting the adoption of urban and mobility planning. Noise and air quality monitoring activities have been carried out in pilot area inanteandpost-operamconditions. The monitoring methods, the measurement techniques, the analysis procedures, able to describe the effects due to Noise LEZ establishment, for both the main environmental issues are reported in this paper, as proposals to be applied in other different contexts. Results of monitoring activities highlight a reduction of noise, in term of sound pressure levels, betweenanteandpost-operam, during the day and particularly during the night period, and it is essentially due to the interventions realised. The effect of the Noise LEZ on air pollution seems to be negligible for combustion related pollutant and carbon fractions of PM, due both to the moderate spatial effects of the measures undertaken and confounding factors due to concomitant emission sources and meteorology

    The double-sided of human leukocyte antigen-G molecules in type 1 autoimmune hepatitis

    The immunomodulatory effects of HLA-G expression and its role in cancers, human liver infections and liver transplantation are well documented, but so far, there are only a few reports addressing autoimmune liver diseases, particularly autoimmune hepatitis (AIH). Method and materialsWe analyzed the genetic and phenotypic characteristics of HLA-G in 205 type 1 AIH patients (AIH-1) and a population of 210 healthy controls from Sardinia (Italy). ResultsAnalysis of the HLA-G locus showed no substantial differences in allele frequencies between patients and the healthy control population. The HLA-G UTR-1 haplotype was the most prevalent in both AIH-1 patients and controls (40.24% and 34.29%). Strong linkage was found between the HLA-G UTR-1 haplotype and HLA-DRB1*03:01 in AIH-1 patients but not controls (D' = 0.92 vs D' = 0.50 respectively; P = 1.3x10(-8)). Soluble HLA-G (sHLA-G) levels were significantly lower in AIH-1 patients compared to controls [13.9 (11.6 - 17.4) U/mL vs 21.3 (16.5 - 27.8) U/mL; P = 0.011]. Twenty-four patients with mild or moderate inflammatory involvement, as assessed from liver biopsy, showed much higher sHLA-G levels compared to the 28 patients with severe liver inflammation [33.5 (23.6 - 44.8) U/mL vs 8.8 (6.1 - 14.5) U/mL; P = 0.003]. Finally, immunohistochemistry analysis of 52 liver biopsies from AIH-1 patients did not show expression of HLA-G molecules in the liver parenchyma. However, a percentage of 69.2% (36/52) revealed widespread expression of HLA-G both in the cytoplasm and the membrane of plasma cells labeled with anti-HLA-G monoclonal antibodies. ConclusionThis study highlights the positive immunomodulatory effect of HLA-G molecules on the clinical course of AIH-1 and how this improvement closely correlates with plasma levels of sHLA-G. However, our results open the debate on the ambiguous role of HLA-G molecules expressed by plasma cells, which are pathognomonic features of AIH-1

    Redshift identification of X-ray-selected active galactic nuclei in the J1030 field: searching for large-scale structures and high-redshift sources

    We publicly release the spectroscopic and photometric redshift catalog of the sources detected with Chandra in the field of the z = 6.3 quasar SDSS J1030+0525. This is currently the fifth-deepest extragalactic X-ray field, and reaches a 0.5 2 keV flux limit of f0:5-2 = 6 × 10-17 erg s-1 cm-2. Using two independent methods, we measure a photometric redshift for 243 objects, while 123 (51%) sources also have a spectroscopic redshift, 110 of which come from an INAF-Large Binocular Telescope (LBT) Strategic Program.We use the spectroscopic redshifts to determine the quality of the photometric ones, and find it to be in agreement with that of other X-ray surveys which used a similar number of photometric data points. In particular, we measure a sample normalized median absolute deviation of NMAD = 1.48 × median(||zphot - zspec||/(1 + zspec)) = 0.065. We use these new spectroscopic and photometric redshifts to study the properties of the Chandra J1030 field.We observe several peaks in our spectroscopic redshift distribution between z = 0.15 and z = 1.5, and find that the sources in each peak are often distributed across the whole Chandra field of view. This confirms that X-ray-selected AGNs can efficiently track large-scale structures over physical scales of several megaparsecs. Finally, we computed the Chandra J1030 z > 3 number counts: while the spectroscopic completeness of our sample is limited at high redshift, our results point towards a potential source excess at z ≥ 4, which we plan to either confirm or reject in the near future with dedicated spectroscopic campaigns

    Population- and individual-specific regulatory variation in Sardinia

    Genetic studies of complex traits have mainly identified associations with noncoding variants. To further determine the contribution of regulatory variation, we combined whole-genome and transcriptome data for 624 individuals from Sardinia to identify common and rare variants that influence gene expression and splicing. We identified 21,183 expression quantitative trait loci (eQTLs) and 6,768 splicing quantitative trait loci (sQTLs), including 619 new QTLs. We identified high-frequency QTLs and found evidence of selection near genes involved in malarial resistance and increased multiple sclerosis risk, reflecting the epidemiological history of Sardinia. Using family relationships, we identified 809 segregating expression outliers (median z score of 2.97), averaging 13.3 genes per individual. Outlier genes were enriched for proximal rare variants, providing a new approach to study large-effect regulatory variants and their relevance to traits.     Seasonal and Event-Based Hydrological and Slope Stability Modeling of Pyroclastic Fall Deposits Covering Slopes in Campania (Southern Italy)

    The pyroclastic fall deposits mantling mountain slopes in the Campania region (Southern Italy) represent one of the most studied geomorphological frameworks of the world regarding rainfall-induced debris flows threating urban areas. The proposed study focused on advancing knowledge about the hydrological response of pyroclastic fall coverings from the seasonal to event-based time scales, leading to the initiation of slope instability. The study was based on two consequential tasks. The first was the analysis of a six-year monitoring of soil pressure head carried out in a sample area of the Sarno Mountains, located above a debris flow initiation zone. The second was based on coupled hydrological and slope stability modeling performed on the physical models of slopes, which were reconstructed by empirical correlations between the slope angle, total thickness, and stratigraphic settings of pyroclastic fall deposits mantling slopes. The results obtained were: (a) The understanding of a soil pressure head regime of the volcaniclastic soil mantle, always ranging in unsaturated conditions and characterized by a strong seasonal variability depending on precipitation patterns and the life cycle of deciduous chestnut forest; and (b) the reconstruction through a deterministic approach of seasonal intensity&ndash;duration rainfall thresholds related to different morphological conditions
