Effect of ß-cyclodextrin on phospholipids and cholesterol of the milk fat globule membrane

    Get PDF
The aim of this study was to evaluate the effect of ß-cyclodextrin (ß-CD) on phospholipids and cholesterol in raw pasteurized milk with the purpuse on removal cholesterol from the milk fat. Total phospholipids decrease slightly in ß-CD treated milk but were not signinificantly affected by the effect of the ß-CD. ß-CD is an effective oligosacharide for cholesterol removal from milk with more than 97% of reduction and does not affect significantly the individual phospholipids (phosphatidylcholine, phosphatidylethanolamine, sphingomyelin, phosphatidylinositol and phosphatidylserina) of the milk fat globule membrane. The authors thanks to the Ministry of Economy and Competitiveness from Spain (grant number AGL-2011-26713).Peer Reviewe

Environmental enrichment results in both brain connectivity efficiency and selective improvement in different behavioral tasks

Exposure to environmental enrichment (EE) has been a useful model for studying the effects of experience on brain plasticity, but to date, few is known about the impact of this condition on the brain functional networks that probably underlies the multiple behavioral improvements. Hence, we assessed the effect of an EE protocol in adult Wistar rats on the performance in several behavioral tasks testing different domains (Open field (OP): locomotor activity; Elevated-zero maze (EZM): anxiety-related behaviors; 5-choice serial reaction time task (5-CSRTT): attentional processes; 4-arm radial water maze (4-RAWM): spatial memory) in order to check its effectiveness in a wide range of functions. After this, we analyzed the functional brain connectivity underlying each experimental condition through cytochrome C oxidase (COx) histochemistry. Our EE protocol reduced both locomotor activity in the OP and anxiety-related behaviors in the EZM. On the other hand, enriched rats showed more accuracy in the 4-RAWM, whereas 5-CSRTT performance was not significantly ameliorated by EE condition. In relation to COx functional connectivity, we found that EE reduced the number of strong positive correlations both in basal and training conditions, suggesting a modulating effect on specific brain connections. Our results suggest that EE seems to have a selective effect on specific brain regions, such as prefrontal cortex and hippocampus, leading to a more efficient brain connectivity.Universidad de Málaga. Real-time detection of riboflavin production by Lactobacillus plantarum strains and tracking of their gastrointestinal survival and functionality in vitro and in vivo using mCherry labeling

    Get PDF
Some strains of lactic acid bacteria (LAB) produce riboflavin, a water-soluble vitamin of the B complex, essential for human beings. Here, we have evaluated riboflavin (B2 vitamin) production by five Lactobacillus plantarum strains isolated from chicha, a traditional maize-based fermented alcoholic beverage from north-western Argentina and their isogenic riboflavin-overproducing derivatives previously selected using roseoflavin. A direct fluorescence spectroscopic detection method to quantify riboflavin production in bacterial culture supernatants has been tested. Comparison of the efficiency for riboflavin fluorescence quantification with and without prior HPLC fractionation showed that the developed method is a rapid and easy test for selection of B2 vitamin-producing strains. In addition, it can be used for quantitative detection of the vitamin production in real time during bacterial growth. On the basis of this and previous analyses, the L. plantarum M5MA1-B2 riboflavin overproducer was selected for in vitro and in vivo studies after being fluorescently labeled by transfer of the pRCR12 plasmid, which encodes the mCherry protein. The labeling did not affect negatively the growth, the riboflavin production nor the adhesion of the strain to Caco-2 cells. Thus, L. plantarum M5MA1-B2[pRCR12] was evaluated for its survival under digestive tract stresses in the presence of microbiota in the dynamic multistage BFBL gut model and in a murine model. After exposure to both models, M5MA1-B2[pRCR12] could be recovered and detected by the pink color of the colonies. The results indicated a satisfactory resistance of the strain to gastric and intestinal stress conditions but a low colonization capability observed both in vitro and in vivo. Overall, L. plantarum M5MA1-B2 could be proposed as a probiotic strain for the development of functional foods.Fil: Mohedano, Mari Luz. Consejo Superior de Investigaciones Científicas. Measurement of prethrombotic markers in the assessment of acquired hypercoagulable states

    Get PDF
Hypercoagulable states can be detected by measuring activation peptides, enzyme-inhibitor complexes, and fibrin/fibrinogen degradation products, which are markers of hemostatic activation. A series of these prethrombotic markers has been evaluated in the elderly, pregnancy, diabetes and acute myocardial infarction patients (n=30 in each group) as well as in hematologic malignancies (n=42). The parameters assayed were: prothrombin fragment 1+2 (F1+2), thrombin-antithrombin III complexes (TAT), fibrinopeptide A (FPA), plasmin-alpha2 antiplasmin complexes (PAP) and D-Dimer. Results were compared with those obtained in a group of 30 healthy subjects. We found a significant increase of F1+2, TAT and FPA in elderly (p<0.05), acute myocardial infarction (AMI) (p<0.01), hematologic malignancies (p<0.01), and pregnancy (p<0.0001), indicating a marked clotting activation. Diabetic patients under strict metabolic control only presented a moderate increase of TAT (p<0.05), suggesting a slight activation. We also observed a highly significant elevation of PAP and D-Dimer in elderly (p<0.001), AMI (p<0.0001), and malignancy (p<0.0001), indicating an activation of the fibrinolytic system. The combination of selected fibrinolytic and coagulation measurements is useful for the detection of a hypercoagulable state in conditions characterized by a risk of thrombosis

Adding Semantics to Enrich Public Transport and Accessibility Data from the Web

    Get PDF
Web technologies and open data practices have now begun to promote new issues and services addressed to both final and specialized users. The smart cities initiative has also introduced new trends and ideas to offer to the public, one of which is the challenge of a more inclusive society that will provide the same opportunities for all. One of the major areas that could benefit from these new initiatives is public transport by, for example, providing open and accessible datasets, which include information by and about people with special needs. In this sense, the Google Transit Feed Specification (GTFS) defines a format to describe public transportation and associated geographic information. It includes details regarding accessibility and what people with special needs might require to get around using public transport. We are, however, of the opinion that this specification has a low granularity and is not sufficient, since it only takes into account only mobility needs. As suggestions for improvement, we propose to enrich GTFS data by combining public transport data from multiple Web sources with semantic metadata techniques. Those data are stored in a public semantic dataset. To define this dataset, we propose a systematic method to extract data from different sources and integrate them. This method is applied to obtain data about the metro system from the website of Metro Madrid and GTFS. Relevant SPARQL queries and two applications are developed to evaluate the usefulness of the dataset obtained

Consuming Web Data in a Guiding App for Public Bus Users

    Get PDF
The complexity of urban public bus networks in big cities makes their use very difficult. This paper presents Notify.me, a set of pervasive services for mobility that employs open data from the public bus network in Madrid. Our solution provides both a guiding service to assist users travelling by bus and a notifying service (visual, acoustical and sensorial) that informs them when a relevant point on their route has been reached (transfer or destination). Notify.me needs a starting point, which can be the user's current location, a destination and the preferences regarding the best route for the user. Notify.me requests a route from the Madrid public bus company via SOAP Web services. The back-end responds with the calculated route, the user's route, which includes the bus lines, the transfers and the pedestrian routes needed to reach the destination. Finally, an empirical evaluation of the experiences of users who employed Notify.me is presented

Clotting activation and impairment of fibrinolysis in malignancy

    Get PDF
Different coagulation and fibrinolysis parameters were investigated in 149 patients with metastatic and non-metastatic tumours and results were compared with those obtained in a healthy population. Results showed a significant increase of thrombin-antithrombin complexes, fibrinopeptide A (FPA) and fibrin monomers in the group of patients (p less than 0.001). There was also a significant prolongation of euglobulin lysis time (p less than 0.005) and an increase of plasminogen activator inhibitor activity (p less than 0.0001), fibrinogen degradation products (p less than 0.001), and D-dimer (p less than 0.05) in the group of patients as compared to controls; FPA levels were also increased in patients with metastases (p less than 0.005). This study demonstrates clotting activation, at the level of fibrinogen to fibrin conversion, and impairment of fibrinolysis in patients with malignancy
