    Variation and genetic structure of Melipona quadrifasciata Lepeletier (Hymenoptera, Apidae) populations based on ISSR pattern

    For a study of diversity and genetic structuring in Melipona quadrifasciata, 61 colonies were collected in eight locations in the state of Minas Gerais, Brazil. By means of PCR analysis, 119 ISSR bands were obtained, 80 (68%) being polymorphic. He and H B were 0.20 and 0.16, respectively. Two large groups were obtained by the UPGMA method, one formed by individuals from Januária, Urucuia, Rio Vermelho and Caeté and the other by individuals from São João Del Rei, Barbacena, Ressaquinha and Cristiano Otoni. The Φst and θB values were 0.65 and 0.58, respectively, thereby indicating high population structuring. UPGMA grouping did not reveal genetic structuring of M. quadrifasciata in function of the tergite stripe pattern. The significant correlation between dissimilarity values and geographic distances (r = 0.3998; p < 0.05) implies possible geographic isolation. The genetic differentiation in population grouping was probably the result of an interruption in gene flow, brought about by geographic barriers between mutually close geographical locations. Our results also demonstrate the potential of ISSR markers in the study of Melipona quadrifasciata population structuring, possibly applicable to the studies of other bee species

    Evolutionary and Experimental Assessment of Novel Markers for Detection of Xanthomonas euvesicatoria in Plant Samples

    BACKGROUND: Bacterial spot-causing xanthomonads (BSX) are quarantine phytopathogenic bacteria responsible for heavy losses in tomato and pepper production. Despite the research on improved plant spraying methods and resistant cultivars, the use of healthy plant material is still considered as the most effective bacterial spot control measure. Therefore, rapid and efficient detection methods are crucial for an early detection of these phytopathogens. METHODOLOGY: In this work, we selected and validated novel DNA markers for reliable detection of the BSX Xanthomonas euvesicatoria (Xeu). Xeu-specific DNA regions were selected using two online applications, CUPID and Insignia. Furthermore, to facilitate the selection of putative DNA markers, a customized C program was designed to retrieve the regions outputted by both databases. The in silico validation was further extended in order to provide an insight on the origin of these Xeu-specific regions by assessing chromosomal location, GC content, codon usage and synteny analyses. Primer-pairs were designed for amplification of those regions and the PCR validation assays showed that most primers allowed for positive amplification with different Xeu strains. The obtained amplicons were labeled and used as probes in dot blot assays, which allowed testing the probes against a collection of 12 non-BSX Xanthomonas and 23 other phytopathogenic bacteria. These assays confirmed the specificity of the selected DNA markers. Finally, we designed and tested a duplex PCR assay and an inverted dot blot platform for culture-independent detection of Xeu in infected plants. SIGNIFICANCE: This study details a selection strategy able to provide a large number of Xeu-specific DNA markers. As demonstrated, the selected markers can detect Xeu in infected plants both by PCR and by hybridization-based assays coupled with automatic data analysis.     Vitimização por homicídios segundo características de raça no Brasil

    OBJETIVO: Descrever a tendência temporal da mortalidade por homicídio no Brasil. MÉTODOS: Estudo de série temporal dos homicídios no Brasil de 2000 a 2009. As variáveis explicativas foram raça/cor, sexo e escolaridade. Os óbitos foram provenientes do Sistema de Informações de Mortalidade. A análise de tendência foi realizada por meio de regressão polinomial para séries históricas (p < 0,05; intervalo de 95% de confiança). RESULTADOS: A população negra representou 69% das vítimas de homicídios em 2009. O número de homicídios aumentou entre a população negra e diminuiu entre a branca, com tendência de crescimento da taxa nos negros e de redução nos brancos no período. As taxas aumentaram nos grupos de maior e menor escolaridade entre negros, enquanto, entre brancos, reduziram para os de menor nível escolar e mantiveram-se estáveis no grupo com maior nível de escolaridade. Em 2009 negros tiveram maior risco de morte por homicídios do que a população branca, independentemente do nível de escolaridade. Entre 2004 e 2009, as taxas de homicídios na população branca diminuíram e aumentaram na negra. CONCLUSÕES: O risco relativo de homicídios cresce na população negra, sugerindo o aumento das desigualdades. A repercussão das medidas antiarmas no Brasil, implantada em 2004, foi positiva na população branca e discreta na população negra. Raça/cor pode predizer a ocorrência de homicídio.OBJETIVO: Describir la tendencia temporal de la mortalidad por homicidio en Brasil. METHODS: A series of homicides in Brazil from 2000 to 2009 were studied. The explanatory variables were race/skin color, gender and education. The death statistics were obtained from the Mortality Information System. A trend analysis was performed by means of a polynomial regression for a historic time series (p < 0.05, 95% confidence interval). RESULTS: The black population represented 69% of the homicide victims in 2009. The homicide rate increased in the black population, while it decreased in the white population in the period studied. The homicide rate increased in groups with both higher and lower education among blacks; among whites, the rate decreased for those with the lowest level of schooling and remained stable in the group with higher educational levels. In 2009, blacks had a higher risk of death than whites from homicide, regardless of education level. Between 2004 and 2009, the homicide rate decreased in the white population, while it increased in the black population. CONCLUSIONS: The relative risk of falling victim to homicide increased in the black population, suggesting an increase in inequality. The effect of the anti-gun measures implemented in Brazil in 2004 was positive in the white population and less pronounced in the black population. Overall, race/skin color predicted the occurrence of homicide

    Expanding the knowledge about Leishmania species in wild mammals and dogs in the Brazilian savannah

    Background: Wild, synanthropic and domestic mammals act as hosts and/or reservoirs of several Leishmania spp. Studies on possible reservoirs of Leishmania in different areas are fundamental to understand host-parasite interactions and develop strategies for the surveillance and control of leishmaniasis. In the present study, we evaluated the Leishmania spp. occurrence in mammals in two conservation units and their surroundings in Brasília, Federal District (FD), Brazil. Methods: Small mammals were captured in Brasília National Park (BNP) and Contagem Biological Reserve (CBR) and dogs were sampled in residential areas in their vicinity. Skin and blood samples were evaluated by PCR using different molecular markers (D7 24Sα rRNA and rDNA ITS1). Leishmania species were identified by sequencing of PCR products. Dog blood samples were subjected to the rapid immunochromatographic test (DPP) for detection of anti-Leishmania infantum antibodies. Results: 179 wild mammals were studied and 20.1% had Leishmania DNA successfully detected in at least one sample. Six mammal species were considered infected: Clyomys laticeps, Necromys lasiurus, Nectomys rattus, Rhipidomys macrurus, Didelphis albiventris and Gracilinanus agilis. No significant difference, comparing the proportion of individuals with Leishmania spp., was observed between the sampled areas and wild mammal species. Most of the positive samples were collected from the rodent N. lasiurus, infected by L. amazonensis or L. braziliensis. Moreover, infections by Trypanosoma spp. were detected in N. lasiurus and G. agilis. All 19 dog samples were positive by DPP; however, only three (15.8%) were confirmed by PCR assays. DNA sequences of ITS1 dog amplicons showed 100% identity with L. infantum sequence. Conclusions: The results suggest the participation of six species of wild mammals in the enzootic transmission of Leishmania spp. in FD. This is the first report of L. amazonensis in N. lasiurus
