    Commitment versus persuasion in the three-party constrained voter model

    In the framework of the three-party constrained voter model, where voters of two radical parties (A and B) interact with "centrists" (C and Cz), we study the competition between a persuasive majority and a committed minority. In this model, A's and B's are incompatible voters that can convince centrists or be swayed by them. Here, radical voters are more persuasive than centrists, whose sub-population consists of susceptible agents C and a fraction zeta of centrist zealots Cz. Whereas C's may adopt the opinions A and B with respective rates 1+delta_A and 1+delta_B (with delta_A>=delta_B>0), Cz's are committed individuals that always remain centrists. Furthermore, A and B voters can become (susceptible) centrists C with a rate 1. The resulting competition between commitment and persuasion is studied in the mean field limit and for a finite population on a complete graph. At mean field level, there is a continuous transition from a coexistence phase when zeta= Delta_c. In a finite population of size N, demographic fluctuations lead to centrism consensus and the dynamics is characterized by the mean consensus time tau. Because of the competition between commitment and persuasion, here consensus is reached much slower (zeta=Delta_c) than in the absence of zealots (when tau\simN). In fact, when zeta<Delta_c and there is an initial minority of centrists, the mean consensus time asymptotically grows as tau\simN^{-1/2} e^{N gamma}, where gamma is determined. The dynamics is thus characterized by a metastable state where the most persuasive voters and centrists coexist when delta_A>delta_B, whereas all species coexist when delta_A=delta_B. When zeta>=Delta_c and the initial density of centrists is low, one finds tau\simln N (when N>>1). Our analytical findings are corroborated by stochastic simulations.Comment: 25 pages, 6 figures.     A roadmap for high-resolution satellite soil moisture applications – confronting product characteristics with user requirements

    Soil moisture observations are of broad scientific interest and practical value for a wide range of applications. The scientific community has made significant progress in estimating soil moisture from satellite-based Earth observation data, particularly in operationalizing coarse-resolution (25-50 km) soil moisture products. This review summarizes existing applications of satellite-derived soil moisture products and identifies gaps between the characteristics of currently available soil moisture products and the application requirements from various disciplines. We discuss the efforts devoted to the generation of high-resolution soil moisture products from satellite Synthetic Aperture Radar (SAR) data such as Sentinel-1 C-band backscatter observations and/or through downscaling of existing coarse-resolution microwave soil moisture products. Open issues and future opportunities of satellite-derived soil moisture are discussed, providing guidance for further development of operational soil moisture products and bridging the gap between the soil moisture user and supplier communities

    Consensus statement on blocking interleukin-6 receptor and interleukin-6 in inflammatory conditions: an update

    Background: Targeting interleukin (IL)-6 has become a major therapeutic strategy in the treatment of immune-mediated inflammatory disease. Interference with the IL-6 pathway can be directed at the specific receptor using anti-IL-6Rα antibodies or by directly inhibiting the IL-6 cytokine. This paper is an update of a previous consensus document, based on most recent evidence and expert opinion, that aims to inform on the medical use of interfering with the IL-6 pathway. Methods: A systematic literature research was performed that focused on IL-6-pathway inhibitors in inflammatory diseases. Evidence was put in context by a large group of international experts and patients in a subsequent consensus process. All were involved in formulating the consensus statements, and in the preparation of this document. Results: The consensus process covered relevant aspects of dosing and populations for different indications of IL-6 pathway inhibitors that are approved across the world, including rheumatoid arthritis, polyarticular-course and systemic juvenile idiopathic arthritis, giant cell arteritis, Takayasu arteritis, adult-onset Still’s disease, Castleman’s disease, chimeric antigen receptor-T-cell-induced cytokine release syndrome, neuromyelitis optica spectrum disorder and severe COVID-19. Also addressed were other clinical aspects of the use of IL-6 pathway inhibitors, including pretreatment screening, safety, contraindications and monitoring. Conclusions: The document provides a comprehensive consensus on the use of IL-6 inhibition to treat inflammatory disorders to inform healthcare professionals (including researchers), patients, administrators and payers

    Comparative cellular analysis of motor cortex in human, marmoset and mouse

    The primary motor cortex (M1) is essential for voluntary fine-motor control and is functionally conserved across mammals(1). Here, using high-throughput transcriptomic and epigenomic profiling of more than 450,000 single nuclei in humans, marmoset monkeys and mice, we demonstrate a broadly conserved cellular makeup of this region, with similarities that mirror evolutionary distance and are consistent between the transcriptome and epigenome. The core conserved molecular identities of neuronal and non-neuronal cell types allow us to generate a cross-species consensus classification of cell types, and to infer conserved properties of cell types across species. Despite the overall conservation, however, many species-dependent specializations are apparent, including differences in cell-type proportions, gene expression, DNA methylation and chromatin state. Few cell-type marker genes are conserved across species, revealing a short list of candidate genes and regulatory mechanisms that are responsible for conserved features of homologous cell types, such as the GABAergic chandelier cells. This consensus transcriptomic classification allows us to use patch-seq (a combination of whole-cell patch-clamp recordings, RNA sequencing and morphological characterization) to identify corticospinal Betz cells from layer 5 in non-human primates and humans, and to characterize their highly specialized physiology and anatomy. These findings highlight the robust molecular underpinnings of cell-type diversity in M1 across mammals, and point to the genes and regulatory pathways responsible for the functional identity of cell types and their species-specific adaptations.Cardiovascular Aspects of Radiolog

    Composition of woody species in a dynamic forest-woodland-savannah mosaic in Uganda: implications for conservation and management

    Forest¿woodland¿savannah mosaics are a common feature in the East African landscape. For the conservation of the woody species that occur in such landscapes, the species patterns and the factors that maintain it need to be understood. We studied the woody species distribution in a forest¿woodland¿savannah mosaic in Budongo Forest Reserve, Uganda. The existing vegetation gradients were analyzed using data from a total of 591 plots of 400 or 500 m2 each. Remotely sensed data was used to explore current vegetation cover and the gradients there in for the whole area. A clear species gradient exists in the study area ranging from forest, where there is least disturbance, to wooded grassland, where frequent fire disturbance occurs. Most species are not limited to a specific part of the gradient although many show a maximum abundance at some point along the gradient. Fire and accessibility to the protected area were closely related to variation in species composition along the ordination axis with species like Cynometra alexandri and Uvariopsis congensis occurring at one end of the gradient and Combretum guenzi and Lonchocarpus laxiflorus at the other. The vegetation cover classes identified in the area differed in diversity, density and, especially, basal area. All vegetation cover classes, except open woodland, had indicator species. Diospyros abyssinica, Uvariopsis congensis, Holoptelea grandis and all Celtis species were the indicator species for the forest class, Terminalia velutina and Albizia grandbracteata for closed woodland, Grewia mollis and Combretum mole for very open woodland and Lonchocarpus laxiflorus, Grewia bicolor and Combretum guenzi for the wooded grassland class. Eleven of the species occurred in all cover classes and most of the species that occurred in more than one vegetation cover class showed peak abundance in a specific cover class. Species composition in the study area changes gradually from forest to savannah. Along the gradient, the cover classes are distinguishable in terms of species composition and vegetation structure. These classes are, however, interrelated in species composition. For conservation of the full range of the species within this East African landscape, the mosaic has to be managed as an integrated whole. Burning should be varied over the area with the forest not being burnt at all and the wooded grassland burnt regularly.     Tocilizumab in patients admitted to hospital with COVID-19 (RECOVERY): a randomised, controlled, open-label, platform trial

    Background: In this study, we aimed to evaluate the effects of tocilizumab in adult patients admitted to hospital with COVID-19 with both hypoxia and systemic inflammation. Methods: This randomised, controlled, open-label, platform trial (Randomised Evaluation of COVID-19 Therapy [RECOVERY]), is assessing several possible treatments in patients hospitalised with COVID-19 in the UK. Those trial participants with hypoxia (oxygen saturation &lt;92% on air or requiring oxygen therapy) and evidence of systemic inflammation (C-reactive protein ≥75 mg/L) were eligible for random assignment in a 1:1 ratio to usual standard of care alone versus usual standard of care plus tocilizumab at a dose of 400 mg–800 mg (depending on weight) given intravenously. A second dose could be given 12–24 h later if the patient's condition had not improved. The primary outcome was 28-day mortality, assessed in the intention-to-treat population. The trial is registered with ISRCTN (50189673) and ClinicalTrials.gov (NCT04381936). Findings: Between April 23, 2020, and Jan 24, 2021, 4116 adults of 21 550 patients enrolled into the RECOVERY trial were included in the assessment of tocilizumab, including 3385 (82%) patients receiving systemic corticosteroids. Overall, 621 (31%) of the 2022 patients allocated tocilizumab and 729 (35%) of the 2094 patients allocated to usual care died within 28 days (rate ratio 0·85; 95% CI 0·76–0·94; p=0·0028). Consistent results were seen in all prespecified subgroups of patients, including those receiving systemic corticosteroids. Patients allocated to tocilizumab were more likely to be discharged from hospital within 28 days (57% vs 50%; rate ratio 1·22; 1·12–1·33; p&lt;0·0001). Among those not receiving invasive mechanical ventilation at baseline, patients allocated tocilizumab were less likely to reach the composite endpoint of invasive mechanical ventilation or death (35% vs 42%; risk ratio 0·84; 95% CI 0·77–0·92; p&lt;0·0001). Interpretation: In hospitalised COVID-19 patients with hypoxia and systemic inflammation, tocilizumab improved survival and other clinical outcomes. These benefits were seen regardless of the amount of respiratory support and were additional to the benefits of systemic corticosteroids.     Convalescent plasma in patients admitted to hospital with COVID-19 (RECOVERY): a randomised controlled, open-label, platform trial

    Background: Many patients with COVID-19 have been treated with plasma containing anti-SARS-CoV-2 antibodies. We aimed to evaluate the safety and efficacy of convalescent plasma therapy in patients admitted to hospital with COVID-19. Methods: This randomised, controlled, open-label, platform trial (Randomised Evaluation of COVID-19 Therapy [RECOVERY]) is assessing several possible treatments in patients hospitalised with COVID-19 in the UK. The trial is underway at 177 NHS hospitals from across the UK. Eligible and consenting patients were randomly assigned (1:1) to receive either usual care alone (usual care group) or usual care plus high-titre convalescent plasma (convalescent plasma group). The primary outcome was 28-day mortality, analysed on an intention-to-treat basis. The trial is registered with ISRCTN, 50189673, and ClinicalTrials.gov, NCT04381936. Findings: Between May 28, 2020, and Jan 15, 2021, 11558 (71%) of 16287 patients enrolled in RECOVERY were eligible to receive convalescent plasma and were assigned to either the convalescent plasma group or the usual care group. There was no significant difference in 28-day mortality between the two groups: 1399 (24%) of 5795 patients in the convalescent plasma group and 1408 (24%) of 5763 patients in the usual care group died within 28 days (rate ratio 1·00, 95% CI 0·93–1·07; p=0·95). The 28-day mortality rate ratio was similar in all prespecified subgroups of patients, including in those patients without detectable SARS-CoV-2 antibodies at randomisation. Allocation to convalescent plasma had no significant effect on the proportion of patients discharged from hospital within 28 days (3832 [66%] patients in the convalescent plasma group vs 3822 [66%] patients in the usual care group; rate ratio 0·99, 95% CI 0·94–1·03; p=0·57). Among those not on invasive mechanical ventilation at randomisation, there was no significant difference in the proportion of patients meeting the composite endpoint of progression to invasive mechanical ventilation or death (1568 [29%] of 5493 patients in the convalescent plasma group vs 1568 [29%] of 5448 patients in the usual care group; rate ratio 0·99, 95% CI 0·93–1·05; p=0·79). Interpretation: In patients hospitalised with COVID-19, high-titre convalescent plasma did not improve survival or other prespecified clinical outcomes.     Contingent negative variation and evoked responses recorded by radio-telemetry in free-ranging subjects

Using an 8-channel radio-telemetry system (RTE) records were obtained of the EEG, pulse rate, respiration, evoked responses and Contingent Negative Variation (CNV) in four normal subjects and three patients with intracerebral electrodes. The subjects were free to move about within 30 m of the receiving aerial. Two RTE channels were modified to provide time constants of 7 sec. 2. 2. Auditory stimuli, synchronized with the operation of average response computers, were transmitted to the subjects by a separate radio-control link. These were used as conditional and imperative signals to the subjects to perform various tasks: pressing a button to arrest the imperative signals, turning the pages of a book, inserting pegs in a peg-board and coming to a stop on a bicycle. 3. 3. Responses to the signals were averaged on line with two barrier-grid tubes and a 2-channel Enhancetron and the intrinsic rhythms were analysed with a 2-channel frequency analyser. The pulse rate was indicated by a cardiotachometer. 4. 4. The initil responses and CNV during the reception of paired auditory signals were similar to those seen with direct connection, provided that the subjects were engaged in some task related to the signals. The CNV was attenuated during exercise or conversation only while the subject was inattentive to the signals. 5. 5. The amplitude of the intracerebral responses to the conditional and imperative stimuli in the patients with implanted electrodes was reduced when the scalp CNV was attenuated by isolation or distraction. 6. 6. During the performance of fairly complex tasks following the auditory signals, the CNV terminated only at the completion of the task, not at the moment of muscular effort. 7. 7. The radio-control link was also used to instruct an experimenter when to loss a ball to a subject or to feint. In this situation also the CNV developed only when the subject was sure the ball was in the air and terminated when it was caught. 8. 8. These observations suggest that the interactions of evoked responses and CNV seen in laboratory conditions also accompany normal activity and the performance of everyday tasks
