    Avaliação do ácido trans, trans-mucônico urinário como biomarcador de exposição ao benzeno

    OBJECTIVE: To assess the use of trans, trans-muconic acid as a biomarker of occupational exposure to benzene. METHODS: Trans, trans-muconic acid in urine samples of exposed (exposed group, n=36) and non-exposed (non-exposed group, n=116) workers to benzene. Urinary levels of trans, trans-muconic acid were quantified by high-performance liquid chromatography. The study sample consisted of subjects exposed to benzene in an oil refinery in Belo Horizonte, Brazil. Non-parametric statistical analysis was carried out using Kruskall-Wallis test, Mann-Whitney test and Spearman correlation at pOBJETIVO: Avaliar o uso do ácido trans, trans-mucônico urinário como biomarcador na monitorização da exposição ocupacional ao benzeno. MÉTODOS: Foi estudado o comportamento do ácido trans, trans-mucônico em amostras de urina de indivíduos expostos (N=36) e não expostos (N=116) ocupacionalmente ao solvente. A concentração urinária do ácido foi determinada por Cromatografia Líquida de Alta Eficiência. A amostra foi constituída de indivíduos expostos ao benzeno em uma refinaria de petróleo localizada em Belo Horizonte, MG. Foram empregados os testes estatísticos não-paramétricos de Kruskall-Wallis, Mann Witney e de correlação de Spearman, ao nível de significância de 0,05%. RESULTADOS: A exposição média ao benzeno dos trabalhadores selecionados foi de 0,15±0,05 mg/m³ (0,05 ppm) o que resultou em um valor médio do metabólito urinário de 0,19±0,04 mg/g de creatinina. A faixa de referência do ácido trans, trans-mucônico no grupo não exposto variou de 0,03 a 0,26 mg/g de creatinina (média de 0,10±0,08 mg/g de creatinina). Foi encontrada uma diferença significativa entre os níveis de ácido trans, trans-mucônico do grupo exposto e não exposto. Entretanto, não houve correlação entre os níveis do metabólito urinário e do benzeno no ar. Foi observada a correlação entre ácido trans, trans-mucônico e hábito de fumar no grupo de indivíduos expostos. A ingestão de álcool num período de até 48 horas antes da coleta das amostras não mostrou interferir nos níveis do metabólito nos dois grupos estudados. Foi observada a correlação entre ácido trans, trans-mucônico e idade (faixa etária de 18 a 25 anos) no grupo de não expostos. CONCLUSÕES: Os resultados obtidos evidenciaram a importância de ser mais bem avaliada a influência do hábito de fumar e da faixa etária do trabalhador nos níveis urinários do ácido trans, trans-mucônico

    Antimicrobial resistance of isolated Streptococcus pneumoniae in a hospital of the Brazilian public system

    Streptococcus pneumoniae is the predominant bacterial agent that affects the human population with pneumonia. This disease is an important cause of death in the elderly and the children under five years old. in this study, 29 strains of invasive S. pneumoniae were isolated from 29 patients of pneumonia, bacteremia and meningitis in the laboratory of the Municipal Hospital in Paulinia, Brazil, from May 2006 to October 2007. Patients' age ranged from 8 months old to 60 years old. These strains of S. pneumoniae were isolated from blood, pleural fluid and cerebrospinal fluid (CSF) of patients. After typing of encapsulated strains of S. pneumoniae through quellung reaction, their resistance to antimicrobial agents was gauged through Disc Diffusion Technique followed by determination of minimum inhibitory concentration (MIC). Among the 29 strains analyzed, 23 were methicillin-sensitive and six were methicillin-resistant and penicillin intermediate resistant. No strain presented full resistance to penicillin. Serotyping was performed only in two samples, which belonged to serotype 18.     Annona coriacea Mart. fractions promote cell cycle arrest and inhibit autophagic flux in human cervical cancer cell lines

    Plant-based compounds are an option to explore and perhaps overcome the limitations of current antitumor treatments. Annona coriacea Mart. is a plant with a broad spectrum of biological activities, but its antitumor activity is still unclear. The purpose of our study was to determine the effects of A. coriacea fractions on a panel of cervical cancer cell lines and a normal keratinocyte cell line. The antitumor effect was investigated in vitro by viability assays, cell cycle, apoptosis, migration, and invasion assays. Intracellular signaling was assessed by Western blot, and major compounds were identified by mass spectrometry. All fractions exhibited a cytotoxic effect on cisplatin-resistant cell lines, SiHa and HeLa. C3 and C5 were significantly more cytotoxic and selective than cisplatin in SiHa and Hela cells. However, in CaSki, a cisplatin-sensitive cell line, the compounds did not demonstrate higher cytotoxicity when compared with cisplatin. Alkaloids and acetogenins were the main compounds identified in the fractions. These fractions also markedly decreased cell proliferation with p21 increase and cell cycle arrest in G2/M. These effects were accompanied by an increase of H2AX phosphorylation levels and DNA damage index. In addition, fractions C3 and C5 promoted p62 accumulation and decrease of LC3II, as well as acid vesicle levels, indicating the inhibition of autophagic flow.     Brazilian isolates of Trypanosoma cruzi from humans and triatomines classified into two lineages using mini-exon and ribosomal RNA sequences

    Traditional molecular and biochemical methods, such as schizodeme analysis, karyotyping, DNA fingerprinting, and enzyme electrophoretic profiles, have shown a large variability among Trypanosoma cruzi isolates. In contrast to those results, polymerase chain reaction (PCR) amplification of sequences from the 24Sα ribosomal RNA gene and from the mini-exon gene nontranscribed spacer indicated a dimorphism among T. cruzi isolates, which enabled the definition of two major parasite lineages. In the present study, 86 T. cruzi field stocks (68 isolated from humans with defined presentations of Chagas' disease and 18 from triatomines) derived from four Brazilian geographic areas were typed by the PCR assay based on the DNA sequences of the mini-exon and 24Sα rRNA genes.
