Efficacy Of First Time Furosemide Use In Thoroughbred Racehorses In Brazil
Racehorses are commonly affected by a condition known as Exercise Induced Pulmonary Haemorrhage (EIPH) which consists of rupture of alveolar membranes and extravasation of blood into the airways. EIPH is considered to negatively affect racehorse performance and there is no current treatment. In order to reduce the amount of bleeding the diuretic furosemide has been used prior to exercise and competition in some racing jurisdictions. Efficacy of furosemide in reducing EIPH is documented in this research through endoscopic observation of the airways of horses positive for EIPH in two consecutive races
Occurrence and diversity of yeast species isolated from fish feed and tambatinga gut
The present study aimed to identify yeasts species isolated from fish feed and the intestinal tracts of tambatinga fish (Colossoma macropomum × Piaractus brachypomum) cultivated in a Brazilian fish farm. Twenty tambatingas and 30 fish feed samples from different brands were acquired from two commercial establishments in the state of Piauí, Brazil. The sampled guts were divided into three equal parts, namely the anterior, medium and posterior portions, totaling 60 samples. Molecular identification was performed by PCR amplification and sequencing of the D1/D2 regions of the large rRNA subunit gene. Sixteen Candida nivariensis strains in the gut were identified. The fish feed samples showed a higher variability of yeast species, with the following species isolated: Hyphopichia burtonii (23.08%), Lodderomyces elongisporus (15.39%) and Trichosporon asahii (15.39%), Candida nivariensis (7.69%), C. orthopsilosis (7.69%), C. parapsilosis (7.69%), Rhodotorula minuta (7.69%), Sterigmatomyces elviae (7.69%), Cryptococcus liquefaciens (7.69%). Yeast found in feed, however, was not always isolated from tambatinga gut. Molecular identification allowed for the isolation of yeast species not previously reported in fish feed and gut samples.Fil: Pinheiro, Raizza Eveline E.. Environmental factors driving plant trait distributions in coastal zones of Atlantic Forest

All rights reserved.Environmental filtering has been defined as the effect of environmental gradients on species in a plant community and can be the dominant driver of community assembly. Here, we evaluate the relationship between plant communities and the environment in the Restinga vegetation. For this, we measured 11 functional traits of plant species present along transects covering a marked edaphic environmental gradient. This gradient was characterized through Principal Component Analysis of soil characteristics. The relationships between the edaphic gradient and functional traits were evaluated using linear models. Finally, we compared the contributions of species turnover and intraspecific variation to among-site variation in functional traits. The gradients associated with soil nutrients (PCA axis 1) and soil acidity and organic matter (PCA axis 2) were then used to test the observed changes in community composition and were significant predictors of the distribution of water potential, leaf dry matter content and K content, height and chlorophyll index. Decomposing the total variation in the distribution of functional traits between species turnover and intraspecific variation revealed that species turnover explains a greater proportion of the observed variation. We conclude that community assembly is strongly limited by environmental filters and mediated by functional traits at the species level.Peer reviewe
Generating real-world evidence on the quality use, benefits and safety of medicines in australia: History, challenges and a roadmap for the future
Australia spends more than $20 billion annually on medicines, delivering significant health benefits for the population. However, inappropriate prescribing and medicine use also result in harm to individuals and populations, and waste of precious health resources. Medication data linked with other routine collections enable evidence generation in pharmacoepidemiology; the science of quantifying the use, effectiveness and safety of medicines in real-world clinical practice. This review details the history of medicines policy and data access in Australia, the strengths of existing data sources, and the infrastructure and governance enabling and impeding evidence generation in the field. Currently, substantial gaps persist with respect to cohesive, contemporary linked data sources supporting quality use of medicines, effectiveness and safety research; exemplified by Aus-tralia’s limited capacity to contribute to the global effort in real-world studies of vaccine and dis-ease-modifying treatments for COVID-19. We propose a roadmap to bolster the discipline, and population health more broadly, underpinned by a distinct capability governing and streamlining access to linked data assets for accredited researchers. Robust real-world evidence generation requires current data roadblocks to be remedied as a matter of urgency to deliver efficient and equitable health care and improve the health and well-being of all Australians
Targeting MAGE-C1/CT7 Expression Increases Cell Sensitivity to the Proteasome Inhibitor Bortezomib in Multiple Myeloma Cell Lines
The MAGE-C1/CT7 encodes a cancer/testis antigen (CTA), is located on the chromosomal region Xq26-27 and is highly polymorphic in humans. MAGE-C1/CT7 is frequently expressed in multiple myeloma (MM) that may be a potential target for immunotherapy in this still incurable disease. MAGEC1/CT7 expression is restricted to malignant plasma cells and it has been suggested that MAGE-C1/CT7 might play a pathogenic role in MM; however, the exact function this protein in the pathophysiology of MM is not yet understood. Our objectives were (1) to clarify the role of MAGE-C1/CT7 in the control of cellular proliferation and cell cycle in myeloma and (2) to evaluate the impact of silencing MAGE-C1/CT7 on myeloma cells treated with bortezomib. Myeloma cell line SKO-007 was transduced for stable expression of shRNA-MAGE-C1/CT7. Downregulation of MAGE-C1/CT7 was confirmed by real time quantitative PCR and western blot. Functional assays included cell proliferation, cell invasion, cell cycle analysis and apoptosis. Western blot showed a 70-80% decrease in MAGE-C1/CT7 protein expression in inhibited cells (shRNA-MAGE-C1/CT7) when compared with controls. Functional assays did not indicate a difference in cell proliferation and DNA synthesis when inhibited cells were compared with controls. However, we found a decreased percentage of cells in the G2/M phase of the cell cycle among inhibited cells, but not in the controls (p < 0.05). When myeloma cells were treated with bortezomib, we observed a 48% reduction of cells in the G2/M phase among inhibited cells while controls showed 13% (empty vector) and 9% (ineffective shRNA) reduction, respectively (p < 0.01). Furthermore, inhibited cells treated with bortezomib showed an increased percentage of apoptotic cells (Annexin V+/PI-) in comparison with bortezomib-treated controls (p < 0.001). We found that MAGE-C1/CT7 protects SKO-007 cells against bortezomib-induced apoptosis. O Papel do ensino do português como língua estrangeira na defesa do multicultarismo
As política actuais existentes a nível oficial para a implementação e defesa do ensino da Língua Portuguesa como Língua Estrangeira (L. E.) na Europa e no resto do mundo levam-nos a pensar que são, sobretudo, os casos isolados de leitores portugueses pioneiros, inspirados e marginais que na sua missão individual e afastada lutam pela implementação e defesa desta língua nos seus países de acolhimento.
Segundo Volfgram, “cabe ensinar a alguns que o multiculturalismo não está apenas na teoria e sim ao nosso redor, nos elevando realmente à condição de seres humanos” (2005), e o mesmo é dizer que o multiculturalismo começa nas suas bases pela aprendizagem desinteressada e não interesseira das crianças na sua mais tenra idade. Não é impunemente que em países multiculturais como a Bélgica, a Língua Portuguesa ensinada como segunda língua ou como língua estrangeira desempenha um papel preponderante na defesa e na preservação do Português e, em simultâneo, pugna pela defesa incontestável da necessidade incontornável que o multiculturalismo é hoje.
É indubitável que a luta contra a xenofobia, a luta pela tolerância e o respeito mútuo, bem como o diálogo profícuo biunívoco não podem sobreviver actualmente sem uma consciencialização da importância das línguas minoritárias, da crioulização, da relação com as línguas maioritárias e da conquista da defesa do multiculturalismo hic et nunc.
Abordando algumas opiniões avisadas, esperamos trazer à discussão temas importantes, tais como, a necessidade de articulação de políticas de difusão da língua portuguesa na Europa e no Mundo concertadamente com o Brasil e outros Países Lusófonos, a necessidade de implementação de medidas concretas no terreno para defesa da Língua de Camões fora de Portugal, a sobrevivência do Português que embora sendo minoritária na Europa é uma das línguas mais faladas no mundo, a necessidade da consciencialização para a crescente importância geo-estratégica do Português paralelamente com o recrudescimento do multiculturalismo à escala global
Human cerebral organoids and fetal brain tissue share proteomic similarities
The limited access to functional human brain tissue has led to the development of stem cell-based alternative models. The differentiation of human pluripotent stem cells into cerebral organoids with self-organized architecture has created novel opportunities to study the early stages of the human cerebral formation. Here we applied state-of-the-art label-free shotgun proteomics to compare the proteome of stem cell-derived cerebral organoids to the human fetal brain. We identified 3,073 proteins associated with different developmental stages, from neural progenitors to neurons, astrocytes, or oligodendrocytes. The major protein groups are associated with neurogenesis, axon guidance, synaptogenesis, and cortical brain development. Glial cell proteins related to cell growth and maintenance, energy metabolism, cell communication, and signaling were also described. Our data support the variety of cells and neural network functional pathways observed within cell-derived cerebral organoids, confirming their usefulness as an alternative model. Protective Properties in Hymenaea martiana Hayne against Multi-drug-resistant Staphylococcus aureus
Antibiotic resistance represents a widespread problem in milk production. The identification of compounds for a topically applied ointment used in mastitis therapy remains elusive. Compounds from the genus Hymenaea can be administered in cases of multi-drug-resistant Staphylococcus aureus infection for ruminant species, but the protective properties are not well known. Wi this research the aim is verify the protective effects of H. martiana against S. aureus infection in bovine mammary epithelial cell line (MAC-T) and to obtain an antioxidant profile evaluation in vitro. The MAC-T cells were challenged with S. aureus after being exposed to the extract of the H. martiana in the protective assay. For the verification of the viability of the MAC-T cells, the MTT assay was performed, and was used dilutions of the plant extract, starting at 2.5%. The extract of H. martiana was evaluated for antioxidant aspect in different dilutions by FRAP, ORAC and DPPH. A variety of flavonoids (quercetin, luteolin, etc.) have been identified as the main components by using mass spectrometry, reinforcing our in vitro findings that flavonoids, especially quercetin, have a medicinal profile capable of killing mastitis-causing bacteria. An excellent antioxidant pattern was observed in the 2.5% solution; however, membrane integrity in MAC-T cells was compromised. Those findings suggest low dilutions of H. martiana extract has a desirable protective effect from S. aureus pathogenesis. Our in vitro studies can be gleaned upon for further in vivo studies
