    The diffusion of Constructivist ideas in publications in Germany, 1918-1925

    After a long period of disrupted communication, the years 1918-21 saw a rapid infiltration of Russian post-revolutionary culture into German progressive circles. In the thriving book market of Berlin, resourceful apologists like El Lissitzky and Il'ya Erenburg found conditions favourable to the diffusion of their ideas and played a critical role in encouraging pro-Soviet sentiment among the local vanguard. Their cosmopolitan enthusiasm, shared by a number of artists throughout Europe, bolstered hopes for co-operation between the modernist forces of all countries. For a brief while. Constructivism became a rallying point for those artists who had faith in their ability to foster cultural transformation and felt a common purpose in creating an environment conducive to social harmony. Various initiatives were taken to lay the foundation for a truly cohesive effort but yielded only temporary alliances. Serving as alternative platforms from which to gain influence and individual recognition, periodicals and slim book publications ultimately provided the main element of continuity in the attempt to found a Constructivist International. By 1925, those who had devoted most energy to this collective project had distanced themselves from a strong identification as Constructivists on the Berlin art scene and struck out in new directions

    Non-Financial Conflicts of Interest in Academic Grant Evaluation: A Qualitative Study of Multiple Stakeholders in France

    International audienceBACKGROUND: Peer review is the most widely used method for evaluating grant applications in clinical research. Criticisms of peer review include lack of equity, suspicion of biases, and conflicts of interest (CoI). CoIs raise questions of fairness, transparency, and trust in grant allocation. Few observational studies have assessed these issues. We report the results of a qualitative study on reviewers' and applicants' perceptions and experiences of CoIs in reviews of French academic grant applications. METHODOLOGY AND PRINCIPAL FINDINGS: We designed a qualitative study using semi-structured interviews and direct observation. We asked members of assessment panels, external reviewers, and applicants to participate in semi-structured interviews. Two independent researchers conducted in-depth reviews and line-by-line coding of all transcribed interviews, which were also subjected to TropesÂź software text analysis, to detect and qualify themes associated with CoIs. Most participants (73/98) spontaneously reported that non-financial CoIs predominated over financial CoIs. Non-financial CoIs mainly involved rivalry among disciplines, cronyism, and geographic and academic biases. However, none of the participants challenged the validity of peer review. Reviewers who felt they might be affected by CoIs said they reacted in a variety of ways: routine refusal to review, routine attempt to conduct an impartial review, or decision on a case-by-case basis. Multiple means of managing non-financial CoIs were suggested, including increased transparency throughout the review process, with public disclosure of non-financial CoIs, and careful selection of independent reviewers, including foreign experts and methodologists. CONCLUSIONS: Our study underscores the importance of considering non-financial CoIs when reviewing research grant applications, in addition to financial CoIs. Specific measures are needed to prevent a negative impact of non-financial CoIs on the fairness of resource allocation. Whether and how public disclosure of non-financial CoIs should be accomplished remains debatable

    Short-chain lipid peroxidation products form covalent adducts with pyruvate kinase and inhibit its activity in vitro and in breast cancer cells

    Pyruvate kinase catalyses the last step in glycolysis and has been suggested to contribute to the regulation of aerobic glycolysis in cancer cells. It can be inhibited by oxidation of cysteine residues in vitro and in vivo, which is relevant to the more pro-oxidant state in cancer and proliferating tissues. These conditions also favour lipid peroxidation and the formation of electrophilic fragmentation products, including short-chain aldehydes that can covalently modify proteins. However, as yet few studies have investigated their interactions with pyruvate kinase, so we investigated the effects of three different aldehydes, acrolein, malondialdehyde and 4-hydroxy-2(E)-hexenal (HHE), on the structure and activity of the enzyme. Analysis by LC-MS/MS showed unique modification profiles for each aldehyde, but Cys152, Cys423 and Cys474 were the residues most susceptible to electrophilic modification. Analysis of enzymatic activity under these conditions showed that acrolein was the strongest inhibitor, and at incubation times longer than 2 h, pathophysiological concentrations induced significant effects. Treatment of MCF-7 cells with the aldehydes caused similar losses of pyruvate kinase activity to those observed in vitro, and at lower concentrations than those required to cause cell death, with time and dose-dependent effects; acrolein adducts on Cys152 and Cys358 were detected. Cys358 and Cys474 are located at or near the allosteric or active sites, and formation of adducts on these residues probably contributes to loss of activity at low treatment concentrations. This study provides the first detailed analysis of the structure-activity relationship of C3 and C6 aldehydes with pyruvate kinase, and suggests that reactive short-chain aldehydes generated in diseases with an oxidative aetiology or from environmental exposure such as smoking could be involved in the metabolic alterations observed in cancer cells, through alteration of pyruvate kinase activity

    Colostral antibody induced interference of inactivated bluetongue serotype-8 vaccines in calves

    Since its introduction into northern Europe in 2006, bluetongue has become a major threat to animal health. While the efficacy of commercial vaccines has been clearly demonstrated in livestock, little is known regarding the effect of maternal immunity on vaccinal efficacy. Here, we have investigated the duration and amplitude of colostral antibody-induced immunity in calves born to dams vaccinated against bluetongue virus serotype 8 (BTV-8) and the extent of colostral antibody-induced interference of vaccination in these calves. Twenty-two calf-cow pairs were included in this survey. The median age at which calves became seronegative for BTV was 84 and 112 days as assayed by seroneutralisation test (SNT) and VP7 BTV competitive ELISA (cELISA), respectively. At the mean age of 118 days, 13/22 calves were immunized with inactivated BTV-8 vaccine. In most calves vaccination elicited a weak immune response, with seroconversion in only 3/13 calves. The amplitude of the humoral response to vaccination was inversely proportional to the maternal antibody level prior to vaccination. Thus, the lack of response was attributed to the persistence of virus-specific colostral antibodies that interfered with the induction of the immune response. These data suggest that the recommended age for vaccination of calves born to vaccinated dams needs to be adjusted in order to optimize vaccinal efficacy

    Dilated cardiomyopathy in a cat with congenital hyposomatotropism.

    Case summary A 7-month-old domestic shorthair cat was presented for evaluation of stunted growth, recurrent hypoglycaemia during the first months of its life and altered mentation. Complete blood count and biochemistry were unremarkable, except for mildly elevated serum creatinine concentration (despite low muscle mass) and concurrent isosthenuria. Hyposomatotropism was diagnosed based on persistent low circulating insulin-like growth factor 1 concentrations and a lack of response of circulating growth hormone (GH) concentration after the administration of GH-releasing hormone. Other endocrinopathies such as hypothyroidism and hypoadrenocorticism were excluded. MRI of the brain revealed a fluid-filled empty sella tursica, consistent with a pituitary cyst and atrophy/hypoplasia of the pituitary. Echocardiography was unremarkable at the time of diagnosis of hyposomatotropism. Three months later, ovariohysterectomy revealed immature ovaries, raising the suspicion of luteinising and follicle-stimulating hormone deficiency. At 1 year of age, the cat developed congestive heart failure secondarily to dilated cardiomyopathy (DCM) with severely reduced left ventricular systolic function and died a few days later. Pathology showed atrophy of the adenohypophysis, epithelial delineation of the pituitary cysts, mild cardiomegaly, multifocal fibrosis of the left ventricle and a mild, multifocal, chronic epicarditis. Relevance and novel information GH deficiency is a very rare endocrinopathy in cats. This is the first case to describe the development of DCM with concurrent hyposomatotropism, which has previously been reported in human medicine. Other notable abnormalities that could be related to GH deficiency are juvenile self-limiting hypoglycaemia, behavioural changes and possible nephropathy

    Measurements of nitric oxide and ammonia soil fluxes from a wet savanna ecosystem site in West Africa during the DACCIWA field campaign

    Biogenic fluxes from soil at a local and regional scale are crucial to study air pollution and climate. Here we present field measurements of soil fluxes of nitric oxide (NO) and ammonia (NH3) observed over four different land cover types, i.e. bare soil, grassland, maize field, and forest, at an inland rural site in Benin, West Africa, during the DACCIWA field campaign in June and July 2016. At the regional scale, urbanization and a massive growth in population in West Africa have been causing a strong increase in anthropogenic emissions. Anthropogenic pollutants are transported inland and northward from the megacities located on the coast, where the reaction with biogenic emissions may lead to enhanced ozone production outside urban areas, as well as secondary organic aerosol formation, with detrimental effects on humans, animals, natural vegetation, and crops. We observe NO fluxes up to 48.05 ngNm 2 s 1. NO fluxes averaged over all land cover types are 4:79 5:59 ngNm 2 s 1, and maximum soil emissions of NO are recorded over bare soil. NH3 is dominated by deposition for all land cover types. NH3 fluxes range between 6:59 and 4.96 ngNm 2 s 1. NH3 fluxes averaged over all land cover types are 0:911:27 ngNm 2 s 1, and maximum NH3 deposition is measured over bare soil. The observations show high spatial variability even for the same soil type, same day, and same meteorological conditions. We compare point daytime average measurements of NO emissions recorded during the field campaign with those simulated by GEOS-Chem (Goddard Earth Observing System Chemistry Model) for the same site and find good agreement. In an attempt to quantify NO emissions at the regional and national scale, we also provide a tentative estimate of total NO emissions for the entire country of Benin for the month of July using two distinct methods: upscaling point measurements and using the GEOS-Chem model. The two methods give similar results: 1:170:6 and 1.44 GgN month 1, respectively. Total NH3 deposition estimated by upscaling point measurements for the month of July is 0.21 GgN month1
