    Synthesis and structural characterization of benzyl-functionalized N-heterocyclic carbene platinum complexes: Dramatic substituent effect on anti-cancer activity

    International audienceA series of platinum (II) complexes bearing N-heterocyclic carbene NHC ligands functionalized by various benzyl moieties were synthesized and characterized. The molecular structure of two complexes has been confirmed by X-ray diffraction studies on single crystals. Investigation of in vitro cytotoxic activities against various cancer cell lines revealed a strong substituent effect: IC50 as low as 0.005â€ŻÎŒM were obtained depending on the substituent on the benzyl moiety

    Metal ions (Ag+^{+}, Cd2+^{2+}, Eu3+^{3+}, Sm3+^{3+}) and protons can, through coordination and protonation, modulate in three specific ways the structural information contained in the pyrazine-based heterocyclic strand LL (obtained from 2,5-bis(methylhydrazino)pyrazine and 2 equivalents of 2-pyridinecarboxaldehyde), thus generating two linear rod-like conformations and a bent one. This conformational diversity is associated with a structural one that consists of two diprotonated forms (H2L(PF6)2H_{2}L(PF_{6})_{2} and H2L(CF3SO3)2H_{2}L(CF_{3}SO_{3})_{2}), a polymeric architecture [AgL]n(CF3SO3)n[AgL]_{n}(CF_{3}SO_{3})_{n}, two rack-like complexes ([Eu2H2L3(CF3SO3)6](PF6)2([Eu_{2}H_{2}L_{3}(CF_{3}SO_{3})_{6}](PF_{6})_{2} and [Sm2H2L3(CF3SO3)6](PF6)2)[Sm_{2}H_{2}L_{3}(CF_{3}SO_{3})_{6}](PF_{6})_{2}) and a grid-like structure ([Cd4L4](CF3SO3)8[Cd_{4}L_{4}](CF_{3}SO_{3})_{8})

    In the copper(II) complex, bis­{(E)-1-[(2,4,6-tri­bromo­phen­yl)diazen­yl]naph­thalen-2- olato}copper(II), [Cu(C₁₆H₈Br₃N₂O)₂], (I), the metal cation is coord­inated by two N atoms and two O atoms from two bidentate (E)-1-[(2,4,6-tri­bromo­phen­yl)diazen­yl]naphthalen-2-olate ligands, forming a slightly distorted square-planar environment. In one of the ligands, the tri­bromo­benzene ring is inclined to the naphthalene ring system by 37.4 (5)°, creating a weak intra­molecular Cu...Br inter­action [3.134 (2) Å], while in the other ligand, the tri­bromo­benzene ring is inclined to the naphthalene ring system by 72.1 (6)°. In the isotypic nickel(II) and palladium(II) complexes, namely bis­{(E)-1-[(2,4,6-tri­bromo­phen­yl)diazen­ yl]naphthalen-2-olato}nickel(II), [Ni(C₁₆H₈Br₃N₂O)₂], (II), and bis­{(E)-1- [(2,4,6-tri­bromo­phen­yl)diazen­yl]naphthalen-2-olato}palladium(II), [Pd(C₁₆H₈Br₃N₂O)₂], (III), respectively, the metal atoms are located on centres of inversion, hence the metal coordination spheres have perfect square-planar geometries. The tri­bromo­benzene rings are inclined to the naphthalene ring systems by 80.79 (18)° in (II) and by 80.8 (3)° in (III). In the crystal of (I), mol­ecules are linked by C-H...Br hydrogen bonds, forming chains along [010]. The chains are linked by C-H...Pi inter­actions, forming sheets parallel to (011). In the crystals of (II) and (III), mol­ecules are linked by C-H...Pi inter­actions, forming slabs parallel to (10-1). For the copper(II) complex (I), a region of disordered electron density was corrected for using the SQUEEZE routine in PLATON [Spek (2015). Acta Cryst. C71, 9-18].     Deep-Learning Assessed Muscular Hypodensity Independently Predicts Mortality in DLBCL Patients Younger Than 60 Years.

    [en] BACKGROUND: Muscle depletion (MD) assessed by computed tomography (CT) has been shown to be a predictive marker in solid tumors, but has not been assessed in non-Hodgkin's lymphomas. Despite software improvements, MD measurement remains highly time-consuming and cannot be used in clinical practice. METHODS: This study reports the development of a Deep-Learning automatic segmentation algorithm (DLASA) to measure MD, and investigate its predictive value in a cohort of 656 diffuse large B cell lymphoma (DLBCL) patients included in the GAINED phase III prospective trial (NCT01659099). RESULTS: After training on a series of 190 patients, the DLASA achieved a Dice coefficient of 0.97 ± 0.03. In the cohort, the median skeletal muscle index was 50.2 cm2/m2 and median muscle attenuation (MA) was 36.1 Hounsfield units (HU). No impact of sarcopenia was found on either progression free survival (PFS) or overall survival (OS). Muscular hypodensity, defined as MA below the tenth percentile according to sex, was associated with a lower OS and PFS, respectively (HR = 2.80 (95% CI 1.58-4.95), p < 0.001, and HR = 2.22 (95% CI 1.43-3.45), p < 0.001). Muscular hypodensity appears to be an independent risk factor for mortality in DLBCL and because of DLASA can be estimated in routine practice

The N atoms are protonated and, thus, the structure is a bis-zwitterionic compound in the solid state. The zwitterion shows strong intramolecular N—H...O hydrogen bonds between the iminium N and the naphthalenolate O atoms
