    Composition and dynamics of the fungal population in a typical Phaeozem luvico in Argentina

Here, we studied the dynamics of the soil fungal populations in a typical Phaeozem luvico in Argentina and related it to the management practices. Soil samples (at 0–10cm depth) were collected from a field cultivated with wheat, at different sampling times: at post-harvest, before sowing, and at tillering. The relative abundance of individuals in the population on Nash Snyder and Oxgall agar media was assessed as colony forming units (CFU). The fungal population was classified by numerical taxonomy at the different sampling times. The highest values of CFUg−1of soil were found at post-harvest under reduced tillage and differed significantly from those at conventional tillage. The generaTrichoderma,Fusarium,Aspergillus,Penicillium, andFusarium oxysporumwere present in the largest number of samples and discriminated the fungal community between times. This discrimination could be related to alterations in the availability of carbon sources during stubble degradation at post-harvest and before sowing and were of lesser importance at tillering

    An Analytical Study of Coupled Two-State Stochastic Resonators

    The two-state model of stochastic resonance is extended to a chain of coupled two-state elements governed by the dynamics of Glauber's stochastic Ising model. Appropriate assumptions on the model parameters turn the chain into a prototype system of coupled stochastic resonators. In a weak-signal limit analytical expressions are derived for the spectral power amplification and the signal-to-noise ratio of a two-state element embedded into the chain. The effect of the coupling between the elements on both quantities is analysed and array-enhanced stochastic resonance is established for pure as well as noisy periodic signals. The coupling-induced improvement of the SNR compared to an uncoupled element is shown to be limited by a factor four which is only reached for vanishing input noise.Comment: 29 pages, 5 figure

    Mineralización de los rastrojos de trigo y comunidades microbianas asociadas para el control cultural de la septoriosis

    La siembra directa es una de las prácticas más importantes y efectivas para controlar la erosión, aumentar la materia orgánica y la fertilidad, mejorar el aprovechamiento del agua, disminuir los costos de producción y preservar el ecosistema. Sin embargo, la acumulación de los rastrojos ocasiona un aumento en el potencial de inóculo de ciertos patógenos hemibiótrofos, si no se implementan medidas de control eficientes. En siembra directa y monocultivo los patógenos hemibiótrofos son más severos, debido a que aumenta el período de supervivencia sobre los rastrojos retenidos en el suelo. El trigo debería ser cultivado en el mismo lote, sólo después de la completa mineralización de los restos culturales. Por eso, el conocimiento del tiempo en que el rastrojo del trigo persiste en el suelo en diferentes prácticas culturales y bajo diferentes ambientes, es de fundamental importancia. El objetivo del presente trabajo fue investigar el tiempo y la influencia del ambiente para una total mineralización del rastrojo de trigo infectado para conducir también el control cultural de la Septoriosis del trigo. Se cuantificó la perdida de peso del rastrojo de trigo, durante tres cuatrimestres continuados y se identificó la población de hongos involucrados en el proceso de la degradación, en monocultivo de trigo, bajo dos condiciones de recultivo( con recultivo de trigo y sin recultivo de trigo), dos sistemas de labranza( Siembra Directa y Labranza Convencional) y dos dosis de fertilizante (0 y 160 Kg.N.H-1). La labranza cero y alta dosis de N en el suelo aumentó la frecuencia de especies microbianas en las capas superficiales del suelo; esto se debió a la acumulación heterogénea de residuos a través del tiempo que produjo alta disponibilidad de nutrientes y acentuada actividad microbiológica de la micota celulolítica. La especie predominante, registrada por la técnica de recuento del agua de lavado del rastrojo fue Bipolares sorokiniana se diferenció significativamente de otros microorganismos tales como Mycosphaerella gramianicola, Stagonospora nodorum, Alternaria grupo infectoria y Fusarium graminearum y F. oxysporum. En cambio Fusarium equisetti fue observado en abundancia en labranza convencional. Con respecto a Fusarium spp, también se encontró una estrecha relación de este género con los restos vegetales, cumpliendo un rol importante en la descomposición del sustrato, junto con Trichoderma spp.. Dentro de este género se encuentran especies muy exitosas en cuanto a su capacidad como agentes biocontroladores de hongos patógenos por lo que se deben intensificar estudios con las nuevas cepas que se aíslen. Se concluye que para asegurarse una siembra futura con una población disminuida de potenciales patógenos para el cultivo, en un sistema de monocultivo de trigo, la condición no disturbada, acelera la descomposición del rastrojo, alcanzando la degradación total a los 13 meses de exposición de las trampas a la Labranza Convencional y las dos dosis de fertilizante; en cambio, en un sistema disturbado, fue necesario esperar 21 meses para que ocurriera esta situación. Se concluyó que las poblaciónes de Mycosphaerella graminicola y Septoria tritici sólo se vieron acrecentadas en el período estival, coincidiendo con la madurez del cultivo del primer año de exposición de las trampas; en el año subsiguiente se registró una merma a través de los tiempos de evaluación alcanzando su desaparición total para la primavera del segundo año

    Markov analysis of stochastic resonance in a periodically driven integrate-fire neuron

    We model the dynamics of the leaky integrate-fire neuron under periodic stimulation as a Markov process with respect to the stimulus phase. This avoids the unrealistic assumption of a stimulus reset after each spike made in earlier work and thus solves the long-standing reset problem. The neuron exhibits stochastic resonance, both with respect to input noise intensity and stimulus frequency. The latter resonance arises by matching the stimulus frequency to the refractory time of the neuron. The Markov approach can be generalized to other periodically driven stochastic processes containing a reset mechanism.Comment: 23 pages, 10 figure

    Targeting ErbB-2 nuclear localization and function inhibits breast cancer growth and overcomes trastuzumab resistance

    Membrane overexpression of ErbB-2/HER2 receptor tyrosine kinase (membrane ErbB-2 (MErbB-2)) has a critical role in breast cancer (BC). We and others have also shown the role of nuclear ErbB-2 (NErbB-2) in BC, whose presence we identified as a poor prognostic factor in MErbB-2-positive tumors. Current anti-ErbB-2 therapies, as with the antibody trastuzumab (Ttzm), target only MErbB-2. Here, we found that blockade of NErbB-2 action abrogates growth of BC cells, sensitive and resistant to Ttzm, in a scenario in which ErbB-2, ErbB-3 and Akt are phosphorylated, and ErbB-2/ErbB-3 dimers are formed. Also, inhibition of NErbB-2 presence suppresses growth of a preclinical BC model resistant to Ttzm. We showed that at the cyclin D1 promoter, ErbB-2 assembles a transcriptional complex with Stat3 (signal transducer and activator of transcription 3) and ErbB-3, another member of the ErbB family, which reveals the first nuclear function of ErbB-2/ErbB-3 dimer. We identified NErbB-2 as the major proliferation driver in Ttzm-resistant BC, and demonstrated that Ttzm inability to disrupt the Stat3/ErbB-2/ErbB-3 complex underlies its failure to inhibit growth. Furthermore, our results in the clinic revealed that nuclear interaction between ErbB-2 and Stat3 correlates with poor overall survival in primary breast tumors. Our findings challenge the paradigm of anti-ErbB-2 drug design and highlight NErbB-2 as a novel target to overcome Ttzm resistance.Fil: Cordo Russo, Rosalia Ines.     A Fokker-Planck formalism for diffusion with finite increments and absorbing boundaries

    Gaussian white noise is frequently used to model fluctuations in physical systems. In Fokker-Planck theory, this leads to a vanishing probability density near the absorbing boundary of threshold models. Here we derive the boundary condition for the stationary density of a first-order stochastic differential equation for additive finite-grained Poisson noise and show that the response properties of threshold units are qualitatively altered. Applied to the integrate-and-fire neuron model, the response turns out to be instantaneous rather than exhibiting low-pass characteristics, highly non-linear, and asymmetric for excitation and inhibition.     Ankle proprioception is not targeted by exercises on an unstable surface

    Item does not contain fulltextLaboratory study using a repeated measures design. The aim of this study was to determine if ankle proprioception is targeted in exercises on unstable surfaces. Lateral ankle sprain (LAS) has recurrence rates over 70%, which are believed to be due to a reduced accuracy of proprioceptive signals from the ankle. Proprioceptive exercises in rehabilitation of LAS mostly consist of balancing activities on an unstable surface. The methods include 100 healthy adults stood barefoot on a solid surface and a foam pad over a force plate, with occluded vision. Mechanical vibration was used to stimulate proprioceptive output of muscle spindles of triceps surae and lumbar paraspinal musculature. Each trial lasted for 60 s; vibration was applied from the 15th till the 30th second. Changes in mean velocity and mean position of the center of pressure (CoP) as a result of muscle vibration were calculated. Results show that on foam, the effect of triceps surae vibration on mean CoP velocity was significantly smaller than on a solid surface, while for paraspinal musculature vibration the effect was bigger on foam than on solid surface. Similar effects were seen for mean CoP displacement as outcome. Exercises on unstable surfaces appear not to target peripheral ankle proprioception. Exercises on an unstable surface may challenge the capacity of the central nervous system to shift the weighting of sources of proprioceptive signals on balance

    MAPK-ERK is a central pathway in T-cell acute lymphoblastic leukemia that drives steroid resistance

    (Patho-)physiological activation of the IL7-receptor (IL7R) signaling contributes to steroid resistance in pediatric T-cell acute lymphoblastic leukemia (T-ALL). Here, we show that activating IL7R pathway mutations and physiological IL7R signaling activate MAPK-ERK signaling, which provokes steroid resistance by phosphorylation of BIM. By mass spectrometry, we demonstrate that phosphorylated BIM is impaired in binding to BCL2, BCLXL and MCL1, shifting the apoptotic balance toward survival. Treatment with MEK inhibitors abolishes this inactivating phosphorylation of BIM and restores its interaction with anti-apoptotic BCL2-protein family members. Importantly, the MEK inhibitor selumetinib synergizes with steroids in both IL7-dependent and IL7-independent steroid resistant pediatric T-ALL PDX samples. Despite the anti-MAPK-ERK activity of ruxolitinib in IL7-induced signaling and JAK1 mutant cells, ruxolitinib only synergizes with steroid treatment in IL7-dependent steroid resistant PDX samples but not in IL7-independent steroid resistant PDX samples. Our study highlights the central role for MAPK-ERK signaling in steroid resistance in T-ALL patients, and demonstrates the broader application of MEK inhibitors over ruxolitinib to resensitize steroid-resistant T-ALL cells. These findings strongly support the enrollment of T-ALL patients in the current phase I/II SeluDex trial (NCT03705507) and contributes to the optimization and stratification of newly designed T-ALL treatment regimens

    The effects of stochastic resonance electrical stimulation and neoprene sleeve on knee proprioception

    <p>Abstract</p> <p>Background</p> <p>A variety of knee injuries and pathologies may cause a deficit in knee proprioception which may increase the risk of reinjury or the progression of disease. Stochastic resonance stimulation is a new therapy which has potential benefits for improving proprioceptive function. The objective of this study was to determine if stochastic resonance (SR) stimulation applied with a neoprene sleeve could improve knee proprioception relative to a no-stimulation/no-sleeve condition (control) or a sleeve alone condition in the normal, healthy knee. We hypothesized that SR stimulation when applied with a sleeve would enhance proprioception relative to the control and sleeve alone conditions.</p> <p>Methods</p> <p>Using a cross-over within subject design, twenty-four healthy subjects were tested under four combinations of conditions: electrical stimulation/sleeve, no stimulation/sleeve, no stimulation/no sleeve, and stimulation/no sleeve. Joint position sense (proprioception) was measured as the absolute mean difference between a target knee joint angle and the knee angle reproduced by the subject. Testing was conducted during both partial-weight bearing (PWB) and non-weight bearing (NWB) tasks. Differences in joint position sense between the conditions were evaluated by repeated-measures analysis of variance testing.</p> <p>Results</p> <p>Joint position sense error during the stimulation/sleeve condition (2.48° ± 1.32°) was found to be more accurate (P < 0.05) relative to the control condition (3.35° ± 1.63°) in the PWB task. No difference in joint position sense error was found between stimulation/sleeve and sleeve alone conditions for the PWB task. Joint position sense error was not found to differ between any of the conditions for the NWB task.</p> <p>Conclusion</p> <p>These results suggest that SR electrical stimulation when combined with a neoprene sleeve is an effective modality for enhancement of joint proprioception in the PWB knee. We believe these results suggest the need for further study of the potential of SR stimulation to correct proprioceptive deficits in a clinical population with knee injury/pathology or in subjects at risk of injury because of a proprioceptive deficit.</p
